Human ferritin H-chain protein (FTH1)-based nanoparticles possess a precisely assembled nanometer-scale structure and high safety. However, their applications for imaging and drug delivery towards cancer cells remain limited due to a lack of target specificity. Epidermal growth factor receptor (EGFR) is overexpressed in many malignant tissues including breast cancer, and has been used as a therapeutic target for cancer treatment. Herein, a genetic method is shown to generate EGF-FTH1 chimeric proteins. EGF-FTH1 nanoparticles with EGF on the surface are then produced. The data demonstrate that EGF-FTH1 nanoparticles, with a small size (11.8 ± 1.8 nm), narrow size distribution, and high biosafety, can specifically bind to and then be taken up by breast cancer MCF-7 cells and MDA-MB-231 cells, but not normal breast epithelial MCF-10A cells. In contrast, binding and absorption of nontargeted ferritin-based nanoparticles to breast cancer cells are negligible. In vivo studies show that EGF-FTH1 nanoparticles are accumulated in breast tumors in a mouse xenograft model. Interestingly, the concentration of EGF-FTH1 nanoparticles in the tumor site is significantly reduced when mice are pretreated with an excess of free EGF. These results imply that EGF-EGFR interaction plays an important role in regulating the tumor retention of EGF-FTH1 nanoparticles.  相似文献

Administration of 4,4′-diethylamino-ethoxyhexestrol increased total phospholipids and free cholesterol in rat liver. An elevation of a peculiar glycerophospholipid, lysobisphosphatidic acid, was marked both in liver and spleen. An increase in phosphatidylinositol was also noticed in spleen. However these changes in rats were not so marked as in human cases. There was an accumulation of a substance in rat tissue which was identified as a derivative of the drug, while the intact drug accumulated in large amount in human cases. Differences in the effect of 4,4′-diethylaminoethoxyhexestrol on lipid metabolism between human and rat seem to be related to the difference in drug digesting ability between these two animal species.  相似文献

The goal of the present study was to evaluate techniques for modeling the physiological responses, rectal temperature, and respiratory rate of black and white Holstein dairy cows. Data from the literature (792 data points) and obtained experimentally (5884 data points) were used to fit and validate the models. Each datum included dry bulb air temperature, relative humidity, rectal temperature, and respiratory rate. Two models based on artificial intelligence—artificial neural networks and neurofuzzy networks—and one based on regression were evaluated for each response variable. The adjusted models predict rectal temperature and respiratory rate as a function of dry-bulb air temperature and relative humidity. These models were compared using statistical indices. The model based on artificial neural networks showed the best performance, followed by the models based on neurofuzzy networks and regression; the last two performed similarly.  相似文献

A genetically engineered porcine myoglobin triple mutant (H64V/V68H/H93A) (VHA-Mb) contains 6 non-axial His residues (His24, His36, His48, His81, His82, and His119) besides two candidate axial His residues (His68 and His97). Although previous resonance Raman study on the ferric VHA-Mb were not conclusive for its coordination structure, present EPR parameters of the ferric VHA-Mb were consistent with bis-imidazole coordination of His68/His97. We further investigated the reactivity of these possible His ligands with diethylpyrocarbonate (DEPC) to clarify the coordination structure and their protonation states in ferric form. We found that the non-axial His residues were easily modified with a low concentration of DEPC based on UV spectral changes and MALDI-TOF-MS analyses. On the other hand, the two candidate axial His ligands were protected from the modification due to a limited steric exposure of their imidazoles to solvent, the Fe(3+)-N(epsilon2) coordination bond, and the protonation of N(delta1) by forming a hydrogen bond with their immediate surroundings. However, once N-carbethoxylation occurred at N(epsilon2) of His97, resulting in a disruption of the heme Fe(3+)-N(epsilon2) coordination bond, it facilitated the second N-carbethoxylation to take place at N(delta1) of the same imidazole ring, leading to a bis-N-carbethoxylated derivative and further to a ring-opened derivative. These phenomena were consistent with the bis-His68/His97 coordination. Further, these were not observed at all for cytochrome b(561), a transmembrane di-heme containing protein responsible for the ascorbate-specific transmembrane electron transfer, where only a specific N(delta1)-carbethoxylation of axial His occurred at a low concentration of DEPC, leading to an inhibition of the electron acceptance from ascorbate without a release of the heme. These distinct results might be related to a specific physiological mechanism being operative at the cytosolic heme center of cytochrome b(561).  相似文献

This paper discusses the two public water supply systems in Japan, for domestic and industrial use. Looking first at domestic supplies, it is found that more than 92% of the population benefited from public water services in 1982, the great majority relying on municipal systems. The proportion of demand met from surface water had reached 68% by 1982, and was set to continue rising, partly for reasons of groundwater conservation. Water quality must meet extensive legal standards. Industrial water requirements are met by a variety of systems, including municipal waterworks, recycled water and private supplies; public industrial waterworks play a relatively small part. The paper concludes by outlining the major problems of water supply that Japan must address.  相似文献

Hep27 monoclonal antibody (Hep27 Mab) was raised by immunizing BALB/c mice with cells of the Thai human hepatocellular carcinoma (HCC) cell line HCC-S102 using hybridoma technology. The Hep27 Mab recognizes oncofetal development antigens by reacting with many HCC, other cancers, fetal and newborn liver but not adult liver. The Hep27 Mab alone markedly inhibits the growth of hepatocellular carcinoma cell lines (65% viability on the third day), suggesting its clinical usefulness. Moreover, complementary DNA (cDNA) for active variable regions of both heavy and light chains of the antibody has been cloned. Sequence analysis of the variable region of the Hep27 Mab revealed that the V(H) and V(L) genes belong to the V(H) 7183 and V(K) families, respectively. We have also characterized the reactivity of the Hep27 Mab to synthetic carbohydrate epitopes and 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP)-treated HCC-S102 cells. The results showed that the Hep27 Mab recognizes a neoglycolipid containing a mucin core unit and PDMP treatment reduced Hep27 Mab binding activity to HCC-S102 cells, indicating that the Hep27 Mab recognizes a glycolipid antigen on HCC-S102 cells. This Mab may be potentially useful for studying antigenic expression in hepatocellular carcinoma and as a targeting agent for radioimmunodetection and immunoconjugated therapy.  相似文献
