Alicyclobacillus species are thermo-acidophilic, endospore-forming bacteria that are able to survive pasteurisation and have been implicated in a number of spoilage incidents involving acidic foods and beverages. The aim of this study was to compare three isolation methods used for the detection of Alicyclobacillus acidoterrestris and to investigate the influence of incubation temperature on the growth of A. acidoterrestris and A. acidocaldarius. Peach juice samples inoculated with A. acidoterrestris K47 were analysed using either the International Federation of Fruit Juice Producers (IFU) Method No. 12 (Method A), which involved spread plating onto Bacillus acidoterrestris (BAT) agar at pH 4.0; Method B, which involved pour plating using potato dextrose agar (PDA) at pH 3.7; or Method C, which made use of membrane filtration followed by incubation on K agar at pH 3.7. The performance of the three methods differed significantly, with the IFU Method No. 12 recovering the highest percentage of cells at 75.97%, followed by Method B at 66.79% and Method C at 3.43%. These findings strengthen the proposal of the IFU for the use of the IFU Method No. 12 as a standard international method for the detection of Alicyclobacillus. To investigate the effect on growth of different incubation temperatures A. acidoterrestris (three strains) and A. acidocaldarius (two strains) were incubated at either 45 °C or 25 °C. Growth at 25 °C was slower and maximum cell concentrations were lower (1 × 105-106 cfu/mL compared to 1 × 107-108 cfu/mL) than at 45 °C for A. acidoterrestris. A. acidocaldarius was unable to grow at 25 °C and cell concentrations decreased by 1-2 logs. Since a growth temperature of 25 °C could not inhibit growth of A. acidoterrestris, cooling to room temperature (20°-25 °C) is not an effective control measure for A. acidoterrestris inhibition. 相似文献

BACKGROUND: The freshness of whiting was studied at five stages of ice storage by comparing the analysis of volatile compounds obtained through solid phase microextraction/gas chromatography/mass spectrometry (SPME/GC/MS) with two sensory methods. RESULTS: Of the volatile compounds identified, 38 were analysed using a statistical multivariate approach and classified according to their role in the estimation of freshness during storage as markers of freshness or spoilage. Regarding the evolution of the presence or absence of individual compounds, three categories were defined. For example, the volatile compounds propanal, hexanal, 1‐penten‐3‐ol, pentanal, 2,3‐pentanedione, 1‐penten‐3‐one, heptanal, (E)‐2‐pentenal, 2,3‐octanedione, (Z)‐2‐penten‐1‐ol, 1‐pentanol, butanal, octanal, 3,5,5‐trimethyl‐2‐hexene, 1‐hexanol and 4,4‐dimethyl‐1,3‐dioxane appeared highly relevant, because they are found throughout storage and can be divided into several categories that are directly related to the quality of fish. CONCLUSION: SPME/GC/MS combined with a statistical multivariate approach may be a useful method to identify volatile compounds and characterise fish freshness during storage. Scope: Products of cashew tree (Anacardium occidentale) are used in traditional medicine for various ailments, including diabetes. Methods and results: The anti‐diabetic properties of cashew plant parts were studied using differentiated C2C12 myoblasts (myotubes) and rat liver mitochondria. Hydroethanolic extract of cashew seed (CSE) and its active component, anacardic acid (AA), stimulated glucose transport into C2C12 myotubes in a concentration‐dependent manner. Extracts of other parts (leaves, bark and apple) of cashew plant were inactive. Significant synergistic effect on glucose uptake with insulin was noticed at 100 μg/mL CSE. CSE and AA caused activation of adenosine monophosphate‐activated protein kinase in C2C12 myotubes after 6 h of incubation. No significant effect was noticed on Akt and insulin receptor phosphorylation. Both CSE and AA exerted significant uncoupling of succinate‐stimulated respiration in rat liver mitochondria. Conclusion: Activation of adenosine monophosphate‐activated protein kinase by CSE and AA likely increases plasma membrane glucose transporters, resulting in elevated glucose uptake. In addition, the dysfunction of mitochondrial oxidative phosphorylation may enhance glycolysis and contribute to increased glucose uptake. These results collectively suggest that CSE may be a potential anti‐diabetic nutraceutical. 相似文献

