Despite growing concerns about cross-contamination of ready-to-eat foods with Listeria monocytogenes, our knowledge about the ecology and transmission of L. monocytogenes in retail establishments has remained limited. We conducted a cross-sectional study to characterize the prevalence, distribution, and subtype diversity of L. monocytogenes in 120 New York State retail deli establishments that were hypothesized to present an increased risk for environmental L. monocytogenes contamination (i.e., small establishments and establishments with a history of failed New York State Agriculture and Markets inspections). Analysis of these data along with previously reported data for 121 predominantly larger retail establishments in New York State identified establishment size, geographic location, and inspection history as significant predictors of L. monocytogenes presence and prevalence. The odds of an establishment being L. monocytogenes positive were approximately twice as high for large establishments, establishments located in New York City, or establishments with poor inspection history (as compared with establishments without these attributes), even though correlation between location and inspection history complicated interpretation of results. Within an establishment, L. monocytogenes was significantly more prevalent on nonfood contact surfaces than on food contact surfaces; prevalence was particularly high for floors and in floor drains, sinks, the dairy case, and milk crates. L. monocytogenes subtype diversity differed between sites, with lineage I isolates significantly associated with nonfood contact surfaces and lineage II isolates significantly associated with food contact surfaces. Nonthermal plasma has been shown to be effective in reducing pathogens on the surface of a range of fresh produce products. The research presented here investigated the effectiveness of nonthermal dielectric barrier discharge plasma on Salmonella enterica and Campylobacter jejuni inoculated onto the surface of boneless skinless chicken breast and chicken thigh with skin. Chicken samples were inoculated with antibiotic-resistant strains of S. enterica and C. jejuni at levels of 10(1) to 10(4) CFU and exposed to plasma for a range of time points (0 to 180 s in 15-s intervals). Surviving antibiotic-resistant pathogens were recovered and counted on appropriate agar. In order to determine the effect of plasma on background microflora, noninoculated skinless chicken breast and thighs with skin were exposed to air plasma at ambient pressure. Treatment with plasma resulted in elimination of low levels (10(1) CFU) of both S. enterica and C. jejuni on chicken breasts and C. jejuni from chicken skin, but viable S. enterica cells remained on chicken skin even after 20 s of exposure to plasma. Inoculum levels of 10(2), 10(3), and 10(4) CFU of S. enterica on chicken breast and chicken skin resulted in maximum reduction levels of 1.85, 2.61, and 2.54 log, respectively, on chicken breast and 1.25, 1.08, and 1.31 log, respectively, on chicken skin following 3 min of plasma exposure. Inoculum levels of 10(2), 10(3), and 10(4) CFU of C. jejuni on chicken breast and chicken skin resulted in maximum reduction levels of 1.65, 2.45, and 2.45 log, respectively, on chicken breast and 1.42, 1.87, and 3.11 log, respectively, on chicken skin following 3 min of plasma exposure. Plasma exposure for 30 s reduced background microflora on breast and skin by an average of 0.85 and 0.21 log, respectively. Phytochemical compositions of five varieties of black soybeans (Glycine max) and their stabilities at room temperature, 4 and −80 °C over 14 months were determined by HPLC systems with electrochemical (ECD) and UV detectors. Polyphenol profiling was carried out by a liquid chromatography–electrospray ionisation-mass spectrometry (LC–ESI-MS) with orbitrap as a mass analyser in both positive and negative ion modes, and polyphenols in aglycone forms were quantified by HPLC–ECD. Five different varieties of black soybeans (G. max) contained 249–405 μg/g dry wt of γ-tocopherol and 6.76–14.98 μg/g dry wt of lutein. Major polyphenols in black soybeans (G. max) were daidzein (193–288 μg/g dry wt) and genistein (145–223 μg/g dry wt), mainly present as glucosides and acetyl glucosides. No significant decrease was found in total phenols of black soybeans (G. max) stored at room temperature, 4 or −80 °C for 14 months. On the other hand, lutein and γ-tocopherol degraded significantly within a month of storage at room temperature (p < 0.01), whereas they remained stable up to 6 months at 4 °C and up to 14 months at −80 °C. The current study indicates that black soybeans (G. max) are rich source of γ-tocopherol and phenols (isoflavones, flavonols, flavan-3-ols, proanthocyanidins and anthocyanin) and that the levels vary depending upon varieties. In addition, storage at low temperature is recommended to reduce the loss of fat-soluble phytochemicals in black soybeans (G. max) over an extended period of time. 相似文献

