The moderate consumption of red wine has been proposed to reduce the incidence of colorectal cancer. Its anti-cancer action my in part be mediated by the actions of polyphenols on colon cancer epithelial cells. We show that a red wine phenolic (50 μg/ml) extract devoid of anthocyanins (removed to reflect polyphenol bioavailability to the large intestine) and the major red wine flavonols quercetin, myricetin, laricitrin and syringetin (50 μM) are capable of inhibiting the proliferation of colorectal epithelial adenocarcinoma cells. This anti-proliferative activity was partly mediated by the direct cytotoxic actions of flavonols and also their ability to cause a significant cell cycle blockage in G2/M. The anti-proliferative effects induced by flavonols were preceded, in most cases, by a strong and significant reduction of cyclooxygenase-2 (COX-2) and cyclin D1 expression. These results indicate that the observed inverse correlation between colon cancer and a moderate red wine intake may be partly mediated by the actions of red wine flavonols on the growth of cancer cells. 相似文献

The aim of this study was to isolate and identify strains of Enterococcus from commercial cheeses and then analyze their abilities to produce biogenic amines. The genotypic variability, studied by
randomly amplified polymorphic DNA (RAPD)-PCR, showed a high degree of homogeneity among enterococcal strains. Afterward,
genotypic analysis indicated that all strains contain genes encoding a tyrosine decarboxylase. Our results indicate that a
potential health risk exists if the enterococcal strains are able to survive the pasteurization of milk or appear as post-pasteurization

Flavonoids and saponins from common beans have been widely studied due to their bioactivity. This research evaluated the effect of germination of black beans (Phaseolus vulgaris L.) on the antioxidant capacity and antiproliferative activity against cancer cell lines of saponins and flavonoids extracted from seed coats, cotyledons and sprouts. Principal component analysis was performed to achieve punctual associations between the black bean saponins and flavonoids concentrations to the antioxidant capacity and the antiproliferative activities. Total phenolic content and antioxidant capacity of extracts were higher when obtained from seed coats, mainly from the 3rd germination day. The extracts obtained from seed coats after 3 and 5 germination days inhibited all cancer cell lines proliferation with no cytotoxicity against control cells. Genistein was related with the activity against mammary cancer cells but flavonols and group B saponins were more related with hepatic and colon cancers. Yacon roots are a promising source of inulintype fructans (35 g/100 g dry matter) with a total amount of dietary fiber of about 45 g/100 g dry matter. The polydispersity of the particle size distribution of yacon aqueous suspension was reduced with increasing the degree of homogenization due to two phenomena: aggregation of particles, and disruption of large particles. Nonhomogenized yacon suspensions exhibited large cell clusters that were disrupted into small cell clusters, single cells, and aggregates. The most concentrated suspension exhibited mainly aggregates.The volume fraction of the suspensions decreased significantly with increasing the degree of homogenization. This was attributed to a denser packing of small particles and aggregates in between large particles thus allowing the water within the structure to be released.Yacon suspensions exhibited high elastic modulus (750 Pa) at low water insoluble solids content (0.9% WIS). Additional results suggested that inulintype fructans contributed to the elastic properties of yacon suspensions.The yacon suspensions studied in this work can be considered as semiconcentrated suspensions, i.e. the plot elastic modulus of the suspensions versus concentration exhibited a region immediately after the transition region. Particle interactions seem to be of great importance in this region in the particular case of yacon suspension since the elastic modulus (G′) reached 750 Pa at low water insoluble solids content (< 1%). 相似文献

