The neuronal cytoskeleton is one of the most profoundly altered organelles in late life neuro-degenerative disorders that are characterized by progressive impairments in cognitive abilities. The elucidation of the protein building blocks of these organelles as well as advances in understanding how these proteins become altered in Alzheimer's disease (AD) and other less common dementing illnesses, i.e., diffuse Lewy body disease (DLBD) or the Lewy body variant of AD (LBVAD), will provide insights into the molecular basis of these disorders. Within, we review evidence that normal adult human tau is abnormally phosphorylated and converted into the subunits of AD paired helical filaments (PHFs), and that Lewy bodies (LBs) represent accumulation of altered neurofilament (NF) triplet subunits. Although the precise biological consequences of PHF and LB formation in neurons is unknown, growing evidence suggests that the formation of PHFs and LBs from normal neuronal cytoskeletal proteins could have deleterious effects on neuronal function and survival. Finally, insights into the composition of PHFs and LBs could lead to the development of novel strategies for the timely and accurate diagnosis of AD, DLBD and the LBVAD. 相似文献

The method of x-ray electron spectroscopy is used to investigate lead silicatexPbO(1 -x) · SiO2 (x - 0.3, 0.4, 0.5, 0.55, 0.667) glasses. The concentration dependences of the spectra of inner Pb4f, Si2p, and Ols levels led to the conclusion that for low contents of PbO, lead is present in the glass in the form of modifying ions, whereas in high concentrations, it plays the role of glass former. Restructuring in the glass occurs for about 50% molar concentration of PbO. The Pb - O bond in the glass is more ionic than in PbO. With an increase in the PbO content in the glass, the Pb - O bond becomes closer to that of lead monoxide, i.e., is more covalent. 相似文献

Catastrophic convolutional codes (CC) cause an infinite number of decoded data bit errors when decoding a finite number of code symbols. A CC displays a catastrophic error propagation if the generating polynomials have a common factor. An efficient algorithm for polynomial factorization in GF(2m) is used for detecting catastrophic CC for any rate n/m and constraint length k. The effect of a novel flavonoid, venoruton (a mixture of mono-, di-, tri- and tetrahydroxyethylrutosides) has been investigated in healthy rat lenses and compared with diabetic cataract modelled in vitro. One mM venoruton was added to medium simulating healthy and diabetic conditions for the incubated lenses; damage was followed by either stereoscopic photography of the lenses under a Cooperative Cataract Research Group operating microscope or with our recently developed method: the leakage of lactate dehydrogenase (LDH) into the lens culture media. The increased LDH activity in the medium and observable development of the opacity were correlated with cell damage, which has been found to be associated with globular degeneration and cataract formation. The extent of opacification and LDH release is reduced if 1 mM venoruton is included in the medium. The protective effect may be related to antioxidant activity against reactive oxygen species: decreased luminol luminescence was shown after venoruton addition to either superoxide-generating hypoxanthine plus xanthine oxidase, or hydrogen peroxide. 相似文献

Immunofluorescence microscopic observations indicated that a monoclonal antibody, Vmp 18, raised against the peptide 199-208 of murine interleukin 1 alpha, cross-reacted with an antigenic determinant of Drosophila thorax muscles. Immunoelectron microscopic analysis showed that the gold particles were mainly localized in the Z-line which is the attachment site of thin filaments from adjacent sarcomeres. On the contrary, the antibody failed to mark the Z-line in vertebrate skeletal muscle. A Western blot of total protein extract from Drosophila thorax muscles bound a protein of 43 kDa. Coating technology plays a significant role in a number of applications such as high temperatures, corrosion, oxidation, wear, and interface. In this paper, we investigate the interface cracking between ceramic and/or functionally graded coatings (FGM coatings) and a substrate under antiplane shear. Four coating models are considered, namely single layered homogeneous coating, double layered piece-wise homogeneous coating, single layered FGM coating and double layered coating with an FGM bottom coat. Mode III stress intensity factors (SIFs) are calculated for the different coating models. In the case of μL > μ0 where μ0 is the shear modulus of the substrate and μL the shear modulus of the material at the surface of the coating, it is found that the single layered FGM coating reduces SIF slightly, whereas the coating system with a top homogeneous layer and a thin FGM bottom layer reduces SIF significantly. In the case of μL < μ0 the SIF is found to be larger for the FGM coatings than for the homogeneous coatings. The FGM coating, however, may still be superior to homogeneous coatings in this case as FGM coatings usually provide better bonding strength between the coating and substrate. Finally, the applicability of the SIF concept in the fracture of FGM coatings is discussed. Large modulus gradients in thin coatings may seriously restrict the application of SIFs as the SIF-dominant zone may fall into the crack tip nonlinear deformation and damage zone.
