Ten-eleven translocation (TET) proteins, a family of Fe2+- and 2-oxoglutarate-dependent dioxygenases, are involved in DNA demethylation. They also help regulate various cellular functions. Three TET paralogs have been identified (TET1, TET2, and TET3) in humans. This study focuses on the evolution of mammalian TET genes. Distinct patterns in TET1 and TET2 vs. TET3 were revealed by codon-based tests of positive selection. Results indicate that TET1 and TET2 genes have experienced positive selection more frequently than TET3 gene, and that the majority of codon sites evolved under strong negative selection. These findings imply that the selective pressure on TET3 may have been relaxed in several lineages during the course of evolution. Our analysis of convergent amino acid substitutions also supports the different evolutionary dynamics among TET gene subfamily members. All of the five amino acid sites that are inferred to have evolved under positive selection in the catalytic domain of TET2 are localized at the protein’s outer surface. The adaptive changes of these positively selected amino acid sites could be associated with dynamic interactions between other TET-interacting proteins, and positive selection thus appears to shift the regulatory scheme of TET enzyme function.  相似文献

While the use of amorphous solid dispersions to improve aqueous solubility is well documented, little consideration has traditionally been given to the finished dosage form. The objective of this study was to evaluate the dissolution performance of amorphous solid dispersions containing a dispersed superdisintegrant with binding properties. KinetiSol® dispersing was used to thermally process hypromellose acetate succinate-based compositions containing the drug substance nifedipine (NIF) and a highly compressible grade of low-substituted hydroxypropyl cellulose (New Binder Disintegrants; NBD-grade). Solid-state analysis demonstrated that compositions were rendered amorphous during processing. Tablets containing intra-dispersion NBD were found to exhibit non-sink dissolution performance similar to milled intermediate, demonstrating excellent disintegration characteristics. Conversely, tablets without intra-dispersion NBD were found to release significantly less NIF during dissolution analysis due to particle agglomeration. It was determined that compressibility and particle wetting increased as the level of intra-dispersion NBD increased.  相似文献

To clarify the effect of hydrogen gas on fretting fatigue strength of the materials, which supposed to be used for hydrogen utilization machines, fretting fatigue tests were conducted in hydrogen gas. It is important to take fretting fatigue into account in strength design, because many fatigue failure accidents have occurred at joints or contact parts between components. As a part of the experiments, an austenitic stainless steel was focused in this paper. The material was SUS 304. Fretting fatigue strength in hydrogen gas decreased compared with that in air. Tangential force coefficient increased in the reverse order of fretting fatigue strength. Therefore, one of the reasons of the decrease of fretting fatigue strength was that tangential force was different depending on the environment. Absorption of hydrogen occurred during fretting in hydrogen gas was detected. The absorption could be considered as one of the causes of the decrease of fretting fatigue strength, since fretting fatigue life of pre-charged specimen was decreased and also the crack propagation threshold of short fatigue crack was reduced by hydrogen charge.  相似文献

Effect of water on vapor phase photografting on cellulose was investigated at 60°C. An activated grafting of methyl methacrylate by water contained in the sample was observed in the experiment. The effect of water was commonly recorded irrespective of the type of cellulose derivatives such as cellulose acetate (degree of substitution, DS = 0.18 and 0.33), cellulose nitrate (DS = 0.35 and 0.75), and carboxymethl cellulose (DS = 0.19 and 0.74). Organic solvents can also be used in place of water, indicating that the percent grafting decreases in the order, water > methanol > acetone > cyclohexane. From ESR studies, water in the sample was found to contribute to the decay of cellulose radicals rather than to the radical formation. The decay was accelerated by organic solvents, and the magnitude of the effect was in the order, water ≈ methanol > acetone > cyclohexane. Based on the above investigation, it was presumed that water contained in the sample cannot contribute directly to the formation of cellulose radicals which may initiate grafting, but mostly promotes the penetration of monomer into cellulose fibers. Such penetration could lead cellulose radicals to an effective initiation of grafting.  相似文献

