The numbers of obese people and diabetic patients are ever increasing. Obesity and diabetes are high-risk conditions for chronic diseases, including certain types of cancer, such as colorectal cancer (CRC). The aim of this study was to develop a novel animal model in order to clarify the pathobiology of CRC development in obese and diabetic patients. We developed an animal model of obesity and colorectal cancer by breeding the C57BL/KsJ-db/db (db/db) mouse, an animal model of obesity and type II diabetes, and the C57BL/6J-Apc(Min/+) (Min/+) mouse, a model of familial adenomatous polyposis. At 15 weeks of age, the N9 backcross generation of C57BL/KsJ-db/db-Apc(Min/+) (db/db-Min/+) mice developed an increased incidence and multiplicity of adenomas in the intestinal tract when compared to the db/m-Min/+ and m/m-Min/+ mice. Blood biochemical profile showed significant increases in insulin (8.3-fold to 11.7-fold), cholesterol (1.2-fold to 1.7-fold), and triglyceride (1.2-fold to 1.3-fold) in the db/db-Min/+ mice, when compared to those of the db/m-Min/+ and m/m-Min/+ mice. Increases (1.4-fold to 2.6-fold) in RNA levels of insulin-like growth factor (IGF)-1, IRF-1R, and IGF-2 were also observed in the db/db- Min/+ mice. These results suggested that the IGFs, as well as hyperlipidemia and hyperinsulinemia, promoted adenoma formation in the db/db-Min/+ mice. Our results thus suggested that the db/db-Min/+ mice should be invaluable for studies on the pathogenesis of CRC in obese and diabetes patients and the therapy and prevention of CRC in these patients.  相似文献

In this study, complexes composed of poly-l-tyrosine (pLT) and single-walled carbon nanotubes (SWCNTs) were produced and the dispersibility of the pLT/SWCNT complexes in water by measuring the ζ potential of the complexes and the turbidity of the solution were investigated. It is found that the absolute value of the ζ potential of the pLT/SWCNT complexes is as high as that of SWCNTs modified with double-stranded DNA (dsDNA) and that the complexes remain stably dispersed in the water at least for two weeks. Thermogravimetry analysis (TGA) and visualization of the surface structures of pLT/SWCNT complexes using an atomic force microscope (AFM) were also carried out.  相似文献

In the previous study, we successfully prepared a chitin nanofiber film by regeneration from a chitin ion gel with an ionic liquid using methanol. In this study, we performed surface-initiated graft polymerization of γ-benzyl l-glutamate N-carboxyanhydride (BLG-NCA) from amino groups on a partially deacetylated chitin nanofiber (PDA-CNF) film. First, the chitin nanofiber film was immersed in 40 % NaOH aq. at 80 °C for 7 h for partial deacetylation. Then, the PDA-CNF film was immersed in a solution of BLG-NCA in ethyl acetate at 0 °C for 24 h for graft polymerization from amino groups on nanofibers to give a chitin nanofiber-graft-poly(γ-benzyl l-glutamate) (CNF-g-PBLG) film. The analytical results of the film indicated that graft polymerization of BLG-NCA occur on surface of nanofibers. Furthermore, the film was treated with 1.0 mol/L NaOH aq. to convert PBLG on nanofibers into poly(γ-l-glutamic acid sodium salt) (PLGA). Then, condensation of the resulting carboxylates with amino groups at the terminal ends of PLGAs or the remaining amino groups on nanofibers was performed using the condensing agent to produce a CNF-g-PLGA network film. The resulting film showed the good mechanical properties with high flexibility, which has potentials as promising materials for practical applications.  相似文献

The dynamic behavior of liquid water transport through the gas diffusion layer (GDL) of the proton exchange membrane fuel cell is studied with an ex-situ approach. The liquid water breakthrough pressure is measured in the region between the capillary fingering and the stable displacement on the drainage phase diagram. The variables studied are GDL thickness, PTFE/Nafion content within the GDL, GDL compression, the inclusion of a micro-porous layer (MPL), and different water flow rates through the GDL. The liquid water breakthrough pressure is observed to increase with GDL thickness, GDL compression, and inclusion of the MPL. Furthermore, it has been observed that applying some amount of PTFE to an untreated GDL increases the breakthrough pressure but increasing the amount of PTFE content within the GDL shows minimal impact on the breakthrough pressure. For instance, the mean breakthrough pressures that have been measured for TGP-060 and for untreated (0 wt.% PTFE), 10 wt.% PTFE, and 27 wt.% PTFE were 3589 Pa, 5108 Pa, and 5284 Pa, respectively.  相似文献

