Feeding of butyrate was found to have a positive effects in enhancing gut development and improving growth performance of calves. Equally, glucagon-like peptide 1 and 2 (GLP-1 and GLP-2), secreted from gastrointestinal L-cells in response to nutrient intake, were found to play a significant role in regulating blood glucose homeostasis and improving gut health. However, limited information is available about the relationship between butyrate and release of GLP-1 and GLP-2 in dairy calves. The objective of this study was to evaluate the effects of a pulse-dose ruminal infusion of butyrate on plasma GLP-1 and GLP-2 concentrations in dairy calves. Five ruminally cannulated mature Holstein bull calves (7.2 ± 0.10 mo, and 330 ± 16.0 kg of body weight; mean ± standard deviation) were used in a 5 × 5 Latin square with 4-d periods. On d 1 of each period at 0800 h, calves were ruminally infused with 1 of 5 treatments: 0 (saline), 0.3, 0.6, 0.9, and 1.2 g of butyrate per kg of body weight. Before butyrate infusion, calves were not offered feed overnight, and sequential blood and rumen fluid samples were taken before and after infusion on d 1 of each period. Ruminal butyrate and total volatile fatty acid concentrations increased linearly (2.65, 12.19, 20.99, 30.19, and 36.30; 23.68, 33.07, 40.94, 51.13, and 56.31 µmol/mL, for butyrate and total volatile fatty acids, respectively) in a dose-dependent manner, whereas propionate and isobutyrate increased quadratically. Ruminal and plasma butyrate, β-hydroxybutyrate, GLP-1, GLP-2, insulin, and glucose concentrations were all affected by treatment, time (except GLP-2), and interaction of treatment with time (except GLP-1). The area under the curve (AUC) summarized at different time points relative to the baseline (AUC30, AUC60, AUC120, and AUC240) for ruminal and plasma butyrate, and BHB, increased linearly with the dose of butyrate infused. However, AUC30, AUC60, AUC120, and AUC240 for plasma GLP-2 concentration were affected in a cubic manner unlike the linear effect on AUC30 and AUC60 for GLP-1. Plasma GLP-2 was not correlated with plasma butyrate (r = 0.16), GLP-1 (r = 0.03), or BHB (r = ?0.05). This findings suggest that pulse-dosing of butyrate slightly increased both GLP-1 and GLP-2 concentrations at specific time points and this might be promoted by direct or indirect effect of butyrate on the intestinal L-cells.  相似文献

The objective of this study was to evaluate periprandial plasma concentrations of glucagon-like peptide-2 (GLP-2), glucose, and β-hydroxybutyrate (BHB) in response to a milk meal in preweaning dairy calves. Nineteen Holstein heifer calves were fed either a high (10 L/d; n = 9) or low (5 L/d; n = 10) amount of pasteurized whole milk from d 2 to 50 of life. Calves were housed in individual pens for the first 19 ± 3 d and fed only milk before being moved to a group pen, where they remained on their respective milk treatment and offered calf starter ad libitum. Blood samples were collected sequentially for 240 min following their milk meal at wk 3, 5, and 7 of life to characterize the periprandial response in plasma concentrations of GLP-2, glucose, and BHB. Baseline plasma glucose concentrations were increased, when a high amount was fed; however, we found no difference in area under the curve. Feeding a high amount of whole milk had no effect on baseline or periprandial plasma BHB concentrations. Baseline plasma GLP-2 concentrations decreased as calves aged. Feeding a high amount of whole milk tended to significantly increase baseline GLP-2 concentrations throughout compared with calves fed a low amount. The periprandial response of GLP-2 was not biphasic until calves were 7 wk old. In conclusion, feeding a high amount of milk may increase GLP-2 concentrations in preweaning calves, although its exact mechanism is unknown.  相似文献

This review paper focuses on the recent studies of electroactive polymer actuators that have a triple‐layered configuration composed of an ionic‐gel electrolyte layer sandwiched by nano‐carbon dispersed ionic‐liquid gel electrode layers (bucky‐gel actuator) for the purpose of development of practical devices. The review covers recent studies of the developments of the materials of the bucky‐gel actuators and their electromechanical modeling. Prevention of porosity in partial penetration fibre laser and fibre laser–gas metal arc hybrid welding is investigated. It is found that modulation of laser power prevents porosity formation in fibre laser welding, but that this method is not effective in hybrid welding. However, the addition of a small amount of oxygen to the molten pool can prevent porosity formation in both fibre laser and hybrid welding. This is attributed to stabilisation of the keyhole. During welding, oxygen reacts with dissolved carbon to form carbon monoxide (CO) in the keyhole. The CO partial pressure in the keyhole prevents the intense interaction between laser beam and molten metal, thus stabilising the keyhole. The role of CO formation is confirmed by the enhancement of porosity suppression with increased carbon content of the base metal.  相似文献

