A prospective study was conducted on 25 Legionella pneumophila culture-positive and 98 culture-negative bronchoalveolar lavage fluid samples to compare two DNA preparation methods: a rapid modified Chelex-based protocol and a proteinase K method. PCR was found to be more sensitive with the Chelex-based method (P = 0.03). N difference was found concerning the inhibition rate.  相似文献

Nutritional support of preterm infants by total parental nutrition (TPN) is common; however, TPN compromises intestinal structure and enzyme activity. Our goal was to develop a piglet model to assess the effect of limited enteral stimulation with formula and insulin-like growth factor-I (IGF-I) on intestinal morphology and enzyme activity. A nutritionally complete TPN solution was infused for 7 days and piglets were gavaged twice daily with 4 ml sterile water, formula, or formula containing 1,000 micrograms/l IGF-I. Litter mate piglets fed formula served as orally fed controls. On day 7, body weights and plasma hormone profiles of TPN and orally fed piglets were similar. However, intestinal weight, jejunal and ileal villus height and surface area, and mucosal lactase and sucrase activity of TPN piglets was reduced by approximately 50%. No effect of limited enteral stimulation or IGF-I was observed likely due to the small volume of formula administered enterally, which resulted in a low dose of IGF-I (4.3 micrograms IGF-I/kg/day).  相似文献

BACKGROUND: Vitiligo is a pigmentary disorder of unknown cause characterized by depigmented patches due to destruction of melanocytes. Recently, the inherent cellular defect theory has been discussed. To investigate the biologic characteristics of cultured melanocytes from normal and vitiligo subjects, this study had the purpose to examine the functional and ultrastructural characteristics of these melanocytes and to observe the morphologic and functional changes of melanocytes in response to ultraviolet B irradiation. METHODS: Melanocytes were isolated and cultured from foreskin and arm skin of normal and vitiligo subjects. The DNA synthesis, tyrosinase activity assay, transmission and scanning electron microscopic examination, and the effects of ultraviolet B(UVB)-irradiation on cultured melanocytes were studied. RESULTS: Vitiligo melanocytes showed no significant differences in DNA synthesis and tyrosinase activity compared with normal melanocytes, but the vitiligo melanocytes contained dilated and/or circular rough endoplasmic reticulum (RER) on transmission electron microscopic examination. Exposure of the cultured melanocytes to UVB resulted in increased protein synthesis and tyrosinase activity. Morphologic alterations and changes in DNA synthesis were also noted. Compared with normal melanocytes, the responses of vitiligo melanocyte to UVB showed no significant difference. CONCLUSIONS: Normal and vitiligo melanocytes showed similar biologic characteristics except in the changes of RERs in the vitiligo melanocytes. The ultrastructural aberrations in vitiligo subjects do not seem to be directly related to the biologic characteristics and the responses to UVB irradiation in vitiligo melanocytes.  相似文献

A possible interaction of salmon-calcitonin with opioid systems was studied in isolated tissues. Neurogenic contractions were elicited by electrical stimulation in guinea-pig ileum myenteric plexus-longitudinal muscle strips, rabbit vas deferens and mouse vas deferens. Bremazocine inhibited neurogenic contractions in all three tissues (presumably through kappa-receptors) [D-Pen2, D-Pen5]enkephalin and [Met5]enkephalin inhibited contractions in mouse vas deferens (presumably through delta-receptors), and [D-Ala2, N-Me-Phe4, Gly5-ol]enkephalin (DAMGO) inhibited contractions in guinea-pig ileum and mouse vas deferens (presumably through mu-receptors). All inhibitory effects were concentration-dependent. Salmon-calcitonin 0.1 IU/ml increased the effect of bremazocine in guinea-pig ileum and rabbit vas deferens and also increased the effects of [D-Pen2, D-Pen5]enkephalin and [Met5]enkephalin in mouse vas deferens. In contrast, salmon-calcitonin up to 0.4 IU/ml did not change the effect of bremazocine in mouse vas deferens and the effect of DAMGO in guinea-pig ileum and mouse vas deferens. It is concluded that salmon-calcitonin enhances agonist effects at opioid kappa- and delta- but not at opioid mu-receptors. The level of this interaction remains to be elucidated. The interaction may be the basis of the analgesic effect of salmon-calcitonin in vivo.  相似文献

