The diffusion coefficient (D) values of tert‐butyloxycarbonyl‐glycine, tert‐butyloxycarbonyl‐L ‐tryptophan, tert‐butyloxycarbonyl‐L ‐phenylalanine (Boc‐Phe), and 9‐fluorenylmethoxycarbonyl‐L ‐phenylalanine in Merrifield polystyrene (MPS) gels, poly(ethylene glycol)‐grafted polystyrene (PEG–PS) gels, and crosslinked ethoxylate acrylate (CLEAR) gels, as used in solid‐phase peptide synthesis, were determined by the pulsed‐field‐gradient spin‐echo 1H‐NMR method. From these experimental results, it was found that the amino acids in MPS gels, PEG–PS gels, and CLEAR gels with N,N‐dimethylformamide‐d7 (DMF‐d7) as a solvent had multidiffusion components within a measurement timescale of 10 ms. The D value of Boc‐Phe in polystyrene gels (1% divinylbenzene crosslinked) with tetrahydrofuran‐d8 was much larger than that in the same gels with DMF‐d7. Furthermore, the required time in which an amino acid transferred from a reactive site to a reactive site was estimated, within which the solvents and amino acids in the polymer supports diffused in the swollen beads.© 2003 Wiley Periodicals, Inc. To examine methods for reducing the amount of adsorbed protein on the surface of contact lenses during use, cationic copolymers containing poly(ethylene oxide) units were synthesized and evaluated as surface modifiers. Poly(ethylene oxide) graft‐branched copolymers of composition 70 mol % dimethylaminoethyl methacrylate (DM) and 30 mol % methoxy polyethylene glycol methacrylate (Mp0G; p = 2, 4, 9; the average number of the ethylene oxide units) were obtained using nonionic monomers containing poly(ethylene oxide) units. The copolymers very efficiently prevented protein adsorption on a contact lens. Contact angle measurements showed that immersion in tear fluid made the lens surface hydrophobic because of adsorption of proteins with hydrophobic residues. The copolymer pretreatment made the lens surface hydrophilic, even after dipping in artificial tear fluid. These results suggest that adsorption of the poly(ethylene oxide) branched copolymer on the contact lens would make the lens surface hydrophilic and prevent protein adsorption. © 2012 Wiley Periodicals, Inc. Anatase-type TiO2 powder containing sulfur with absorption in the visible region was directly formed as particles with crystallite in the range 15–88 nm by thermal hydrolysis of titanium(III) sulfate (Ti2(SO4)3) solution at 100°–240°C. Because of the presence of ammonium peroxodisulfate ((NH4)2S2O8), the yield of anatase-type TiO2 from Ti2(SO4)3 solution was accelerated, and anatase with fine crystallite was formed. Anatase-type TiO2 doped with ZrO2 up to 9.8 mol% was directly precipitated as nanometer-sized particles from the acidic precursor solutions of Ti2(SO4)3 and zirconium sulfate in the presence and the absence of (NH4)2S2O8 by simultaneous hydrolysis under hydrothermal conditions at 200°C. By doping ZrO2 into TiO2 and with increasing ZrO2 content, the crystallite size of anatase was decreased, and the anatase-to-rutile phase transformation was retarded as much as 200°C. The anatase-type structure of ZrO2-doped TiO2 was maintained after heating at 1000°C for 1 h. The favorable effect of doping ZrO2 to anatase-type TiO2 on the photocatalytic activity was observed.  相似文献

Anatase-type TiO2 solid solutions doped with 0–10 mol% scandium were formed by hydrothermal crystallization under weak basic conditions above 180°C for 5 h from amorphous co-precipitates that were obtained from the aqueous precursor solutions of TiOSO4 and Sc(NO3)3 using aqueous ammonia. The anatase particles were spindle-like and consisted of nanosized-crystallites (23–25 nm). The lattice parameter c 0 of anatase and the length and width of the spindle-like anatase gradually increased when the scandium content was increased. The diffuse reflectance spectra of the as-prepared TiO2 doped with scandium showed that the onset of absorption slightly shifted to longer wavelengths with increasing scandium content. The band gap of anatase was slightly increased by making solid solutions with scandium oxide.  相似文献

Capuramycins are one of several known classes of natural products that contain an l ‐Lys‐derived l ‐α‐amino‐?‐caprolactam (l ‐ACL) unit. The α‐amino group of l ‐ACL in a capuramycin is linked to an unsaturated hexuronic acid component through an amide bond that was previously shown to originate by an ATP‐independent enzymatic route. With the aid of a combined in vivo and in vitro approach, a predicted tridomain nonribosomal peptide synthetase CapU is functionally characterized here as the ATP‐dependent amide‐bond‐forming catalyst responsible for the biosynthesis of the remaining amide bond present in l ‐ACL. The results are consistent with the adenylation domain of CapU as the essential catalytic component for l ‐Lys activation and thioesterification of the adjacent thiolation domain. However, in contrast to expectations, lactamization does not require any additional domains or proteins and is likely a nonenzymatic event. The results set the stage for examining whether a similar NRPS‐mediated mechanism is employed in the biosynthesis of other l ‐ACL‐containing natural products and, just as intriguingly, how spontaneous lactamization is avoided in the numerous NRPS‐derived peptides that contain an unmodified l ‐Lys residue.  相似文献

