Passage of the mouse-adapted rabies virus strain CVS-24 (where CVS is challenge virus standard) in BHK cells results in the rapid selection of a dominant variant designated CVS-B2c that differs genotypically and phenotypically from the dominant variant CVS-N2c present in mouse-brain- or neuroblastoma-cell-passaged CVS-24. The glycoprotein of CVS-B2c has 10 amino acid substitutions compared with that of CVS-N2c. Because CVS-B2c can be reproducibly selected in BHK cells, it is likely to be a conserved minor subpopulation of CVS-24. CVS-N2c is more neurotropic in vitro and in vivo than CVS-B2c, which replicates more readily in nonneuronal cells in vitro and in vivo. These characteristics appear to be relevant to the pathogenicity of the two variants. CVS-N2c is more pathogenic for adult mice than CVS-B2c. In contrast, CVS-B2c is more pathogenic for neonatal mice. These differences in pathogenicity are reflected in the selection pattern when mixtures of CVS-N2c and CVS-B2c were used to infect neonatal and adult mice. Although CVS-N2c was highly selected in adult mice, no selection for either variant was seen in neonates, suggesting that certain aspects of development, such as maturation of the nervous and immune systems, may contribute to the selection process. We speculate that the existence of different variants within a rabies virus strain may facilitate the virus in overcoming barriers to its spread, both within the host and between species.  相似文献

AIM: To study the antagonism of l-stepholidine (SPD) against bromocriptine (Bro)-inhibition on prolactin (PRL) level. METHODS: Bro (0.5 mg.kg-1.d-1, s.c.) reduced the PRL and caused a dysplasia of mammary gland in lactational rats. The weight growing of newborn rats was retarded. The PRL of the lactational rats was assessed by immunoradiometric assay (IRMA); the weight of newborn rats and development of mammary glands in lactational rats were also examined. Antagonism of SPD was evaluated. RESULTS: SPD (30 & 100 mg.kg-1.d-1, i.p.) obviously antagonized the Bro that induced lowering the PRL level in lactational rats, the PRL was 11 +/- 4 & 23 +/- 6 micrograms.L-1 (NS 7 +/- 2) respectively on d 15 of postpartum and the development of mammary gland in lactational rats was normal. The newborn rats grew rapidly in 11-15 d. CONCLUSION: SPD possessed an antagonism with Bro inhibition on D2 receptors located in the pituitary gland, and was an antagonist of dopamine D2 receptors.  相似文献

The application of very sensitive photocurrent-based spectroscopic techniques have led to the detection of new levels for the electronic structure of the phosphorous donor in n-type CVD diamond. By combining quasi-steady-state photocurrent measurements (PC), photothermal ionisation spectroscopy (PTIS) and the highly sensitive Fourier transform photocurrent spectroscopy (FTPS) technique at different temperatures, ranging from liquid nitrogen temperature to 170 K, the resulting spectra point to a richer structure than assumed up to now. This is the consequence of the improved sample quality over the last years, opening up to a much larger attainable doping window. By using doping levels, ranging from 1019 cm−3 down to 1016 cm−3 on 111 -oriented Ib HPHT substrates, still giving rise to measurable n-type conductivity, spectra showed less line broadening and more fine structure. Finally, the results will be compared with spectra measured on active P-doped polycrystalline n-type films.  相似文献
