A single administration of a physiological dose of noradrenaline to animals. in contrast to adrenaline, stimulates the respiration of mitochondria not only under oxidation of FAD-dependent Krebbs cycle substrate of the succinase but also HAD-dependent substrate of alpha-ketoglutarate. In the both cases the phosphorylation rate increases, since the action of noradrenaline, separating the respiration and oxidative phosphorylation, was not found. 1. A contact of the pteropod mollusk Clione limacina with its prey (small pteropod mollusk Limacina helicina) evokes a complex pattern of hunting and feeding behavior: protraction of tentacles to seize the prey, activation of buccal apparatus to swallow the prey, activation of locomotor system (speeding up of wing beating), reversal of reaction to tactile stimulation of the head, loss of normal (vertical) orientation in space, and swimming in circles. After injection of physostigmine (PhS), the acetylcholinesterase inhibitor, into the hemocoel of intact Clione, all these manifestations of the hunting and feeding behavior could be evoked by tactile stimulation of the head, or they arose spontaneously. 2. In the preparation of the isolated CNS, the effect of PhS on the neural networks controlling different aspects of the hunting and feeding behavior was studied by recording from neurons monitoring activity of different networks (< or = 4 neurons simultaneously). Tactile stimulation of the head was mimicked by a short-term electrical stimulation of the corresponding nerve. Before PhS application, the nerve stimulation evoked elements of the defense reaction, i.e., long-lasting inhibition of all main motor control systems: the locomotor network in the pedal ganglia, the tentacle control network in the cerebral ganglia, and the network controlling radula and hook movements in the buccal ganglia. However, after PhS application, the same stimulus evoked a long-lasting bout of excitation in all the three networks accompanied by activation of the heart-exciting neuron as well as by a modification of the activity of statocyst receptor cells controlling Clione's spatial orientation (the "fictive hunting bout"). Similar hunting bouts could arise spontaneously. 3. Injection of acetylcholine (ACh) into the hemocoel of intact Clione was less effective than injection of PhS. After ACh injection, reversal of reaction to head stimulation was observed in < or = 20% of the experiments (the percentage of positive results was higher if ACh was injected into the head just over the CNS). Bath application of ACh to the isolated CNS did not produce the hunting bouts. However, a short-term local application of ACh to the cerebral ganglia in the isolated CNS resulted in activation of the main motor systems controlling locomotion, protraction of tentacles, and movements of buccal mass. 4. During spontaneous PhS-induced bouts, excitation of different networks involved in hunting behavior was sometimes not quite synchronous. The goals of this study were 1) to compare the effects of transforming growth factor-beta (TGF-beta) and parathyroid hormone-related protein (PTHrP) on mouse blastocyst attachment and outgrowth in vitro, 2) to determine whether TGF-beta acts through a mechanism involving PTHrP, 3) to examine effects of PTHrP on preimplantation mouse embryo development, and 4) to determine the pattern of expression of PTHrP protein in the uterus of the mouse during early gestation. In the first set of experiments, hatched blastocysts were placed in fibronectin-coated wells. Cultures were treated with PTHrP or TGF-beta1 and assessed at 24, 48, and 72 h for attachment and surface area of blastocyst outgrowth. Results showed that both PTHrP and TGF-beta1 increased blastocyst outgrowth significantly. A PTHrP-neutralizing antibody blocked the stimulatory effect of both PTHrP and TGF-beta1, suggesting that TGF-beta1 acts to increase endogenous production of PTHrP by the blastocyst. Immunoassay of conditioned medium from blastocysts treated with either TGF-beta1 or PTHrP 1-34 confirmed a 3- to 4-fold increase in levels of PTHrP 1-141. In the second series of experiments, pronuclear zygotes were cultured in various concentrations of PTHrP for 96 h. Blastocysts then were subjected to differential fluorescent staining of inner cell mass and trophectoderm cells. Treatment of mouse embryos with the various concentrations of PTHrP altered neither the number developing to the blastocyst stage nor the number of inner cell mass or trophectoderm cells in the resulting blastocysts. In the third experiment, pregnant mice were killed at Days 3, 4, 5, 6, and 7 of gestation, and uterine horns were processed for immunohistochemistry. Uterine sections were stained with antibodies to PTHrP, desmin, and laminin. On Days 3, 4, and 5, uterine luminal and glandular epithelial cells stained intensely for PTHrP, while stromal cells were negative. By Days 6 and 7, decidualized stromal cells stained positively for PTHrP, desmin, and laminin. In this paper, two methods are presented. One involves the separation of calcipotriol, a new synthetic analogue, from two related compounds, specifically cholecalciferol (Vitamin D3) and calcitriol (1,25-dihydroxyvitamin D3). The other involves the isolation and assay of calcipotriol from a topical ointment. The study was performed with reversed-phase high-performance liquid chromatography using an RP18 column and ultraviolet detection. Applying the method of Snyder, a mobile phase mixture containing methanol-acetonitrile-water (67:23:10, v/v) was found which achieved a total separation within 18 min. A mobile phase of methanol-water (80:20, v/v) attained a slower elution of calcipotriol. For isolation and assay of calcipotriol from an ointment (Daivonex), dissolution in chloroform gave the highest recovery (> 98%). The isolation and assay process can be performed within 2 h. 相似文献