The effects of single-walled carbon nanotubes (SWCNTs) on the properties of carbide-derived carbon (CDC)-based electroactive polymer (EAP) actuators were studied. SWCNTs were used as an additive to increase the mesoporosity and electrical conductivity of the electrodes, and also to support the CDC matrix. EAP actuators with various ratios of SWCNTs to CDC in the electrodes were fabricated and their electromechanical and electrochemical characteristics were examined. The addition of SWCNTs to CDC-based electrodes significantly increased the bending strain and stress (bending force) of the actuators. The actuator assembled with electrodes containing SWCNTs and CDC in the ratio 50/50 (wt.%/wt.%) showed the highest strain output among the samples at lower frequencies (<0.1 Hz). The increase in maximum strain was more than twice that of pure CDC-based EAPs (0.85% vs. 0.35% at an applied voltage of ±2 V). Also, the high frequency (5–50 Hz) response of the combined SWCNT/CDC-based actuators was considerably improved.  相似文献

PURPOSE: To characterize changes in the retina, retinal pigment epithelium (RPE), and choriocapillaris with fluorescein angiography (FA) and histology after hydraulic or abrasive RPE debridement in 26 domestic short-haired cats. METHODS: Hydraulic debridement was produced by injecting balanced salt solution forcefully into the subretinal space. For abrasive debridement, RPE were removed with a silicone-tipped cannula after creating a localized retinal detachment. The FAs were performed after surgery, and tissue was prepared for light microscopy (LM) and scanning electron microscopy (SEM). RESULTS: Sixty-seven blebs were examined by FA 1 hour after surgery, and RPE debridement was confirmed by SEM or LM in 15 blebs from 10 animals. Hyperfluorescence and variable central fluorescein leakage were seen 1 week after surgery in 52 of 53 blebs (which includes all 27 blebs from the 1-week timepoint and 26 of 29 blebs from the 4-week timepoint that were studied by FA 1 week after surgery). Choriocapillary filling delays were seen in no hydraulic debridements, but in 11 of 14 abrasive blebs, especially in areas showing leakage late in the angiogram. In 1 of 13 hydraulic and 12 of 14 abrasive debridements, areas of late dye leakage had no RPE with outer retinal degeneration. At the 4-week timepoint, 1 of 17 hydraulic and 10 of 12 abrasive debridements had foci of delayed or absent choriocapillary perfusion by FA, with degenerated outer retina, no RPE, and choriocapillary atrophy by histologic analysis. CONCLUSIONS: Abrasive debridement is more commonly associated with abnormal FAs and with incomplete RPE repopulation, choriocapillaris atrophy, and outer retinal degeneration than is hydraulic debridement. This clinicopathologic study may give insight into FA interpretation after choroidal neovascular membrane removal in human patients.  相似文献

We studied 38 superficial-type hyperplastic polyps of more than 0.6 cm in size on radiographs to examine their morphologic features. The 38 lesions ranged in size from 0.6 cm to 2.5 cm, and 22 were located in the sigmoid and transverse colon (Table 1, Fig.1). The cross-section and surface configurations of the 38 lesions studied radiographically are illustrated in Fig. 2. 24 lesions without central depression were classified into 5 groups (1-a to 1-e). The 14 other lesions had central depression (2-a). The cross-section configuration was sharp in all lesions, and a finely serrated arrangement was seen in 30 of them. In surface configuration, several granular patterns were seen, but full-blown undulated folds and/or regular granular patterns were characteristic in groups 1-c, 1-d and 2-a. These radiologic features of cross-section and surface configuration may be regarded as a possible means of distinguishing superficial-type hyperplastic polyps from adenomas and carcinomas of the large intestine.  相似文献

Incidence of Kawasaki disease in Japan is 10 times higher than in the United States. Approximately 10% of patients have atypical clinical presentations. Because echocardiographic or angiographic evidence of coronary artery complications is needed for diagnosis, such atypical cases often result in either delay or omission of intravenous gamma globulin therapy and higher incidence of coronary artery aneurysms than those patients with classical presentations. Despite ongoing research, no universally accepted etiologic theory exists today. Intravenous gamma globulin remains the mainstay of the acute phase treatment of Kawasaki disease. However, reported transmission of hepatitis C virus via a brand of intravenous gamm globulin leaves us with a lingering concern about the safety of this treatment. In terms of long-term follow-up, emergencence of a number of newer diagnostic modalities is promising and warrants careful evaluation. Surgical therapy for coronary artery disease needs to be approached cautiously.  相似文献