Maloalcoholic fermentation (MAF) of grape must by Schizosaccharomyces pombe immobilised in calcium‐alginate double‐layer beads (ProMalic®) was studied in Erlenmeyer flasks and in a total recycle fixed‐bed reactor operating in batch mode. The reaction is pseudo‐first order with respect to l ‐malic acid and under similar conditions deacidification is faster in the recycle reactor. This was attributed to mass transfer limitations which were confirmed in the recycle reactor by studying the influence of yeast load on the rate of MAF. Mass transfer limitations are also responsible for the lower activation energy of fermentation with the immobilised yeast (67 ± 9 kJ mol?1) in comparison with the free cells (126 ± 19 kJ mol?1). Alcoholic fermentation and MAF were performed simultaneously, both in the recycle reactor and in the industrial trials, confirming the efficacy of immobilised S. pombe to reduce grape must acidity without interfering with the main fermentation. Altogether, the present results are useful for the scale‐up of a recycle reactor to process large volumes of grape must. 相似文献

The chemical composition, intake, digestibility, ultrastructure and microbial degradation of rice straw from Camargue were compared with barley straw. These variables were observed in two different herbivore digestive ecosystems: the sheep rumen and the donkey caecum. The two straws differed essentially in their ash content, which was three times higher in rice, owing to its silica content. Other chemical components were comparable, except a higher phenolic acids‐to‐lignin ratio in rice. Rice straw was better ingested than barley straw. Organic matter and neutral detergent fibre digestibilities were the same in both straws. Dry matter and cell wall disappearances could be adjusted to the exponential modelling equation with lag time, and differed between animals but not between straws. The sheep rumen had a higher extent of degradation, but the donkey caecum had a higher degradation rate. Statistical analysis revealed that cell‐wall components degradation was similar in the two straws except for ferulic acid, which was more degraded in rice straw. Formation of pesticide nonextractable (bound) residues in soil: magnitude, controlling factors and reversibility 总被引:1,自引:0,他引:1
The analysis of the coherent data on nonextractable (bound) residues (NER) from the literature and EU pesticide registration dossiers allows the identification of general trends, in spite of the large variability and heterogeneity of data. About 50% of the pesticides reviewed exhibit a low proportion of NER (less than 30% of the initial amount) while only 12% of pesticides have a proportion of NER exceeding 70%. The lowest proportion of NER was found for dinitroanilines (<20%), and the largest value was obtained for carbamates, and in particular dithiocarbamates. The presence of chemical reactive groups, such as aniline or phenol, tends to yield a larger proportion of NER. NER originating from N-heteroatomic ring were found to be lower than those from phenyl-ring structures. Among the environmental factors affecting the formation of NER, microbial activity has a direct and significant effect. Concerning the NER uptake or their bioavailability, consistent data suggest that only a small percentage of the total amounts of NER can be released. The analysis of NER formation kinetics showed that incubation experiments are often stopped too early to allow a correct evaluation of the NER maturation phase. Therefore, there is a need for longer term experiments to evaluate the tail of the NER formation kinetics. Still, the heterogeneity of the NER data between pesticides and for specific pesticides calls for great care in the interpretation of the data and their generalization. 相似文献

The present work aimed at evaluating the Cucurbita maxima Duchesne defatted seeds flour (CDSF) as fat replacer in beef patty. Pastes obtained from CDSF containing 60 and 72 % water were prepared and used to replace fat in beef patty at 25, 50, 75 and 100 % respectively. A control patty was processed using kidney fat from beef. The proximate composition, technological yield, textural and sensorial properties of different samples were evaluated. Results showed that CDSF contains 77.76/100 g proteins based on dried matter. Protein content of patty increased with fat substitution rates (FSR) from 18.82/100 g wet matter (WM) to 23.24/100 g WM respectively for the control (P0) and the sample in which fat was substituted at 100 % with the CDSF paste with a water content of 60 % (P460). The lipid content dropped with the increase in the fat substitution rate and water content of CDSF paste from 10.70/100 to 0.28/100 g WM respectively for the sample in which fat was substituted at 25 % with the CDSF paste with a water content of 60 % (P160) and the sample in which fat was substituted at 100 % with the CDSF paste containing 72 % water (P472). Similarly, hardness dropped from 54.78 N (P0) to 37.56 N (P472) respectively. The cooking yield increased significantly (p < 0.05) with the increase in the water content of the CDSF paste and the fat substitution rate. From the sensory analysis, patties with 75 and 100 % of FSR were the most accepted. Thus, the CDSF paste can be used in beef patty as a fat replacer to reduce the detrimental effect of animal fat consumption. 相似文献

