ABSTRACTFor the first time, we have fabricated insect-derived cell sheets by using temperature-responsive culture surfaces having a phase-transition temperature below 25°C. We prepared the temperature-responsive cell culture surfaces (tissue culture polystyrene, TCPS) by grafting a copolymeric gel consisting of hydrophobic N-tert-butylacrylamide (tBAAm) and N-isopropylacrylamide (IPAAm) units. First, to characterize the hydrophilic and hydrophobic properties of the copolymeric gel-grafted surfaces, static water contact angles of each surface were measured at various temperatures. By increasing the amount of tBAAm in the grafted copolymeric gel, the transition temperature of the gel was shifted to lower temperatures. At 25°C, the grafted copolymeric gel was dehydrated, and the insect-derived cells (AeAl2 cells) adhered on all the copolymeric gel-grafted surfaces. At 20°C, AeAl2 cells cannot adhere on the P(IPAAm-1.62tBAAm)-TCPS surface (the initial molar ratio of IPAAm and tBAAm (tBAAm?=?1.62 mol%)) better than on other surfaces (TCPS and tBAAm?=?4.88, 8.17 mol%). These two findings implied that the lower critical solution temperature of the copolymeric gel-grafted-TCPS existed from 20°C to 25°C. The laminin-coated P(IPAAm-1.62tBAAm)-TCPS surface showed temperature-dependent cell attachment and detachment properties, while AeAl2 cells were not detached from the extracellular matrix uncoated P(IPAAm-1.62tBAAm)-TCPS surface. Food poisoning from Staphylococcus aureus is sometimes caused by improper handling of food items in food preparation facilities. Prevention of contamination by employees is particularly important in facilities where a significant amount of food preparation is performed by hand. Some experiments have been performed to describe bacterial cross-contamination in the food preparation process, but there have been few studies of cross-contamination in actual food preparation facilities. Aiming to shed light on the transmission of S. aureus in food preparation facilities, this study collected samples of 66 strains of this bacterium from the fingers of food preparation staff, foodstuffs, prepared foods, cooking utensils, and cooking equipment and typed them with the ribotyping method. S. aureus from the same ribogroup was detected on the hands of a study participant, a faucet, knife, frying pan, and a salad, indicating that bacteria found on the hands of the study participant was transmitted to cooking utensils and prepared foods. The hair lipid composition collected from 44 Japanese females between 1 and 81 years of age was examined for eight lipids including hydrocarbons (HCs), squalene (SQ), wax esters (WEs), triglycerides (TGs), fatty acids (FAs), cholesterol (CH), ceramides (CERs), and 18-methyl eicosanoic acid (MEA). In this study, the 5-cm length from the proximal root end of hair fibers, which had never been exposed to any chemical treatment, was used after 5-min incubation with hexane following shampooing. Hair lipids were extracted with solvent and subsequent alkali-solvent and were then analyzed by a combination of chromatography. Although the average contents of the lipids showed great fluctuations among individuals, there were significant correlations between the levels of each lipid, which allowed for the classification of the hair lipids into four groups: group A: SQ, WEs, TGs, and FAs (designated as endogenous lipids based upon their sebum origin); group B: CH and CERs (designated as endogenous lipids); group C: HC (unknown origin); and group D: MEA (the other endogenous lipid). A principal component analysis for eight lipids revealed that the hair lipid composition was characterized by a predominant negative correlation between each lipid for groups A and B. This negative correlation suggests that the endogenous lipids in group B serve as a barrier against the penetration of predominantly sebum-derived exogenous lipids (group A). Endogenous lipids consisting of CH and CERs (group B) and MEA (group D) should be designated as intrinsic internal lipids of human hair. 相似文献

A two-dimensional microarray of ten thousand (100 x 100) hepatocyte heterospheroids, underlaid with endothelial cells, was successfully constructed with 100 microm spacing in an active area of 20 x 20 mm on microfabricated glass substrates that were coated with poly(ethylene glycol) brushes. Cocultivation of hepatocytes with endothelial cells was essential to stabilize hepatocyte viability and liver-specific functions, allowing us to obtain hepatocyte spheroids with a diameter of 100 microm, functioning as a miniaturized liver to secret albumin for at least one month. The most important feature of this study is that these substrates are defined to provide an unprecedented control of substrate properties for modulating cell behavior, employing both surface engineering and synthetic polymer chemistry. The spheroid array constructed here is highly useful as a platform of tissue and cell-based biosensors and detects a wide variety of clinically, pharmacologically, and toxicologically active compounds through a cellular physiological response. 相似文献

Polymer-based nanohybrid materials were created using sustainable sepiolite clay composed from ubiquitous elements. Although sepiolite is generally recognized as a fibrous natural clay mineral, it turned out to be an acicular microcrystal because of the organo-modification of the outermost surface. Surface modification was performed using phosphonic acid derivatives containing hydrocarbon chains or fluorocarbon chains. Formation of a bidentate bond enhanced the desorption temperature and made nanocomposite preparation possible by melt compounding with polymers having a high melting point. As a result of organo-modification, amphiphilic sepiolite was obtained, and nanodispersion in an organic solvent was achieved. This technology was useful for detailed evaluation of sepiolite morphology. The nanocomposite of crystalline polymers/organo-modified sepiolites achieved uniform dispersion of these nanofillers in the matrix polymer. The introduction of 1 wt% nanofillers did not impair the transparency of the matrix polymer. As a result, a lamellae structure of the polymer developed, the crystallinity increased, and the mechanical properties improved. In addition, the crystallization temperature was improved, indicating that organo-modified sepiolites may act as a nucleating agent. It was found that sepiolite nanofiller with a highly aggregated tendency can achieve a well-nanodispersed state, even in phase-separable fluoropolymers, by applying fluorocarbon modification. POLYM. The shrinking mechanism of comb-type grafted poly(N-isopropylacrylamide) gel was investigated by small-angle X-ray scattering (SAXS). The SAXS reveals that the microdomain structure with characteristic spacing of 460 Å is developed in the comb-type grafted poly(N-isopropylacrylamide) gel during the shrinking process. These observations suggest that the freely mobile characteristics of the grafted chains are expected to show the rapid dehydration to make tightly packed globules with temperature, followed by the subsequent hydrophobic intermolecular aggregation of the dehydrated graft chains. The dehydrated grafted chains created the hydrophobic cores, which enhance the hydrophobic aggregation of the networks. These aggregations of the NIPA chains contribute to an increase in void volume, which allow the gel having a pathway of water molecules by the phase separation. 相似文献
