Ultrasonic arrays are increasingly widely used in nondestructive evaluation (NDE) due to their greater flexibility and potentially superior performance compared to conventional monolithic probes. The characterization of small defects remains a challenge for NDE and is of great importance for determining the impact of a defect on the integrity of a structure. In this paper, a technique for characterizing reflectors with subwavelength dimensions is described. This is achieved by post-processing the complete data set of time traces obtained from an ultrasonic array using two algorithms. The first algorithm is used to obtain information about reflector orientation and the second algorithm is used to distinguish between point-like reflectors that reflect uniformly in all directions and specular reflectors that have distinct orientations. Experimental results are presented using a commercial 64-element, 5-MHZ array on two aluminum test specimens that contain a number of machined slots and side-drilled holes. The results show that the orientation of 1-mm-long slots can be determined to within a few degrees and that the signals from 1-mm-long slots can be distinguished from that from a 1-mm-diameter circular hole. Techniques for quantifying both the orientation and the specularity of measured signals are presented and the effect of processing parameters on the accuracy of results is discussed.  相似文献

The biological properties of spherical nucleic acids (SNAs) are largely independent of nanoparticle core identity but significantly affected by oligonucleotide surface density. Additionally, the payload-to-carrier (i.e., DNA-to-nanoparticle) mass ratio of SNAs is inversely proportional to core size. While SNAs with many core types and sizes have been developed, all in vivo analyses of SNA behavior have been limited to cores >10 nm in diameter. However, “ultrasmall” nanoparticle constructs (<10 nm diameter) can exhibit increased payload-to-carrier ratios, reduced liver accumulation, renal clearance, and enhanced tumor infiltration. Therefore, we hypothesized that SNAs with ultrasmall cores exhibit SNA-like properties, but with in vivo behavior akin to traditional ultrasmall nanoparticles. To investigate, we compared the behavior of SNAs with 1.4-nm Au102 nanocluster cores (AuNC-SNAs) and SNAs with 10-nm gold nanoparticle cores (AuNP-SNAs). Significantly, AuNC-SNAs possess SNA-like properties (e.g., high cellular uptake, low cytotoxicity) but show distinct in vivo behavior. When intravenously injected in mice, AuNC-SNAs display prolonged blood circulation, lower liver accumulation, and higher tumor accumulation than AuNP-SNAs. Thus, SNA-like properties persist at the sub-10-nm length scale and oligonucleotide arrangement and surface density are responsible for the biological properties of SNAs. This work has implications for the design of new nanocarriers for therapeutic applications.  相似文献

Phosphate retention plays an important role in the pathogenesis of secondary hyperparathyroidism in patients with renal failure. In in vitro studies, high extracellular phosphate levels directly stimulate PTH secretion in rat and bovine parathyroid tissue. The present study evaluates the effect of high phosphate levels on the secretion of PTH and the production of prepro PTH mRNA in human hyperplastic parathyroid glands. The study includes parathyroid glands obtained from patients with primary adenomas and from hemodialysis and kidney-transplant patients with diffuse and nodular secondary hyperplasia. The experiments were performed in vitro using small pieces of parathyroid tissue. The ability of high calcium levels to decrease PTH secretion was less in adenomas than in secondary hyperplasia; among the secondary hyperplasia, nodular was less responsive to an increase in calcium than diffuse hyperplasia. In diffuse hyperplasia, PTH secretion was increased in response to 3 and 4 mM phosphate compared with 2 mM phosphate, despite a high calcium concentration in the medium; prepro PTH mRNA levels increased after incubation in 4 mM phosphate. Similar results were obtained with nodular hyperplasia, except that the elevation of PTH secretion in response to 3 mM phosphate did not attain statistical significance. In adenomas, high calcium concentrations (1.5 mM) did not result in inhibition of PTH secretion, independent of the phosphate concentration, and the prepro PTH mRNA was not significantly increased by high phosphate levels. In conclusion, first, the PTH secretory response to an increase in calcium concentration is less in nodular than diffuse hyperplasia; second, high phosphate levels directly affect PTH secretion and gene expression in patients with advanced secondary hyperparathyroidism.  相似文献

Although hypertension is a major risk factor for cerebrovascular disease (CVD) and is highly prevalent in African Americans, little is known about how blood pressure (BP) affects brain–behavior relationships in this population. In predominantly Caucasian populations, high BP is associated with alterations in frontal–subcortical white matter and in executive functioning aspects of cognition. We investigated associations among BP, brain structure, and neuropsychological functioning in 52 middle–older-age African Americans without diagnosed history of CVD. All participants underwent diffusion tensor imaging for examination of white matter integrity, indexed by fractional anisotropy (FA). Three regions of interest were derived in the anterior (genu) and posterior (splenium) corpus callosum and across the whole brain. A brief neuropsychological battery was administered from which composite scores of executive function and memory were derived. Blood pressure was characterized by mean arterial blood pressure (MABP). When controlling for age, higher MABP was associated with lower FA in the genu, and there was a trend for this same relationship with regard to whole-brain FA. When the sample was broken into groups on the basis of treatment for BP regulation (medicated vs. nonmedicated), MABP was related to genu and whole-brain FA only in the nonmedicated group. Neither MABP nor FA was significantly related to either neuropsychological composite score regardless of medication use. These data provide important evidence that variation in BP may contribute to significant alterations in specific neural regions of white matter in nonmedicated individuals without symptoms of overt CVD. In this study Inconel 718 cylinders were fabricated by selective laser melting in either argon or nitrogen gas from a pre-alloyed powder. As-fabricated cylinders oriented in the build direction (z-axis) and perpendicular to the build direction (x-axis) exhibited columnar grains and arrays of γ″ (body-centered tetragonal) Ni3Nb oblate ellipsoidal precipitates oriented in a strong [2 0 0] texture determined by combined optical metallography, transmission electron microscopy, and X-ray diffraction analysis. Fabricated and hot isostatic pressed (HIP) components exhibited a more pronounced [2 0 0] columnar γ″ phase precipitate architecture parallel to the laser beam or build direction (spaced at ∼0.8 μm), and a partially recrystallized fcc grain structure. Fabricated and annealed (1160 °C for 4 h) components were ∼50% recrystallized and the recrystallized regions contained spheroidal γ′ precipitates distributed in a dense field of fine γ″ precipitates. The γ″ precipitates were always observed to be coincident with {1 0 0} planes of the γ-fcc NiCr matrix. Some δ phase precipitates in the unrecrystallized/recrystallized interfaces and recrystallized grain boundaries were also observed in the annealed samples. The microindentation (Vickers) hardness was 3.9 GPa for the as-fabricated materials, 5.7 GPa for the HIP material, and 4.6 GPa for the annealed material. Corresponding tensile properties were comparable with wrought Inconel 718 alloy.  相似文献

The circadian clock (CC) is a daily system that regulates the oscillations of physiological processes and can respond to the external environment in order to maintain internal homeostasis. For the functioning of the CC, the clock genes (CG) act in different metabolic pathways through the clock-controlled genes (CCG), providing cellular regulation. The CC’s interruption can result in the development of different diseases, such as neurodegenerative and metabolic disorders, as well as cancer. Leukemias correspond to a group of malignancies of the blood and bone marrow that occur when alterations in normal cellular regulatory processes cause the uncontrolled proliferation of hematopoietic stem cells. This review aimed to associate a deregulated CC with the manifestation of leukemia, looking for possible pathways involving CG and their possible role as leukemic biomarkers.  相似文献