Dicalcium silicate, which is found in steelmaking slag for dephosphorization, exists as the hexagonal α phase at high temperatures. The α-dicalcium silicate forms a solid solution with tricalcium phosphate in the entire composition range, although the reason for high solubility of phosphorus remains unclear in view of the crystal structure. It has previously been reported that the crystal structure of α-dicalcium silicate consists of a symmetric arrangement of Ca2+ ions and SiO44− tetrahedra, although other polymorphs exhibit asymmetric arrangements. However, because the occupation probability of each atomic site in the α polymorph is not limited to unity, it has not been qualified how these ions are exactly arranged. In this study, the ionic distribution in the α polymorph of dicalcium silicate was evaluated by first-principles calculation based on density functional theory (DFT), as well as by molecular dynamics (MD) simulation with a polarizable ion model optimized by DFT calculation. The results indicated that the completely symmetric ionic arrangement, as reported for the α phase, is the most unstable. Electronic-state calculation and MD simulation indicated that a highly disordered ionic arrangement spontaneously forms in the α-phase crystal for structure relaxation when held at high temperatures, or when phosphorus is incorporated.  相似文献

In a placebo-controlled double-blind study, we examined the effects of dressing containing plant sterol (PS) on blood lipids and the safety in Japanese borderline or mildly hypercholesterolemic subjects. Fifty-nine subjects [total cholesterol (TC) concentration > or = 200 mg/dL] were randomly divided into two groups and were given daily 15 g of dressing containing 800 mg of PS [PS(+)-group] or without PS [PS(-)-group] for 12 weeks. Every 4 weeks, fasting blood was examined and subjective symptoms were analyzed. Serum TC, low-density lipoprotein cholesterol (LDL-C) and apolipoprotein B (ApoB) concentrations did not change in the PS(-)-group, while TC and ApoB significantly decreased in the PS(+)-group at 8 and 12 weeks and LDL-C at 4, 8 and 12 weeks. Moreover, serum TC, LDL-C and ApoB concentrations were significantly lower than those of PS(-)-group at 8 and 12 weeks. Other laboratory tests were all in normal ranges and no adverse events were observed. The results indicated that PS-containing dressing decreased serum TC, LDL-C and ApoB concentrations in borderline or mildly hypercholesterolemic subjects. It is therefore proved that the dressing containing PS is helpful in maintaining blood cholesterol level normal and hence, the health of Japanese.  相似文献

One of the most effective precipitation methods for submicrometer particles is the charged droplet scrubber. The dust collection in the charged droplet scrubber is considered to consist of some processes with respect to the interaction between charged dust particles and a charged water droplet. That is, the charged dust particles are attracted to, collide with, adhere to, penetrate into, rebound from, accumulate on, and reentrain from a charged water droplet. The direct observation of the collection process for dust particles by a charged water droplet was conducted by using the experimental setup designed by the authors. It was found from the experiments that particles with good wettability were captured inside the water droplet, particles with poor wettability floated at regular intervals on the droplet surface, and particles with medium wettability were trapped on or inside the water droplet. When particles were trapped on the droplet surface the growth of dendritic depositions was found, and their behavior was very distinct from those obtained on the fibrous filter due to the fluidity of the droplet surface. The deposition mechanisms of charged particles on the charged water droplet are discussed qualitatively.  相似文献

Stable dispersions of carbon black and nanodiamond in culture medium were prepared by adding a pre-mixed dispersion of commercial carbon black or nanodiamond in aqueous bovine serum albumin (BSA) solution to culture medium. Dynamic light scattering revealed that carbon black and nanodiamond dispersions prepared in both NaCl solution and culture medium were highly stable. From DLVO theory and the results of zeta potential measurements, the theoretical effect of the electrostatic interactions between adsorbed BSA molecules was found to be minimal. The asymmetric flow field-flow fractionation measurements revealed that 0.05 or 0.56 mg/mL of BSA molecules were adsorbed on 0.11 or 1.09 mg/mL of carbon black, respectively, indicating 1:2 complexation of BSA with the colloidal particles. In the case of nanodiamond, 0.06 or 0.60 mg/mL of BSA molecules were adsorbed on 0.10 or 1.05 mg/mL of nanodiamond, respectively, indicating 1:2 complexation of BSA, which is the same ratio as in the case of carbon black. The adsorbed BSA molecules served as an effective stabilizing agent for the carbon black and nanodiamond, ensuring dispersion stability for at least 1 week. The preparation of the dispersions can be easily carried out by other researchers for toxicity studies.  相似文献
