Contractile response, membrane activity, and protein kinase A (PKA) activity were measured on the longitudinal muscle taken from the estrogen-treated rat uterus, and the influence of Mn ion on the inhibitory effects caused by db cAMP and forskolin was investigated. Phasic contractions generated in the muscle taken from the middle portion of uterus were depressed to 48 and 83% by 30 microM db cAMP and 0.1 microM forskolin in Mg-free Krebs solution, respectively; phasic contractions were more strongly depressed by the agents in the solution containing 0.2 mM Mn. Action potentials consisted of spike and plateau components, and the duration of the plateau potential was reduced by the application of the agents; membrane activity was more strongly depressed in the presence of 0.2 mM Mn. The contractile depression caused by db cAMP was reduced and by forskolin was enhanced by pretreatment of the tissue with 0.6 mM Mn for 30 min. The PKA activity was increased by 39 and 6% of the control, when 30 microM db cAMP and 0.1 microM forskolin were applied, respectively; the PKA activity in response to db cAMP and forskolin was reduced and enhanced, respectively, when the tissues were pretreated with 0.6 mM Mn. It was proposed that Mn ions permeated into cell interior when the muscle was exposed to 0.6 mM Mn, so that the effects of the agents were differently affected. It was also shown that plateau potential dominated in the muscle taken from the ovarian portion, and the contractile inhibition caused by the agents was far weaker.  相似文献

Nanostructured nickel oxide samples having different average particle sizes are synthesized through a wet chemical route. Room temperature magnetic hysteresis of the samples are recorded using a vibrating sample magnetometer. The magnetic properties of the samples are found to be markedly different from those of single crystalline nickel oxide. The sample with an average particle size of 2-3 nm showed superparamagnetism with magnetization curves defined by the Langevin function. Anomalously large uncompensated magnetic moment associated with this sample is attributed to the multisublattice magnetic structure. Interestingly, samples with larger average particle sizes of 13 and 18 nm exhibited superantiferromagnetism with the magnetization curves varying linearly with applied field and susceptibility values larger than that of bulk nickel oxide. The results highlight the importance of surface atoms and surface driven spin rearrangements in determining the magnetic properties of nanostructured nickel oxide.  相似文献

The change in the microstructure on aging at 12 K below a low-temperature structural phase transition in La1.5Sr0.1Nd0.4CuO4 has been examined by transmission electron microscopy. From a seriesof 100 forbidden dark field images obtained inin situ observation, spotty-shape regions with a Pccn/LTT tilt of the CuO6 octahedron were found to be nucleated and grow inside a LTO domain, accompanying the annihilation of some spotty-shape regions. In addition, there exists a tweed texture in fundamental dark field images, which is due to the macroscopic strain involved in the transition. Because a mapof the tweed texture is different from that of the spotty-shape region, the tilt of the octahedron is not always coupled to a proper spontaneous strain.  相似文献

Normal and brown midrib mutant (bmr) maize (Zea mays L) were examined for variations in their morphological composition. The degradability of the leaf blade, leaf sheath and stem, proportional area of specific tissues in leaf blade, and the ultraviolet (UV) absorption spectra of cell walls were measured and related to variations in cell wall degradability by rumen microorganisms. The UV and infrared (IR) absorption spectra of the lignins isolated from leaf blades of both types, before and after reduction with sodium borohydride, were recorded. The bmr3 maize had higher dry matter (DM) and neutral detergent fibre (NDF) degradabilities for leaf blade, leaf sheath, and stem than the normal counterpart. Approximately 35% and 26% of the observed difference in DM degradability was attributed to the difference in DM degradability of stem and leaf blade, respectively, and 39% to the difference in DM composition of stem. Distinct differences in tissue degradation of the leaf blades were observed for mesophyll cell walls in the midrib portion, which were thinner and of greater number in the bmr3 maize. Sclerenchyma cells were present only in the vascular bundles in the bmr3 leaf blade, while in the normal type those cells were underneath the epidermis tissue. The bmr3 plant also had large epidermal cells. UV microspectrometry of mesophyll cell walls of the bmr3 maize showed their lower UV absorbance around 320 nm compared to that of the normal, but not at 280 nm. Considerable increase in the UV absorbance at 280 nm was observed for the isolated lignins after reduction, suggesting a lesser degree of lignification in the bmr3 maize tissues. Lowered UV absorbance of the isolated lignin around 320 nm after reduction was associated with the removal of the IR bands at 1730, 1660, 1600, and 1250 cm?1.  相似文献
