Sphingomyelinase C (SMC) was purified to homogeneity from the culture supernatant of Streptomyces griseocarneus NBRC13471. The purified enzyme appeared as a single band of 38 kDa by using an electropherogram trace. The molecular mass of the enzyme as determined by MALDI-TOF MS was 32,102 Da, indicating that SMC is monomeric in nature. Under experimental conditions, the highest enzyme activity was found at pH 9.0 and 50–55 °C, and the enzyme was stable from pH 5 to 10 and up to 37 °C. The SMC activity requires Mg2+ or Mn2+ and the order of potency to enhance the activity was Zn2+ ≥ Mn2+ > Cu2+ ≥ Fe2+. Phenylmethylsulfonyl fluoride and EDTA inhibited the enzyme activity, showing that SMC belongs to a group of metalloenzymes and a class of serine hydrolases. The enzyme activity was inhibited by DTT, but not by mercaptoethanol and iodoacetamide. SDS inhibited the enzyme activity; by contrast, Triton X-100 stimulated the activity. The N-terminal and internal amino-acid sequences were determined as H2N-APAAATPSLK, AREIAAAGFFQGND, and NTVVQETSAP. The gene encoding SMC consisted of 1020 bp encoding a signal peptide of 42 amino acids and a mature protein of 297 amino acids with a calculated molecular mass of 32,125 Da. The conserved region of DNase I-like family enzymes and the amino acid residues that are highly conserved in the active center of other bacterial SMCs were also found in the deduced amino acid sequence of S. griseocarneus SMC. 相似文献

Tunable distributed feedback lasing was achieved in a guest-host polymeric waveguide using holographic dynamic gratings. Tris(8-quinolinolato)aluminum (Alq) as a host and 4-(dicyanomethylene)-2-methyl-6-(4-dimethylaminostyryl)-4H-pyran (DCM) as a guest emitter were dispersed in poly(N-vinyl carbazole) (PVCz) matrix. Emission of both the TE(0) and the TM(0) modes was measured. Effective energy transfer from PVCz to DCM through Alq assisted in the reduction of the threshold of laser emission. The threshold of 38 nJ/pulse was measured for first-order lasing and that of 53 nJ/pulse for second-order lasing. 相似文献

Bacterial spores (Bacillus subtilis subsp. subtilis NBRC 16183) inoculated onto a stainless steel Petri dish and treated at nine levels of water activity (a(w)) for 2 days were inactivated by infrared radiation heating (IRH) using three kinds of infrared heaters with different radiation spectra. The peak wavelengths used were 950, 1,100 and 1,150 nm. In general, the inactivating efficacy of IRH treatment against bacterial spores with shorter wavelength heater (950 nm) was greater than that with other heaters. The decimal reduction times (D value) calculated using the linear portion of survival curves were affected by both the initial a(w) values and the spectra of the infrared rays. Spores at approximately 0.9, 0.7 and 0.6 a(w) were most resistant to IRH at wavelengths of 950, 1,100 and 1,150 nm, respectively. The a(w) values that led to maximum D values for bacterial spores increased as the wavelength was shortened. Optimum a(w) values were identified for the inactivation of bacterial spores by IRH. Spore resistance to IRH could also be affected by the spectral characteristics of the infrared absorption, which varied with the a(w) of bacterial spores. 相似文献

We present significant electrical conductivity responses of the pelletized as-prepared and oxidized (ox-) single-wall carbon nanohorns (SWNHs) on adsorption of CO(2) and O(2). The morphological and pore structures of both pelletized SWNHs were examined by transmission electron microscopy (TEM) and nitrogen adsorption isotherm, leading to explicit evidences of the formation of nanoscale windows on the wall by oxidation. The SWNH and ox-SWNH induced a semiconducting behavior, strongly responded to CO(2) and O(2) adsorptions, and each exhibited n-type- and p-type-like conductivities. The electrical conductivity increase and decrease for CO(2) and O(2) adsorption, respectively, were observed for SWNH, whereas ox-SWNH showed a marked electrical conductivity drop on CO(2) adsorption and almost no change on O(2) adsorption. The dramatically different electrical conductivity response of ox-SWNH is presumed to be ascribed to the annihilation of pentagons in the single graphene wall by oxidation. 相似文献

Paclitaxel is an essential drug in the chemotherapy of ovarian, non-small cell lung, breast, gastric, endometrial, and pancreatic cancers. However, it frequently causes peripheral neuropathy as a dose-limiting factor. Animal models of paclitaxel-induced peripheral neuropathy (PIPN) have been established. The mechanisms of PIPN development have been elucidated, and many drugs and agents have been proven to have neuroprotective effects in basic studies. In addition, some of these drugs have been validated in clinical studies for their inhibitory PIPN effects. This review summarizes the basic and clinical evidence for therapeutic or prophylactic effects for PIPN. In pre-clinical research, many reports exist of neuropathy inhibitors that target oxidative stress, inflammatory response, ion channels, transient receptor potential (TRP) channels, cannabinoid receptors, and the monoamine nervous system. Alternatively, very few drugs have demonstrated PIPN efficacy in clinical trials. Thus, enhancing translational research to translate pre-clinical research into clinical research is important. 相似文献

Commercial purity aluminum (1100Al) bars were severely plastic deformed by torsion deformation at room temperature. The specimens were deformed to ultrahigh equivalent strain of 5.85 in maximum. Microstructure evolution during the torsion deformation was characterized using electron back scatter diffraction analysis on two different sections: the longitudinal section parallel to the torsion axis and transverse section perpendicular to the torsion axis. The grain size decreased and the fraction of high angle grain boundary increased with increasing equivalent strain. Elongated ultrafine grained structure was obtained after an equivalent strain of 3.27. We have found that the microstructure evolution in the specimen deformed by torsion exhibited similar behavior to those in the same material heavily deformed by accumulative roll bonding. The average grain size of 0.32 μm with the high angle boundary fraction of 0.76 was achieved in the specimen deformed to an equivalent strain of 5.27. Though the microstructure and hardness on the transverse section varied depending on the radial positions, they could be arranged as a simple function of equivalent strain.
