Snake venoms, especially from the Crotalidae family, contain a variety of enzymes that prevent blood coagulation by virtue of their fibrinolytic enzymes. Nineteen snake venoms were screened for fibrinolytic activity and the highest activity was found in the venom of Crotalus basiliscus basiliscus venom. The active principle, basilase, was isolated, purified, and found to have fibrinolytic and fibrinogenolytic activity. It had a molecular weight of 22,000 and 1 mol of zinc per mole of protein associated with it. The proteolytic activity of the enzyme against dimethyl casein was inhibited by ethylenediaminetetraacetic acid and alpha 2-macroglobulin. It did not inactivate alpha 2-macroglobulin. Basilase did not have any of the following activities: thrombin-like, factor X-like, protein C activating, or urokinase-like. It caused neither hemorrhage nor platelet aggregation. In spite of its proteolytic activity, basilase did not hydrolyze the membranes of platelets. Basilase had 24% alpha-helix, 31% beta-sheet, 25% turns, and 20% unordered structure, as determined by Fourier Transform Infrared spectroscopy. OBJECTIVES: This study sought to evaluate the effects of veno-venous ultrafiltration on myocardial contractility in children undergoing cardiopulmonary bypass (CPB) for repair of congenital heart defects. BACKGROUND: Ultrafiltration (UF) is currently used to diminish postoperative fluid accumulation following CPB in children. Previous reports indicate improvement in hemodynamics immediately after UF, but the mechanism of its action is unknown. METHODS: Twenty-three patients (ages 2 months to 9.1 years; 13 males, 10 females) underwent UF for 10 min after CPB. Twelve patients underwent UF immediately after CPB (Group A). They were studied: (1) before and (2) after CPB, (3) after UF, and (4) 10 min after UF. Eleven patients underwent UF 10 min after CPB (Group B). They were studied: (1) before and (2) after CPB, (3) after a 10-min delay before UF, and (4) after UF. Contractility was determined by the difference in the observed and predicted velocity of circumferential fiber shortening for the measured wall stress, using transesophageal echocardiography. Left ventricular wall thickness was also measured. RESULTS: There was significant improvement in contractility after UF in both groups (mean+/-SD, Group A: -0.28+/-0.13 to -0.01+/-0.21 circ/s, p < 0.05; Group B: -0.26+/-0.16 to -0.11+/-0.17 circ/s, p < 0.05). Myocardial thickness to cavity dimension decreased in both groups following UF (Group A: 0.19+/-0.04 to 0.14+/-0.03, p < 0.05; Group B: 0.18+/-0.04 to 0.14+/-0.03, p < 0.05). CONCLUSIONS: UF improves hemodynamics by improving contractility and possibly by reducing myocardial edema in children following cardiac surgery. Enhanced patient outcome after ultrafiltration may in part be due to these changes. 相似文献

