Antagonism of l-stepholidine against bromocriptine-inhibition on prolactin level in lactational rats

AIM: To study the antagonism of l-stepholidine (SPD) against bromocriptine (Bro)-inhibition on prolactin (PRL) level. METHODS: Bro (0.5 mg.kg-1.d-1, s.c.) reduced the PRL and caused a dysplasia of mammary gland in lactational rats. The weight growing of newborn rats was retarded. The PRL of the lactational rats was assessed by immunoradiometric assay (IRMA); the weight of newborn rats and development of mammary glands in lactational rats were also examined. Antagonism of SPD was evaluated. RESULTS: SPD (30 & 100 mg.kg-1.d-1, i.p.) obviously antagonized the Bro that induced lowering the PRL level in lactational rats, the PRL was 11 +/- 4 & 23 +/- 6 micrograms.L-1 (NS 7 +/- 2) respectively on d 15 of postpartum and the development of mammary gland in lactational rats was normal. The newborn rats grew rapidly in 11-15 d. CONCLUSION: SPD possessed an antagonism with Bro inhibition on D2 receptors located in the pituitary gland, and was an antagonist of dopamine D2 receptors.     

Sex difference and testosterone modulation of pheromone-induced NeuronalFos in the Ferret's main olfactory bulb and hypothalamus

A carnivore, the ferret possesses a vomeronasal organ--accessory olfactory bulb (VNO-AOB) projection to the hypothalamus; however, little is known about its function. Pheromones in soiled bedding from estrous female ferrets or an artificial peppermint odor significantly augmented nuclear Fos protein immunoreactivity (Fos-IR), a marker of neural activation, in several main olfactory bulb (MOB) sites but not in the AOB of gonadectomized male and females. Testosterone propionate (TP) significantly augmented the MOB's neuronal Fos responses to estrous females' pheromones, but not to peppermint. Estrous odors, but not peppermint, also augmented neuronal Fos-IR in the medial preoptic area (mPOA) of female, but not male, subjects. Pheromones in soiled bedding from breeding male ferrets significantly augmented neuronal Fos-IR in the MOB and in the medial amygdala of gonadectomized, TP-treated male and female subjects. Again, male pheromones failed to influence neuronal Fos-IR in the AOB of either sex, and only females showed significant increases in neuronal Fos-IR in the lateral aspect of the ventromedial nucleus and mPOA. These results point to an essential role among higher mammals of the main olfactory epithelium-MOB projection to the hypothalamus in detecting and processing pheromones. Gonadectomized ferrets showed significant increases in sniffing behavior when placed on either female or male bedding. This occurred regardless of whether they had received TP or oil vehicle, suggesting that testosterone's facilitation of neuronal Fos responses to estrous females' odors in the MOB of both sexes cannot be attributed to increased scent gathering. Androgen receptor-IR was present in the MOB granule cell layer of male and female ferrets, raising the possibility that testosterone acts directly on these cells to augment their responsiveness to pheromones.     

Digital quantification of the enzyme-linked immunospot (ELISPOT)

Enzyme-linked immunospot (ELISPOT) is a sensitive technique for the detection of cytokines released by immune cells. The technique is well established and correlates closely with the enzyme-linked immunosorbent assay (ELISA) technique. Here, we introduce an integrated imaging-analysis system to quantitate the spots formed by the ELISPOT assay. The spots can be easily counted when the background is clear and the spots are few. However, when the number of spots increases to > 100, this task becomes challenging and time-consuming. To minimize time and standardize the counting procedure, we have used a digital camera linked to a computer system for reading the plates. In general, the computer is able to count more spots and has a smaller standard deviation when compared with the manual microscopic count. This integrated system is commercially available, and we believe that this method is objective, time-saving and consistent in routine ELISPOT assay counting, especially when numerous spots are present.     

Microsurgical sutures with non-transfixing staples. An experimental study of 15 rat aortas

This study, conducted in rats, studied a new system of anastomosis, by nontransfixing clips, pinching each edge of the artery with minimal trauma. Histological examinations were performed at one week and one month in order to investigate the vascular wall in the line of anastomosis. Clinical application of this procedure was undertaken in view of the encouraging and satisfactory results obtained.