The extractable, protein-bound and fibre-bound condensed tannin (CT) concentrations in the leaves of tropical legumes grown in both Colombia and Northern Australia were determined by the butanol–HCl method, whilst extractable CT was also determined by the vanillin–HCl method. With the exception of Senna siamea all species contained CT. The very high CT concentration found in many plants growing in Colombia may have been partly due to soil fertility being much lower at the Colombian than the Northern Australian site. Acacia boliviana , Arachis pintoi , Centrosema latidens , Senna velutina and Gliricidia sepium contained <55 g total CT kg−1 DM, which suggests that they could comprise a reasonable proportion of ruminant diets. All other species grown in South America contained 100–240 g CT kg−1 DM, which suggests that they should only be fed in small amounts as supplements to dilute the CT concentration. Leucaena species and Calliandra calothyrsus grown in Northern Australia contained intermediate concentrations of total CT (60–90 g kg−1 DM). Most species contained 70–95% of total CT as extractable CT, with the exception of Flemingia macrophylla , where 60% was extractable and 40% bound, and Gliricidia sepium , where almost all the CT was bound to protein. Values for Flemingia macrophylla differed between accessions. Extractable CT determined with vanillin–HCl was generally higher than extractable CT determined with butanol–HCl. Three accessions showed negligible (<1 g kg−1 DM) extractable CT with butanol–HCl but 10–12 g extractable CT kg−1 DM with vanillin–HCl. Two accessions showed undetectable levels of extractable CT but substantial levels of protein-bound CT, illustrating the importance of using a bound CT method for identifying forages containing CT. Relative to freeze drying, oven drying of Leucaena species reduced the concentration of extractable CT and increased concentrations of bound CT. The significance of the results for the nutrition of ruminant livestock are discussed, including the possible roles of protein-bound and fibre-bound CT. 相似文献

Sorghum (cv. Maldandi M35-1) was modified to 12, 14 and 16% moisture content (m.c.) and heat-treated with microwave energy at 3 levels, for 30 sec (=4.5, 9 and 18 kJ), and 60 sec (=9, 18 and 36 kJ). The effect of microwave heating on rise and subsequent fall in grain temperature, reduction in m.c. and quality characteristics including germination, seedling dry matter, free fatty acids (FFA) and contaminant fungi was determined. The temperature attained and the moisture loss in the sorghum grain was affected by grain m.c. and the time of exposure. At the lowest and highest microwave treatment level grain temperatures reached 30–40°C and 90–101°C, respectively. However, a 60-sec treatment at the highest energy level was lethal for the grain, particularly at 14 and 16% m.c. The FFA values were unaffected by microwave treatment. Statistical analyses showed that the microwave power level and time individually, and power level × time interactions were significant for most quality characteristics. The fungi present most abundantly on the sorghum grain were Eurotium spp., Aspergillus candidus, A. niger and Penicillium spp. Increasing m.c. and microwave heating resulted in elimination of most fungi after a 30-sec exposure time. The formation of the components of the cell envelope of Acinetobacter sp. 199A was investigated by measuring the incorporation of [3H]leucine into protein, [14C]galactose into lipopolysaccharide, 32P into phospholipid, and [3H]diaminopimelic acid into peptidoglycan. Whereas the lipopolysaccharide and intrinsic protein of the outer membrane were stable, some of the regularly arranged surface protein, the alpha-protein, was lost into the growth medium. Only newly synthesized alpha-protein was lost. The peptidoglycan of the murein layer was also labile. Selective inhibition of the formation of individual components of the cell envelope with penicillin, chloramphenicol, and bacitracin showed that incorporation of protein into the outer membrane required the simultaneous formation of complete lipopolysaccharide. In human peripheral blood polymorphonuclear leukocytes and lymphocytes, GSH-oxidizing agents promote the movement of surface-bound concanavalin A (Con A) into caps and inhibit the assembly of microtubules (MT) that is normally induced by Con A binding. Con A capping and inhibition of MT assembly occur when GSH levels in cell suspensions are decreased by 30-70%, and return to GSH to control levels is accompanied by the appearance of cytoplasmic MT and by inhibition of the capping response with Con A. Oxidation of GSH markedly stimulates the hexose monophosphate shunt, and regeneration of GSH occurs rapidly. The data indicate that MT cannot be assembled or maintained in the face of decreased GSH levels. Thus, GSH homeostasis becomes critical during physiological events such as phagocytosis which simultaneously induce the assembly of MT and the production of agents like H2O2 that can oxidize GSH. 相似文献

It is argued that affective judgments of a hedonic nature whereby individuals react adversely to an unexpected positive as well as a negative event have been observed on an infrequent basis, primarily because the facilitating conditions have not been specified. Hedonic affective reactions to the disconfirmation of expectations of a recently introduced automobile were examined under 3 conditions representing ego involvement, attitudinal commitment, and task interest. Correlations between degree of absolute disconfirmation and affective judgments made by 488 undergraduates were calculated for 8 treatment combinations. Hedonic reactions were observed only when involvement, commitment, and interest were high. A separate chi-square analysis of the correlation z transformations showed that two-thirds of the cell variance was due to treatment main effects.
