1. The depressor response to sodium nitroprusside (SNP) and the pressor response to Nomega-nitro-L-arginine methyl ester (L-NAME) were investigated in anaesthetized and ganglion-blocked 6 week old (young) and 20 week old (adult) spontaneously hypertensive rats (SHR), and the results were compared with those in age-matched normotensive Wistar-Kyoto (WKY) rats. 2. SNP produced a dose-dependent decrease of the mean blood pressure (BP) in both strains, and no differences in vascular sensitivity to SNP were observed between the strains. 3. L-NAME caused dose-dependent pressor responses in both strains. The sensitivity and the maximal response to L-NAME in SHR were significantly greater than those in age-matched WKY (P< 0.05 or 0.01; t-test, 13 d.f. in both ages). However, there were no significant differences in the responses between ages in each strain. 4. Acute reduction of BP induced by 7-O-ethylfangchinoline did not affect the responses to SNP and L-NAME in the adult SHR. 5. These results indicate that a greater amount of NO is tonically released in SHR and that its contribution to BP control is greater in SHR than in WKY, whereas vascular sensitivity to NO does not differ between the strains. In addition, acute changes in BP do not affect the degree of dependency on NO for BP control.  相似文献

Discharge stabilization, long-term operation, output power characteristics, and efficiency of the high-pressure CW CO2laser have been investigated under sealed conditions. A comparison is made with low-pressure CW CO2lasers. Two types of electrode structures suitable for operations in the pressure range 100-760 torr are presented. Effects of O2and CO on the discharge stability and unsaturated gain are described. By using molecular sieve3Aas an adsorbent of water vapor, which was the most detrimental impurity, sealed operation with no decrease in output power was achieved at 0.5-1.5 kW for more than 150 h elapsed time including about 30 h of discharge time. It has been demonstrated that high efficiency can be obtained in spite of high-pressure and sealed operation. The efficiency was improved by reducing the cavity loss due to the absorption of intracavity radiation by CO2molecules in the unexcited region, and by finding the optimum of gas mixture. A maximum efficiency of 19 percent was obtained at a 1 kW power level for a 100 torr gas mixture of either CO2-CO-N2-He = 2-1-19-19 or CO2-CO-N2-He-Ar = 2-1-18-10-10. The effects of Ar and N2proportion on the unsaturated gain and saturation parameter are discussed.  相似文献

Two chicken B cell lines, termed MuH1 and MuH4, which are resistant to ouabain, were established as fusion partners for production of chicken monoclonal antibody from the parental cell lines R27H1 and R27H4 which were deficient in thymidine kinase activity. The MuH1 synthesized, but did not secrete mu chain, whereas the MuH4 secreted non-specific IgM. Fusions of the MuH1 and MuH4 with spleen cells from chickens immunized with human IgG resulted in successful preparations of hybridomas secreting antibodies specific to human IgG. Transformed cells insensitive to HAT appeared in many culture wells after cell hybridizations with R27H4, but on hybridization with MuH1 or MuH4 the appearance of such unfavorable cells could be avoided by the inclusion of ouabain in HAT medium. Thus, in comparison with the R27H4 line, the MuH1 and MuH4 lines did not provide higher fusion efficiencies, but gave increased opportunities to obtain hybridomas producing specific antibodies. Among a total of eight hybridomas, four derived from HuM1 and four derived from MuH4, seven secreted both IgG and IgM, and the remaining one derived from MuH1 secreted only IgG. In all eight hybridoma lines IgG was specifically reactive to human IgG.  相似文献

Apparatus comprising a scanning tunneling microscopy (STM) and an atomic force microscopy (AFM) has been developed for use under supra-atmospheres. Observations of highly oriented pyrolytic graphite (HOPG) were carried out by STM and contact AFM operating in air and various gas atmospheres (hydrogen, helium, neon and argon) under pressures up to 1.1 MPa. Atomic resolution images of the HOPG were obtained by STM in all the gas atmospheres studied. However, it was found that the presence of water vapor gave rise to a noise current at increased pressures. Using contact AFM, the atomic resolution in an argon atmosphere decreased with increasing pressure, while atomic images were obtained under the other gas atmospheres at 1.1 MPa.  相似文献

The kinetics of the hydriding and dehydriding reaction of Mg2Ni and Mg-10 wt % Ni was investigated. The reaction rate at various hydride compositions was measured by the pressure change in a constant volume. In the case of Mg2Ni, at the beginning of the reaction the amount of absorbed/desorbed hydrogen can be expressed by n = k(p0peq) (nsn0), where n is the ratio of H to Mg in the solid, p0 and peq are the initial hydrogen pressure of the system and the equilibrium pressure, and n0 and ns are the values of n at the start of the reaction and a constant around 2, respectively. After a transient period of approximately 1 min, a diffusion-controlled stage took place as dn/dt = k′(p - peq)/t, where p is the pressure at time t. In the case of Mg-10 wt % Ni, throughout a run the absorption/desorption rate was expressed by dn/dt = k″ (p - peq)1.25.  相似文献

Genotyping of F-specific RNA phages is currently one of the most promising approaches to differentiate between human and animal fecal contamination in aquatic environments. In this study, a total of 18 river water and sediment samples were collected from the Tonegawa River basin, Japan, in order to describe the genogroup distribution of F-specific RNA and DNA phages using genogroup-specific real-time PCR assays. F-specific phages were detected in nine (100%) river water and six (67%) sediment samples. Eighty-five phage plaques were isolated from these samples and subjected to real-time PCR assays specific for the phages. F-specific RNA phages of human genogroups (II and III) were detected in 32 (38%) plaques, whereas those of animal genogroups (I and IV) were detected in 17 (20%) plaques. No correlation was observed between the genogroup distribution of F-specific RNA phages and the occurrence of human adenovirus genomes, suggesting that genotyping of the phages alone is inadequate for the evaluation of the occurrence of viruses in aquatic environments. SYBR Green-based real-time PCR assay revealed the presence of F-specific DNA phages in four (5%) plaques, which were further classified into two genogroups (fd- and f1-like phages) by sequence analysis. Thirty-two (38%) plaques were not classified as the F-specific phage genogroups, indicating the limited applicability of these real-time PCR assays to a wide range of aquatic environmental samples worldwide.  相似文献
