Large myelinated club ending and small-vesicle bouton synapses on the distal part of the lateral dendrite of the goldfish Mauthner cell were investigated with thin section, freeze-fracture, and immunocytochemical electron microscopic methods. Large myelinated club endings form mixed synapses, having both gap junctions and chemical synaptic junctions. The correlation of the number of gap junction particles (connexons) and the data from electrophysiological studies of single large myelinated club ending synapses suggest that only a small fraction of gap junction channels are open at any given time during electrical synaptic transmission. The chemical synaptic junctions at the large myelinated club ending synapse have large, round synaptic vesicles, indicating that they are excitatory. This result is in agreement with electrophysiological data demonstrating the excitatory nature of this chemical synapse. Freeze-fracture of these excitatory chemical synaptic junctions reveals the presence of the intramembrane particle aggregates in the postsynaptic E face. Small-vesicle boutons form chemical synaptic junctions with small, flat or oval synaptic vesicles. These structural data, in combination with previous electrophysiological studies, suggest that the small-vesicle bouton synapses are inhibitory. In support of this theory, the cytoplasmic side of the postsynaptic membrane of some of these synapses show positive immunocytochemical reaction to monoclonal antibodies against the rat glycine receptor. Freeze-fracture data reveal intramembrane particle aggregates in the postsynaptic P face of some small-vesicle bouton synapses which could possibly represent glycine receptor aggregates.  相似文献

The conformational change and the alignment behavior of poly-4BCMU were investigated in compression, de-compression and re-compression process using LB method. In compression process, the conformation of poly-4BCMU changed from random-coil conformation to rigid-rod one. With the increase of surface pressure, poly-4BCMU with rigid-rod conformation gathered into bundles and they aligned perpendicular to the compressing direction. Once stopping the compression and releasing the surface pressure, the bundle textures released packing. In this process (de-compression process), the conformational change never occurred. Discrete slender fibrils gathered again in re-compression process, resulting in the formation of close-packed bundles with high molecular ordering.  相似文献

Miscibility of binary and ternary polymer blends composed of thermotropic liquid crystalline polycarbonate (LCPC), poly(vinyl alcohol) (PVA), and chitosan was investigated by viscosity method, FTIR spectrum, and scanning electron microscope techniques. Effect of addition of chitosan as a compatibilizer on miscibility and morphology of binary LCPC/chitosan and PVA/chitosan and ternary LCPC/PVA/chitosan polymer blends was discussed. These measurements indicated that addition of chitosan into the blends of LCPC with PVA leads to an increase of miscibility and a formation of clear fibril structures on fractured surfaces, which are due to intermolecular hydrogen‐bonding interaction between LCPC, PVA, and chitosan chains. It was suggested that side‐chain hydroxy group of PVA and amino and hydroxy groups of chitosan play an important role in the formation of miscible phase and improvement of morphology in binary and ternary blends composed of LCPC, PVA, and chitosan. © 2004 Wiley Periodicals, Inc. Four chelating resins containing triethylenetetramine side chains and/or thiol groups were made from macroreticular 2,3-epithiopropyl methacrylate, styrene–divinylbenzene (DVB), or methyl methacrylate–DVB copolymer beads, and then the resins bearing metal ions such as Ag+, Cu2+, and Zn2+ were made. The antibacterial activity of the resins bearing metal ions against Escherichia coli (E. coli) or Staphylococcus aureus (S. aureus) was investigated. The resins containing thiol groups showed the higher adsorption capacity for silver ions than for other metal ions. The resins, which contain both triethylenetetramine side chains and thiol groups, bearing silver ions (RE-TTA-Ag) exhibited high antibacterial activity against bacteria, especially E. coli, without the residual silver ions in water after contacting with bacteria. Copolymer beads (RCCS-4G) with many chloromethyl groups were prepared by treating macroreticular chloromethylstyrene-tetraethyleneglycol dimethacrylate (4G) copolymer beads with chloromethylether. Copolymer beads (RAAS-4G) with benzylamino groups were prepared by treating RCCS-4G with potassium phthalimide. Then the copolymer beads containing phenol derivatives were prepared by treating RAAS-4G with p-hydroxybenzoic acid (pHBA), 2,4-dihydroxybenzoic acid (DHBA), and 3,4,5-trihydroxybenzoic acid (gallic acid, GA) in N,N-dimethylformamide. The antibacterial activity of the obtained resins was examined against Escherichia coli and Staphylococcus aureus. Resins containing phenolic hydroxy groups of 2.3–7.7 mequiv/g were obtained. Antibacterial activity of the resins containing various phenol derivatives against E. coli or S. aureus increased in the order of RAAS-4G-GA > RAAS-4G-DHBA > RAAS-4G-pHBA. The resins containing phenol derivatives exhibited higher antibacterial activity against E. coli than against S. aureus and high activity even against bacteria in NaCl solution. Scanning electron micrographs showed that high antibacterial activity was brought about by the phenolic hydroxyl groups in the resin. © 1997 John Wiley & Sons, Inc. Polyacrylonitrile (PAN) fibers with mean diameters in 270-400 nm range were prepared by electrospinning for use as a filter media. Compared to commercial filters made of polyolefin and glass, the fibers of electrospun filters were more uniform in diameter. The performance of electrospun filters was evaluated by measuring the penetration of monodisperse NaCl nanoparticles (below 80 nm in size) through the filters. It was found that electrospun filters could be made which had nanoparticle penetration values comparable to commercial filters but with substantially less filter mass. The penetration of nanoparticles through the electrospun filter media could be reduced by increasing the filter thickness, which is controlled by the collection time during the electrospinning process. Nanoparticle collection by electrostatic forces was found to be negligible for electrospun filters. Filter quality factors and single fiber collection efficiencies were found to be independent of filter thickness for electrospun filters, and the penetration of nanoparticles through electrospun filters was in better agreement with theoretical predictions than was the measured penetration through a commercial filter. This study shows that electrospinning is a promising technology for the production of high performance nanoparticle filters.  相似文献