To define the optimal blood collection parameters for plasma human immunodeficiency virus type 1 (HIV-1) viral load testing, plasma HIV-1 RNA levels were quantitated with the NASBA HIV-1 RNA QT System from blood specimens that were collected, processed, and stored under a variety of conditions that might have affected HIV-1 RNA stability. We determined that when whole blood was processed within 2 h of specimen collection the levels of HIV-1 RNA detected in EDTA-, heparin-, and acid citrate dextrose (ACD)-anticoagulated plasma samples were comparable. The levels of HIV-1 RNA in serum specimens (mean = 4.126 log units) were significantly lower (P < 0.01) than the levels in corresponding plasma samples (mean = 4.501 log units). One cycle of freeze-thaw (-70 degrees C) did not significantly reduce the level of HIV-1 RNA detected in EDTA-, heparin-, or ACD-anticoagulated plasmas. The EDTA-anticoagulated plasmas showed the smallest decrease in HIV-1 RNA copies (0.050 log units). HIV-1 RNA levels decreased over a 6-month time period in serum as well as in EDTA-, ACD-, and heparin-anticoagulated plasmas stored at -70 degrees C. However, the only significant decreases were for serum (mean decrease = 0.317 log units) and heparin-anticoagulated samples (mean decrease = 0.384 log units). A comparison of the levels of HIV-1 RNA in cell-free plasma collected in VACUTAINER EDTA Plasma Preparation Tubes and in standard VACUTAINER EDTA tubes determined that HIV-1 RNA levels were stable for up to 30 h after collection when stored at either room temperature (mean standard deviation [SD] = +/- 0.101 log units) or at 4 degrees C (mean SD = +/- 0.102 log units) as cell-free plasma or as EDTA-anticoagulated whole blood (mean SD = +/- 0.109 log units). When the thoracic spinal cord of the North American opossum is transected early in development, supraspinal axons grow through the lesion. In the experiments reported here, we asked whether regeneration of cut axons contributes to such growth. Fast Blue (FB) was injected into the lumbar cord on postnatal day (PD)5, 8, 15, or 20. Five days later, FB was removed by gentle suction, and the spinal cord was transected at thoracic levels. Fourteen days later, rhodamine B dextran was injected between the site of the FB injection and the lesion. The pups were maintained for an additional 7-10 days before killing and perfusion. We assumed that supraspinal neurons that contained FB survived axotomy and those that contained both FB and rhodamine B dextran supported regenerating axons. In the PD5 group (lesioned at PD10), regenerative growth was documented for axons originating in all of the supraspinal nuclei that innervate the lumbar cord by PD10. When the injections were made at the later ages, however, neurons that supported regenerative growth were fewer in number and regionally restricted. In some cases, they were limited primarily to the red nucleus, the medullary raphe, and the adjacent reticular formation. Our results show that regeneration of cut axons contributes to growth of supraspinal axons through the lesion after transection of the thoracic cord in developing opossums and that the critical period for regenerative growth is not the same for all axons. 相似文献

The carbothermic reduction of magnesia to produce magnesium offers the potential of a lower energy and higher productivity
route for metal production compared to existing industrial routes. The reaction of magnesia and carbon produces a magnesium
and carbon monoxide vapor. Slow cooling of that vapor will allow the reaction to quickly revert and the prevention of this
reversion reaction is a major technical challenge. Two main approaches can be taken to prevent reversion and allow recovery
of the metal product: rapid quenching of the vapor and dissolving the magnesium directly in a suitable metal solvent before
reversion can occur. The commercial viability of either carbothermic route to magnesium is closely connected to the physical

Constraint satisfaction problems can be expressed very elegantly in state-based formal methods such as B. But can such specifications be directly used for solving real-life problems? In other words, can a formal model be more than a design artefact but also be used at runtime for inference and problem solving? We will try and answer this important question in the present paper with regard to the university timetabling problem. We report on an ongoing project to build a curriculum timetable validation tool where we use a formal model as the basis to validate timetables from a student’s perspective and to support incremental modification of timetables. Drift analysis is a powerful tool used to bound the optimization time of evolutionary algorithms (EAs). Various previous works
apply a drift theorem going back to Hajek in order to show exponential lower bounds on the optimization time of EAs. However,
this drift theorem is tedious to read and to apply since it requires two bounds on the moment-generating (exponential) function
of the drift. A recent work identifies a specialization of this drift theorem that is much easier to apply. Nevertheless,
it is not as simple and not as general as possible. The present paper picks up Hajek’s line of thought to prove a drift theorem
that is very easy to use in evolutionary computation. Only two conditions have to be verified, one of which holds for virtually
all EAs with standard mutation. The other condition is a bound on what is really relevant, the drift. Applications show how
previous analyses involving the complicated theorem can be redone in a much simpler and clearer way. In some cases even improved
results may be achieved. Therefore, the simplified theorem is also a didactical contribution to the runtime analysis of EAs. 相似文献

Confocal microscopy and rheology studies of two bijel systems are presented to elucidate relationships between the physicochemical properties of bijels and their ability to be utilized as soft matter templates for materials synthesis. For the first time, the origins of viscoelasticity in these systems are investigated using conventional rheometry and a direct correspondence between the elastic storage modulus, particle loading, and the departure from criticality is observed. Further, the rheological transitions that accompany fluid re‐mixing in bijels are characterized, providing key insights into the synergistic role of interfacial tension and interparticle interactions in mediating their mechanical robustness. Bijels that are predominantly stabilized by interfacial tension are also highly sensitive to gradients in chemical composition and more easily prone to mechanical failure during processing. Despite this increased sensitivity, a modified strategy for processing these more delicate systems is developed and its efficacy is demonstrated by synthesizing a bicontinuous macroporous hydrogel scaffold. 相似文献
