Telomerase is thought to be responsible for cell immortality. The telomerase activity in carcinomas has been remarked since 1995. We examined telomerase activity in colorectal carcinoma by TRAP (Telomeric repeat amplification protocol) assay, and investigated its relationship to clinicopathological findings. We could analyse telomerase activities in 33 cases (66%) of 50 colorectal carcinomas, whereas the activity was not found in all 13 cases of noncancerous colorectal mucosa. There was no relation between the telomerase activity and the clinicopathological findings or metastatic status. We confirmed telomerase activities in much of colorectal carcinomas in spite of their progression. The carcinoma cells might be immortal from their early stage of progression by means of telomerase activity.  相似文献

Integral-skin foams of rigid polyurethane are sandwich structures consisting of a core layer of closed cells enclosed in rigid surface layers on both sides. We examined the layer composition of integral-skin foam with the objective of maximum flexural strength, and then studied possibilities of reconciling the strength and thermal insulating properties in housings for evaporators in car air conditioners; i.e., unit cases. This examination showed that the most practical density range (250 ≦ ρpall ≦ 500 kg/m3) provides vibratile resistance and thermal insulating properties. In actual car-running tests, a maximum 0.1 MPa stress was generated on unit cases with overall densities of 350 kg/m3, We found this to be 0.4% of the flexural strength of an integral-skin foam and 2% of the fatigue strength. In the forcible vibratile test, a stress of 0.5 to 1.0 MPa was generated at the resonance point of a unit case with 250 to 500 kg/m3 overall density. We found that these values are 2 to 5% of integral-skin foam's flexural strength and 10 to 25% of its fatigue strength. These values are of the same level as the conventional unit case made of polypropylene blended with talc. An integral-skin foam with an overall density of 250 kg/m3, nearly equal to half the weight of polypropylene, has the same level of resistance to vibration.  相似文献

A β-alumina tube was formed by hydrostatic pressing of raw powder, lathing and subsequent firing. Preferred orientation of tabular crystallites of β-alumina was determined by X-ray diffraction and polarizing microscope technique. The basal planes of the crystallites had a tendency to be perpendicular to the radial direction of the tube. Ionic conductivity in the axial direction was higher by a factor of 1.5 than that in the radial direction. This was consistent with the result on preferred orientation of the crystallites.  相似文献

Transient expression of genes coding for the poly-gamma-glutamate (gammaPGA) synthetase system (pgs) was investigated in tobacco plants. Three genes of the pgs, pgsA, pgsB and pgsC, were separately placed under the control of the CaMV 35S promoter and introduced into tobacco leaves via Agrobacterium infection. Synthesized gammaPGA in plant tissues was detected immunologically with mouse anti-gammaPGA antiserum which specifically reacts with gammaPGA on a nitrocellulose membrane. Confirmation of gammaPGA biosynthesis in the transient expression analysis in tobacco tissue indicates that subunits of pgs complex were expressed and reassembled in a functional form.  相似文献

An analytical method was developed for the determination of phenmedipham (PM) in agricultural products using reversed-phase high-performance liquid chromatography with UV detection. A sample was extracted with acetonitrile, and the acetonitrile layer was separated by salting-out. The acetonitrile phase was isolated and evaporated. The extract was dissolved in diethyl ether-hexane (1 : 1), and then cleaned up on a Florisil column. The column was washed with diethyl ether-hexane (1 : 1), and PM was eluted with acetone-hexane (3 : 7), and the eluate was evaporated. The residue was dissolved in acetone-hexane (2 : 8), and the sample solution was cleaned up on SAX/PSA cartridge. The SAX/PSA cartridge was washed with acetone-hexane (2 : 8), and PM was eluted with acetone-hexane (3 : 7). If required, the eluate of the Florisil column was cleaned up with SAX/PSA and ENVI-Carb/ NH2 cartridges. The SAX/PSA cartridge was washed with acetone-hexane (2 : 8), and connected to be ENVI-Carb/NH2 cartridge. The cartridges were washed with acetone-hexane (3 : 7), and then the SAX/PSA cartridge was removed. PM was eluted with acetonitrile-toluene (3 : 1) from the ENVI-Carb/NH2 cartridge. PM in the eluate was separated isocratically on an ODS column (4.6 mm i.d. x 150 mm, 5 microm) using acetonitrile-water (6 : 4) as a mobile phase (flow-rate 1.0 mL/min, temp. 40 degrees C), with monitoring at 235 nm. The calibration curve was linear from 0.005 microg/mL to 10 microg/mL of PM. The recoveries of PM from eight kinds of agricultural products spiked at levels of 0.1 and 0.02 microg/g were 80.8-98.7%. To define the etiologic factors of Japanese cerebrovascular diseases, a pathological research was conducted on autopsy cases of stroke in Akita Prefecture. In most cases of cerebral hemorrhages, especially in the middle age group (30-59 of age), arteriosclerotic deviations in basal berebral arteries of the circle of Willis and intracerebral small arteries are not found. The authors conclude that cerebral hemorrhage may occur without relationship to disturbances of lipid metabolism, biochemically, and to atherosclerosis, pathologically. Concerning cerebral infarction, especially in the old generation (over 60 years of age), severe arteriosclerotic deviations were recognized both in basal cerebral and intracerebral arteries. These changes were highly influenced by the grade and duration of hypertension, and rarely influenced by hypercholesterolemia. According to the above-mentioned facts, the results obtained from out epidemiological survey were confirmed by the pathological studies.  相似文献

To investigate the mechanism(s) of degradation of leptin, the protein product of ob (obese) gene, we measured serum leptin levels in 70 patients with chronic renal failure (CRF). The median of serum leptin concentrations of 36 male and 34 female patients with CRF were 7.3 ng/ml ranging from 0.5 to 39.0 ng/ml and 34.9 ng/ml from 1.1 to 76.1 ng/ml, respectively, while those of 29 male and 29 female healthy subjects were 5.8 ng/ml ranging from 0.5 to 37.7 ng/ml and 12.0 ng/ml from 2.0 to 45.2 ng/ml, respectively. The difference in male and female serum leptin concentrations between CRF group and the normal counterpart was statistically significant (p<0.005 and p<0.05, respectively). However, there was no significant correlation, either between serum creatinine or BUN, and serum leptin concentrations. These findings suggest that leptin is degraded and/or filtered in renal tissue.  相似文献