As a basis for theories of psychopathology, clinical psychology and related disciplines need sound taxonomies that are generalizable across diverse populations. To test the generalizability of a statistically derived 8-syndrome taxonomic model for youth psychopathology, confirmatory factor analyses (CFAs) were performed on the Youth Self-Report (T. M. Achenbach & L. A. Rescorla, 2001) completed by 30,243 youths 11-18 years old from 23 societies. The 8-syndrome taxonomic model met criteria for good fit to the data from each society. This was consistent with findings for the parent-completed Child Behavior Checklist (Achenbach & Rescorla, 2001) and the teacher-completed Teacher's Report Form (Achenbach & Rescorla, 2001) from many societies. Separate CFAs by gender and age group supported the 8-syndrome model for boys and girls and for younger and older youths within individual societies. The findings provide initial support for the taxonomic generalizability of the 8-syndrome model across very diverse societies, both genders, and 2 age groups. Sterol balance studies, using both isotopic and chromatographic techniques, were carried out in hamsters fed semipurified diets to detect changes in sterol metabolism during the early period of the lithogenic stimulus. The balance studies examined animals in the first two weeks on the experimental lithogenic diets. The variables were as follows: dose of cholesterol (group 1, 0.05% vs. group 2, 0.2%); dietary fat (fatty acid) (group 2, butterfat vs. group 4, palmitic acid); source of hamster [group 2, Sasco (Omaha, NE) vs. group 3, Charles River (Wilmington, MA)]; average weight of animals (group 4, 60 g vs. group 5, 119 g). Animals in groups 1, 2, 3 and 5 maintained almost constant weight throughout the two-week balance study. Liver and plasma cholesterol levels increased in groups 2–5 with increasing dose of dietary cholesterol. The highest levels were found in group 4 (liver cholesterol, 32.7 mg/g; plasma cholesterol, 367 mg/dL). Sterol balance measurements showed that bile acid synthesis remained low (range 0.55–1.01 mg/d) for all groups regardless of the intake of dietary cholesterol (range, 3.27–20.90 mg/d). The dietary cholesterol absorbed from the intestine (range, 2.91–18.91 mg/d) was stored in the liver; this storage was reflected in the negative values for cholesterol balance for all groups (range, −0.70 to −14.97 mg/d). These studies did not reveal any correlations between parameters of sterol balance and cholelithiasis.  相似文献

The Fresnel zone plate (FZP) is a type of lens for focusing X-rays. We have fabricated a kinoform style FZP with a 30-step structure composed of concentric multilayer of alternating Cu layer, Al layer, and 28 composite material (Cu, Al) layers. The multilayer was deposited using a magnetron sputtering apparatus with two DC-sputtering guns. The focusing characteristics were evaluated at the synchrotron radiation beamline of SPring-8, and the focused beam size measured by knife-edge scanning was 0.8 μm at 40 keV. The peak diffraction efficiencies measured using ion chambers for 1st- and 2nd-order diffraction were 42% and 12% at 40 keV and 20 keV, respectively. High-order X-ray diffraction was also examined using the data obtained from a CCD camera.  相似文献

Cystatin C (CST3) is an endogenous cysteine protease inhibitor, which is implicated in cerebral amyloid angiopathy (CAA). In CAA, CST3 is found to be aggregated. The purpose of this study is to investigate whether this aggregation could alter the activity of the protein relevant to the molecular pathology of CAA. A system of CST3 protein aggregation was established, and the aggregated protein was characterized. The results showed that CST3 aggregated both at 80 °C without agitation, and at 37 °C with agitation in a time-dependent manner. However, the levels of aggregation were high and appeared earlier at 80 °C. Dot-blot immunoassay for oligomers revealed that CST3 could make oligomeric aggregates at the 37 °C condition. Electron microscopy showed that CST3 could make short fibrillary aggregates at 37 °C. Cathepsin B activity assay demonstrated that aggregated CST3 inhibited the enzyme activity less efficiently at pH 5.5. At 7.4 pH, it lost the inhibitory properties almost completely. In addition, aggregated CST3 did not inhibit Aβ1-40 fibril formation, rather, it slightly increased it. CST3 immunocytochemistry showed that the protein was positive both in monomeric and aggregated CST3-treated neuronal culture. However, His6 immunocytochemistry revealed that the internalization of exogenous recombinant CST3 by an astrocytoma cell culture was higher when the protein was aggregated compared to its monomeric form. Finally, MTT cell viability assay showed that the aggregated form of CST3 was more toxic than the monomeric form. Thus, our results suggest that aggregation may result in a loss-of-function phenotype of CST3, which is toxic and responsible for cellular degeneration.  相似文献