Acute renal failure (ARF) has changed in its aetiology in developed Western countries. This study compares our experiences of the aetiology and incidence of ARF a decade after a previously recorded paper in the literature. The data of 226 patients with a diagnosis of ARF during a 3-year period from 1986 to 1988 was analysed. ARF in our study occurred at a younger age. Medical causes of ARF remain the dominant subgroup. There were female patients presenting with ARF associated with self-induced abortions. Toxins of a herbal variety and infections remain the commonest aetiological factors in the medical subgroup. Infections have replaced nephrotoxins as being the principal medical cause. There has been no major change in the incidence or aetiology of ARF over a decade. The prevention of ARF in blacks in sub-Saharan Africa is primarily concerned with the eradication of nephrotoxins and infections and a laxity in the abortion laws of the country. Only by an improvement in education, nutrition, socio-economic status and sanitation can one eradicate or diminish ARF in developing countries.  相似文献

We examined the issue of reliability of InP-based high-electron mobility transistors (HEMTs), focusing on the increase of drain resistance Rd. In investigations of the mechanism of Rd increase, we took note of contaminant incorporation and of the relations between the device lifetime and the strength of the channel electric field. In the fabrication process, reducing contaminants, especially fluorine, significantly suppressed the increase of source and drain resistances. Cross-sectional views of the gate of improved devices, which had a long lifetime, confirmed an almost contaminant-free surface around the gate. In acceleration tests, the most negative impact on drain resistance stability among several bias conditions was found when the current density was high and the channel electric field was large at the same time. The dependence of drain-gate electric field strength E showed that the device lifetimes of HEMTs determined from Rd increase obeyed exp(1/E), which means that impact ionization was the main cause of degradation. We elucidate that the interactions of hot carriers with contaminants around the gate are the main causes of the Rd increase in HEMTs. Suppression of device degradation was achieved by optimizing the fabrication process around the gate. In this way, device lifetime was remarkably enhanced.  相似文献

Antimony ions were implanted into ZnO films grown on c-plane sapphire by pulsed-laser deposition. Raman scattering modes of the Sb-implanted samples were found to be influenced by the implantation dose. A characteristic peak at 576 cm− 1 was observed with an asymmetric shape due to ion damage to the lattice of the implanted ZnO films. When the implant dose was low, the height of the peak was reduced by rapid thermal annealing at 400-600 °C and the symmetry of the spectra was recovered. However, when the Sb dose exceeded 1 × 1015 cm− 2, the peak maintained unchanged after rapid thermal annealing at temperatures up to 600 °C. A broad and low Raman peak was observed at 437 cm− 1, which is related to the surface damage caused by the energetic ion bombarding. Photoluminescence measurement showed a decrease of the bandedge emission at 3.36 eV, a clear effect of defects induced by the implantation, and confirmed partial recovery of the crystal by rapid annealing.  相似文献

The seesaw-oscillation of a small circular pad on a single droplet was studied both numerically and experimentally. The circular pad with a diameter of 2.0–3.8 mm onto a water or glycerol droplet with a volume of 1–10 μL, and a bottom substrate with a smaller diameter than that of the pad were used in the experiment. The pad was then tilted and then the tilting fixture was quickly removed. The pad alternately oscillated and then finally stabilized in a horizontal position. The numerical model considering the surface tension and the viscous force of the droplet was developed and calculated using the same configurations as those in the experiment. The experimental and numerical data showed good agreement not only in terms of the oscillating frequency and damping ratio but the transient motion of the circular pad and instantaneous droplet surface shape.  相似文献