Among natural biopolymers, polysaccharides and proteins are very promising for biodegradable and edible wraps with different characteristics, so that their formulations can be tailor‐made to suit the needs of a specific commodity. Films prepared from polysaccharides have good gas barrier properties but exhibit lower resistance to moisture compared to protein films (edible) or polylactide films (biodegradable). Protein‐based films show better mechanical and oxygen barrier properties compared to polysaccharide films. For that reason, film performances may be enhanced by producing blend systems, where hydrocolloids (mixtures of proteins and/or polysaccharides) form a continuous and more cohesive network. However, the lower water barrier properties of hydrocolloid films and their lower mechanical strength in comparison with synthetic polymers limit their applications in food packaging. Therefore, the enhancement of biopolymer film properties has been studied to attain appropriate applications. This review provides an extensive synthesis of the improvement of the properties of edible polysaccharide–protein films by way of various chemical, enzymatic, and physical methods. These methods primarily aim at improving the mechanical resistance. They also permit to ameliorate the water and gas barrier properties and related functional properties. 相似文献

Antimicrobial edible films based on sodium caseinate, glycerol, and 2 food preservatives (nisin or natamycin) were prepared by classical thermomechanical processes. Food preservatives were compounded (at 65 °C for 2.5 min) with sodium caseinate in a twin‐screw extruder. Anti‐Listeria activity assays revealed a partial inactivation of nisin following compounding. Thermoplastic pellets containing food preservatives were then used to manufacture films either by blown‐film extrusion process or by heat‐press. After 24 h of incubation on agar plates, the diameters of K. rhizophila growth inhibition zones around nisin‐incorporated films prepared by solution casting (control), extrusion blowing or heat pressing at 80 °C for 7 min of nisin‐containing pellets were 15.5 ± 0.9, 9.8 ± 0.2, and 8.6 ± 1.0 mm, respectively. Since heat‐pressing for 7 min at 80 °C of nisin‐incorporated pellets did not further inactivate nisin, this indicates that nisin inactivation during extrusion‐blowing was limited. Moreover, the lower diameter of the K. rhizophila growth inhibition zone around films prepared with nisin‐containing pellets compared to that observed around films directly prepared by solution casting confirms that nisin inactivation mainly occurred during the compounding step. Natamycin‐containing thermoplastic films inhibited Aspergillus niger growth; however, by contrast with nisin‐containing films, heat‐pressed films had higher inhibition zone diameters than blown films, therefore suggesting a partial inactivation of natamycin during extrusion‐blowing. 相似文献

European eel (Anguilla anguilla) populations are in decline. Glass eel recruitment has fallen 10-fold since the early 1980s. Estuaries play a fundamental role in the life history of eels because glass eels must pass through them to reach freshwater ecosystems. Unfortunately, because of their geographical position at the upstream basin slopes, estuaries accumulate metals like cadmium and are important sites of hypoxia events. In this context, we studied the effect of the oxygen level on the ventilation of the glass eel. In parallel, glass eels were submitted to different dissolved cadmium concentrations (0, 2, and 10 microg L(-1)) under two oxygen levels (normoxia PO2 = 21 kPa and Hypoxia PO2 = 6 kPa). The expression level of various genes involved in the mitochondrial respiratory chain, in the cellular response to metal and oxidative stresses, was investigated. Our results showed that hypoxia enhances (1) ventilation of the postlarval stage and (2) Cd accumulation in gills only at the lowest metal water concentration tested (2 microg Cd L(-1)). At the gene level, Cd exposure mimics the effect of hypoxia since we observed a decrease in expression of genes involved in the respiratory chain and in the defense against oxidative stress. 相似文献