Teeth are the hardest tissues covered with enamel produced by ameloblasts. The ameloblast differentiation is controlled by sequential epithelial-mesenchymal interactions during tooth morphogenesis. However, the molecular mechanism of ameloblast differentiation remains unclear. To address this question, we developed an in vitro assay system to evaluate the molecular mechanism of amelogenesis. First, we established dental epithelium-derived clones from 6-day-old rat incisors and established that cells of the clone SRE-G5 were the largest producers of amelogenin mRNA. Next, we analyzed the effects of several chemicals on the amelogenin expression in SRE-G5 cells. Only mitogen-activated protein kinase (MAPK) activators enhanced amelogenin mRNA expression. This finding corresponded to the immunohistochemical data showing the presence of phosphorylated forms of p38, c-Jun N-terminal kinase (JNK), and extracellular signal-regulated kinase (ERK) during ameloblast differentiation. To examine the roles of MAPK signals, we compared the effects of anisomycin and sodium salicylate on the expression of tooth-related differentiation markers. Both anisomycin and sodium salicylate induced amelogenin, Abcg2, and Bmp4 mRNA and down-regulated p75NGFR mRNA. On the other hand, ALP, ectodin, Bmp2 and Fgf8 mRNA were up-regulated only by anisomycin. These results indicate that MAPK signaling functions, at least in part, as the inducer of ameloblast differentiation.  相似文献

This paper presents a regression-based daily peak load forecasting method using multiple-year data with trend cancellation and trend estimation techniques. Daily peak load heavily depends on daytime temperature and is influenced by the other weather factors such as humidity. Since the characteristic of the load is varying, peak loads just before a forecasting day are more significant for the forecasting. The regression model can represent relationships between these weather factors and peak loads. However, the forecasting model is sometimes not adequate for precise load forecasting. The regression model is well matched with the late data, but the model causes large forecasting errors in transitional seasons because of seasonal change of load characteristics. In order to forecast precisely through a year, a method of using seasonal or whole year data from past years is proposed. In this paper, two kinds of trend data processing techniques are described. The first is trend cancellation. The second is trend estimation. The trend cancellation technique removes annual load growth by means of division or subtraction processes with morning load on the forecasting day. The trend estimation technique estimates the trend between the forecasting year's load and the past year's load by using the variable transformation techniques. We applied Pixon deconvolution as introduced in Part I to several practical, examples of low signal-to-noise ratio (SNR), electron energy-loss spectra with a goal toward restoring their fine spectral features and/or improving the energy resolution. We demonstrate that by directly fitting the two-dimensional spectral data recorded on the CCD; the method enables us to reveal fine spectral structures. Consequently, Pixon reconstruction extends the ability to probe electronic states in very spatially localized areas, a capability currently unique to our method.  相似文献

Laboratory and field tests were conducted to investigate the bearing and pullout capacities of steel piles with a continuous helix wing during cyclic loading. Both continuous helix and straight-sided piles were subjected to monotonic compressive, monotonic tensile, and cyclic reversal loading in the laboratory, while only the continuous helix pile was tested in the field. Both the laboratory and the field tests showed that the bearing and pullout capacities of the continuous helix pile under cyclic reversal loading decreased to approximately 60–80% of those of the pile under monotonic loading, with a larger reduction seen in the laboratory tests. The decrease in resistance was mainly due to the reduction in shaft friction, which was likely to be the result of soil disturbance and loosening around the pile with cyclic loading. The laboratory tests also showed that the tip resistance of the straight-sided pile under cyclic reversal loading was reduced, similarly due to the loosening of the soil, particularly underneath the pile tip. The tip resistance of the continuous helix pile, in contrast, did not degrade with cyclic loading, owing to the presence of the wing immediately above the pile tip that inhibited the loosening of the soil. As a means of diagnosing partial discharge (PD) signals propagate inside a Gas Insulated Switchgear (GIS), a study for the leakage of electromagnetic waves (EM-waves) emitted from the insulating spacer was implemented. The EM-waves leaking out from the solid insulator have the resonance frequencies depend on the spacing between adjacent bolts in the direction of the flange circumference, because the leakage portion is the equivalent of a slot antenna. In this paper, using an electromagnetic analysis model which has a simulated spacer on a concentrically-shaped GIS tank, the output characteristics of the EM-waves that leaked out from the slit were analyzed under various conditions such as the spacing between adjacent bolts, the width of the spacer, the dielectric constant of the spacer and the form of the flange. Also the actual measurement by the experimental equipment used to simulate the model was implemented for comparison with the analytical results. Consequently, the optimal specifications of the sensor and the measurement method used to achieve highly-sensitive detection for practical use were summarized and proposed as well as evaluating the effectiveness of the electromagnetic analysis model adopted in this paper.  相似文献