The arrangement of permanent magnets in double‐layer interior permanent‐magnet motors is optimized for variable‐speed applications. First, the arrangement of magnets is decided by automatic optimization. Next, the superiority of the optimized motor is discussed by considering d‐ and q‐axis equivalent circuits that take into account the magnetic saturation of the rotor core. Finally, experimental veri?cation is carried out with a prototype motor. It is con?rmed that the maximum torque of the optimized motor under both low‐speed and high‐speed conditions is higher than that of conventional motors because of the relatively large q‐axis inductance and small d‐axis inductance. © 2013 Wiley Periodicals, Inc. In Pb–Bi-cooled direct contact boiling water small fast reactor (PBWFR), steam is generated by direct contact of feedwater with primary Pb–Bi coolant above the core, and Pb–Bi coolant is circulated by steam lift pump in chimneys. Safety design has been developed to show safety features of PBWFR. Negative void reactivity is inserted even if whole of the core and upper plenum are voided hypothetically by steam intrusion from above. The control rod ejection due to coolant pressure is prevented using in-vessel type control rod driving mechanism. At coolant leak from reactor vessel and feedwater pipes, Pb–Bi coolant level in the reactor vessel required for decay heat removal is kept using closed guard vessel. Dual pipes for feedwater are employed to avoid leak of water. Although there is no concern of loss of flow accident due to primary pump trip, feedwater pump trip initiates loss of coolant flow (LOF). Injection of high pressure water slows down the flow coast down of feedwater at the LOF event. The unprotected loss of flow and heat sink (ATWS) has been evaluated, which shows that the fuel temperatures are kept lower than the safety limits.  相似文献

The purpose of this study is to develop electrodes for electrical stimulation of the nervous system using conductive polymers, polypyrrole (PPy) and/or poly(3,4-ethylenedioxythiophene), PEDOT. We evaluated biocompatibility in fibroblast and/or myoblast of mouse. Cultured cells on PPy and/or PEDOT extended their neuritis and survived over a week. These experiments have demonstrated that conductive polymers such as PPy, PEDOT, etc. have high biocompatibility, and PPy and/or PEDOT are applicable to nerve stimulation electrodes.  相似文献

The final stage of a postulated energetic core disruptive accident (CDA) in a liquid metal fast breeder reactor is believed to involve the expansion of a high-pressure core-material bubble against the overlying pool of sodium. Some of the sodium will be entrained by the CDA bubble which may influence the mechanical energy available for damage to the reactor vessel. The following considerations of liquid surface instability indicate that the Kelvin–Helmholtz (K–H) mechanism is primarily responsible for liquid entrainment by the expanding CDA bubble. First, an instability analysis is presented which shows that the K–H mechanism is faster than the Taylor acceleration mechanism of entrainment at the high fluid velocities expected within the interior of the expanding CDA bubble. Secondly, a new model of liquid entrainment by the CDA bubble is introduced which is based on spherical-core-vortex motion and entrainment via the K–H instability along the bubble surface. The model is in agreement with new experimental results presented here on the reduction of nitrogen-gas-simulant CDA bubble work potential. Finally, a one-dimensional air-over-water parallel flow experiment was undertaken which demonstrates that the K–H instability results in sufficiently rapid and fine liquid atomization to account for observed CDA gas-bubble work reductions. An important byproduct of the theoretical and experimental work is that the liquid entrainment rate is well described by the Ricou–Spalding entrainment law.  相似文献

A photosensitive TiO2 thin film embedded with Ag nanoparticles has been prepared from a Ti(OBu)4–acetylacetone solution, containing dispersed Ag nanoparticles, by the sol–gel method. UV–visible absorption spectra showed that the thin film obtained has two absorption bands, characteristic of the acetylacetone chelate rings and plasmon resonance from Ag nanoparticles. After the irradiation of UV light, the absorption band from the chelate rings almost disappeared, ascribed to structural changes associated with dissociation of the chelate rings. The thin film after the UV irradiation exhibited a broad absorption band in the IR spectrum, indicating that a Ti–O–Ti network was formed in the thin film. HRTEM and EDX spectra revealed that Ag nanoparticles were present and dispersed in the TiO2 thin film. Micro-patterns of 50 μm dots have been fabricated by UV irradiation through a corresponding photomask, followed by leaching.  相似文献