BACKGROUND: The increasing consumption of produce all over the world has resulted in increasing concern by the regulatory agencies with respect to the level of safety performed by the processors. The objective of the present study was to investigate the hazards involved in the various steps of fresh‐cut lettuce processing (reception/selection of raw material, washing, rinsing, sanitisation and final product) by means of microbiological analyses of microbial groups used as indicators of hygienic conditions and of pathogens. RESULTS: High microbial loads of mesophilic and psychrotrophic bacteria and Pseudomonas spp. were found in the ram reception (~6 log colony‐forming units (CFU) g?1), which were reduced by a single logarithmic cycle for the last two microbial groups after the sanitisation step (P < 0.05), the latter being ineffective against the first microbial group (P > 0.05). Lower counts of yeasts and moulds, total coliforms (35 °C) and faecal coliforms (44 °C) were observed in the initial step (3.49–4.53 log CFU g?1, 0.65–1.55 log most probable number (MPN) g?1 and 0.50–0.90 log MPN g?1 respectively), these values increasing significantly after the sanitisation step for yeasts and moulds (~5 log CFU g?1) but remaining unaltered for coliforms (P > 0.05). Salmonella spp. were not found in any of the experiments carried out, while the presence of Escherichia coli was observed in the final product. CONCLUSIONS : Practices compromising the hygienic quality of the final product during commercial storage were observed and corrective measures suggested. Excessive nerve growth factor (NGF) production by the ovary, achieved via a transgenic approach, results in arrested antral follicle growth, reduced ovulatory capacity, and a predisposition to cyst formation in response to mildly elevated LH levels. Two salient features in these mutant mice (termed 17NF) are an elevated production of 17α-hydroxyprogesterone (17-OHP(4)), testosterone, and estradiol (E(2)) in response to gonadotropins, and an increased frequency of granulosa cell (GC) apoptosis. In this study, we show that the increase in steroidal response is associated with enhanced expression of Cyp17a1, Hsd17b, and Cyp19a1, which encode the enzymes catalyzing the synthesis of 17-OHP(4), testosterone, and E(2) respectively. Using a proteomic approach, we identified stathmin (STMN1), as a protein that is overproduced in 17NF ovaries. In its phosphorylated state, STMN1 mediates a cell death signal initiated by tumor necrosis factor α (TNF). STMN1 is expressed in GCs and excessive NGF increases its abundance as well as that of its forms phosphorylated at serine (Ser) 16, 25, and 38. TNF synthesis is also increased in 17NF ovaries, and this change is abolished by blocking neurotrophic tyrosine kinase receptors. Inhibiting TNF actions in vivo by administering a soluble TNF receptor prevented the increase in total and phosphorylated STMN1 production, as well as GC apoptosis in NGF-overproducing ovaries. These results indicate that an excess of NGF in the ovary promotes steroidogenesis by enhancing the expression of enzyme genes involved in 17-OHP(4), testosterone, and E(2) synthesis, and causes GC apoptosis by activating a TNF/ STMN1-mediated cell death pathway. 相似文献

There is a great variability in cooking time and final texture among cassava cultivars and one factor in determining these characteristics is the age of roots at harvest. Final texture after cooking is an important attribute for the acceptability of cassava and starch gelatinization has a role in softening the tissue. Two cassava cultivars at two different harvest maturities were cooked at 90 °C and at boiling water temperature (98 °C) and the extent of gelatinization was determined using an iodine colorimetric method. A generalized model for starch gelatinization, considering temperature and cooking time as continuous variables and age at harvest as qualitative variable, was used to model the cooking process and applied for each cultivar. At 90 °C IAPAR-19 Pioneira reached maximum gelatinization in 32 min for the 12 month old sample and 88 min for the 25 month old sample, while double the time was needed for Catarina Amarela. At 98 °C Pioneira gelatinized in 12 min for the 12 month and 32 min for the 25 month old sample while Catarina Amarela needed 28 and 56 min. Calculated activation energy for starch gelatinization was 122.3±5.2 and 98.6±4.7 kJ/mol for cultivar IAPAR-19 Pioneira and Catarina Amarela, respectively. 相似文献

A sensitive electrochemical acetylcholinesterase (AChE) biosensor was successfully developed on polyaniline (PANI) and multi-walled carbon nanotubes (MWCNTs) core-shell modified glassy carbon electrode (GC), and used to detect carbamate pesticides in fruit and vegetables (apple, broccoli and cabbage). The pesticide biosensors were applied in the detection of carbaryl and methomyl pesticides in food samples using chronoamperometry (CA). The GC/MWCNT/PANI/AChE biosensor exhibited detection limits of 1.4 and 0.95μmolL(-1), respectively, for carbaryl and methomyl. These detection limits were below the allowable concentrations set by Brazilian regulation standards for the samples in which these pesticides were analysed. Reproducibility and repeatability values of 2.6% and 3.2%, respectively, were obtained in the conventional procedure. The proposed biosensor was successfully applied in the determination of carbamate pesticides in cabbage, broccoli and apple samples without any spiking procedure. The obtained results were in full agreement with those from the HPLC procedure. 相似文献