Interleukin (IL)-6, a multifunctional cytokine originally described as a T cell-derived factor, is also produced by different cell types, and it influences a wide variety of physiological and pathophysiological processes. Recent studies further suggest that IL-6 has a role in down-regulating hormone production by endocrine organs and can negatively affect the steroidogenic capacity of both ovaries and testes. Thus, the aims of this investigation were to examine whether IL-6 plays a role in luteolysis and, more specifically, to determine whether luteal cells express the IL-6 gene, whether this expression is developmentally and hormonally regulated in pregnancy, and whether the corpus luteum could be a target for IL-6 action. Using semiquantitative RT-PCR, messenger RNA (mRNA) encoding both components of the IL-6 receptor [the ligand-binding subunit (IL-6 R) and the IL-6 R-associated signal transducer (gp130)] were found to be highly expressed in corpora lutea throughout pregnancy. In contrast, IL-6 mRNA expression was barely detectable from day 4 through the end of pregnancy, whereas a sharp and abrupt expression of IL-6 mRNA occurred immediately after parturition. Although the corpus luteum does not express IL-6 mRNA during most of pregnancy, it could be induced to express this gene with an in vivo injection of the bacterial endotoxin, lipopolysaccharide. In addition, when corpora lutea from day-15 pregnant rats were isolated and maintained in culture, IL-6 mRNA that was undetectable at 0 h increased in a time-related manner and reached significant levels after 4 h of incubation, followed by a similar increase in IL-6 protein secreted in the culture media. Isolation of the small and large luteal cells by elutriation indicated that both cell populations can secrete IL-6 in culture. The apparent ability of luteal cells to spontaneously express IL-6 in vitro, together with the lack of IL-6 expression during most of pregnancy, led us to examine whether the IL-6 gene is silenced throughout pregnancy by luteotropic hormones. Corpora lutea from day-15 pregnant rats were cultured in the presence of different doses of progesterone; the synthetic glucocorticoid, dexamethasone; 17beta-estradiol; and PRL. Progesterone and dexamethasone markedly inhibited IL-6 mRNA expression, whereas 17beta-estradiol had a minimal inhibitory effect, and PRL did not affect IL-6 mRNA expression. In summary, results of this investigation have revealed that the rat corpus luteum expresses the IL-6 receptor system and that luteal cells are able to secrete IL-6. However, IL-6 gene expression is silenced during most of pregnancy, probably by the high levels of progesterone locally produced in the corpus luteum. The salient finding that progesterone and glucocorticoid strongly inhibit the expression of IL-6 in the corpus luteum suggests that one important luteotropic role of progesterone and glucocorticoids could be to prevent the expression of IL-6, which might have a deleterious effect on luteal function.  相似文献

Marked T wave abnormalities in 12-lead electrocardiograms often appear after radiofrequency ablation in patients with Wolff-Parkinson-White (WPW) syndrome. Durations of the QRS complex before ablation have been reported to be significantly greater in patients with repolarization abnormalities after ablation than in those with normal repolarization. However, there has been no report concerning the relationship between QRS duration and repolarization abnormalities before ablation. It has been reported that QRST isointegral maps are dependent on local cellular repolarization properties but largely independent of activation sequence, and that they provide quantitative information about repolarization abnormalities. Isointegral maps were constructed from data recorded in 34 patients with WPW syndrome and QRS durations of 0.1 second or longer (21 patients with left-sided accessory pathways and 13 with right-sided pathways) in order to investigate the relationship between QRS duration and abnormalities in repolarization properties during preexcitation. Body surface ECGs were recorded with an 87-lead system at a sampling interval of 1 ms. The QRST values (mean +/- 2SD) from 607 normal individuals were taken to represent the normal range. The area with abnormally low QRST values was designated as the -2SD area and the sum of the QRST values (sigma QRST) in this area was calculated. The QRS duration was found to be significantly related to sigma QRST. These results demonstrate that the patients with WPW syndrome had abnormalities in local repolarization properties that were significantly related to QRS duration during preexcitation. The degree of preexcitation may be important in producing these abnormalities in patients with manifest WPW syndrome. This may result in T wave abnormalities after ablation of the accessory pathway attributable to cardiac memory.  相似文献
