Effects of lipase,lipoxygenase, peroxidase and free fatty acids on volatile compound found in boiled buckwheat noodles 总被引:1,自引:0,他引:1

BACKGROUND: Relationships between buckwheat (Fagopyrum esculentum Moench) flour lipase, lipoxygenase and peroxidase activity, along with levels of individual free fatty acids (FFAs) and levels of headspace volatile compounds of boiled buckwheat noodles, were investigated for 12 different buckwheat varieties. Enzyme activities and FFA levels in flour were correlated with their respective varietal arrays of boiled noodle headspace volatile compounds, measured by gas chromatography–mass spectrometry. RESULTS: The volatiles hexanal, tentative butanal, tentative 3‐methylbutanal and tentative 2‐methylbutanal showed significant positive correlation with one another, indicating that they may be generated through similar mechanisms. These important volatile components of buckwheat flavor were also positively correlated with lipase and/or peroxidase activity, indicating that enzymatic reactions are important in flavor generation in boiled buckwheat noodles. On the other hand, pentanal, which showed no significant correlation with any enzyme activity, showed a significant positive correlation to the levels of C18:2 and C18:3 FFAs, suggesting the existence of a ‘non‐enzymatic’ and/or ‘uncertain enzymatic pathway’ for flavor generation in boiled buckwheat noodles. CONCLUSION: Lipase and peroxidase in buckwheat flour are important for flavor generation of boiled buckwheat noodles. The cellulose binding domain (CBD) of cellulosome-integrating protein A from Clostridium thermocellum NBRC 103400 was genetically fused to FMN-dependent NADH-azoreductase (AZR) and glucose 1-dehydrogenase (GDH) from Bacillus subtilis. The fusion enzymes, AZR-CBD and CBD-GDH, were expressed in Escherichia coli Rosetta-gami B (DE3). The enzymes were purified from cell-free extracts, and the specific activity of AZR-CBD was 15.1 U/mg and that of CBD-GDH was 22.6 U/mg. AZR-CBD and CBD-GDH bound strongly to 0.5 % swollen cellulose at approximately 95 and 98 % of the initial protein amounts, respectively. After immobilization onto the swollen cellulose, AZR-CBD and CBD-GDH retained their catalytic activity. Both enzymes bound weakly to 0.5 % microcrystalline cellulose, but the addition of a high concentration of microcrystalline cellulose (10 %) improved the binding rate of both enzymes. A reactor for flow injection analysis was filled with microcrystalline cellulose-immobilized AZR-CBD and CBD-GDH. This flow injection analysis system was successfully applied for the determination of glucose, and a linear calibration curve was observed in the range of approximately 0.16–2.5 mM glucose, with a correlation coefficient, r, of 0.998. 相似文献

The sintering behavior of Pt metal particles was studied by supporting them on silica-coated alumina. Silica coating was found to be effective for the retention of a large surface area of alumina even after calcination at elevated temperatures. Before sintering, the size of Pt metal particles on all the silica-coated aluminas, including the uncoated alumina, was identical, while the particle size was larger on silica than on alumina. After sintering the Pt catalyst at 1073 K, the particle size increased on uncoated alumina as well as on alumina coated with thicker silica layers, especially on the supports previously calcined at >1473 K. On the other hand, the size of Pt metal particles did not increase much on alumina coated with monolayer silica. The observed suppression of sintering of Pt metal particles resulted from the retention of a large surface area of alumina with a thinner silica layer. In the case of a thicker silica layer, although a large surface area was maintained after calcination at elevated temperatures, the existence of a bulk silica-like property of the support did not favor the suppression of sintering of Pt metal particles. 相似文献

Pitting corrosion is typical corrosion observed on coated hold frames in way of cargo holds of bulk carriers, which exclusively carry coal and iron ore. Extensive survey on the effect of pitting corrosion on structural strength under a wide variety of loading conditions is necessary to clarify the relationship between pitting intensity and residual strength in detail. In the present study, a series of tests has been conducted on structural models which consist of web, shell and face plates to investigate the effect of pitting corrosion on strength of web plates subjected to patch loading. In these tests, artificial pitting was made on the web plates and two equal concentrated loads have been applied vertically at the one third points of simply supported models. It was found that web crippling behavior is strongly affected by the pit distribution on the web plates. According to the FE-analyses following the experiment, ultimate strength of the web plates with pitting under patch loading is a little smaller than or almost the same as that of the web plates with uniform corrosion in terms of average thickness loss. 相似文献

In this study, we attempted to classify the methylotrophic yeasts based on diversities of alcohol oxidase (AOD), i.e. zymogram patterns and partial amino acid sequences. According to zymogram patterns for AOD, members of the methylotrophic yeasts separate into two major lineages, one group involving strains having a single AOD and the other group, including Pichia methanolica, Candida pignaliae and C. sonorensis, showing nine AOD isozymes. Based on partial amino acid sequences of AOD, the methylotrophic yeasts could be divided into five groups, and this classification agrees mostly with grouping based on 26S domain D1/D2 rDNA nucleotide sequences, except for some strains. Moreover, the strains having AOD isozymes constitute one group with P. trehalophila, P. glucozyma and Pichia sp. strain BZ159, although these strains are divided into two types, based on amino acid sequences of second AODs. On the other hand, these AOD isozymes consist of two subunits; the first subunits are induced not only by methanol but also by glycerol and pectin, although the second subunits are mainly induced by methanol. These data indicate that AOD isozymes and second AOD genes distribute widely in several methylotrophic yeasts in the natural environment, and second AOD genes may have evolved as methylotrophic genes that can adapt to the environmental conditions of higher methanol concentrations. 相似文献

An overview is given of previously-proposed measurement-based connection admission control (CAC) methods. First we address requirements for CAC methods, and then provide a taxonomy for CAC methods. Measurement-based CAC methods are discussed in detail, classified according to the taxonomy, and compared against each other with respect to the requirements. We conclude that measurement-based CAC methods based on effective bandwidth and bufferless models are promising because they do not require complex hardware and are less dependent on assumptions regarding traffic than methods in which the effect of buffer is considered. 相似文献

A microbial growth model was presented for estimating the dynamic behavior of cell growth and substrate consumption in the biodegradation of phenol containing a heavy metal, such as zinc or copper ions. The application of the model for experiments in a batch culture and a continuous culture was examined. The values calculated according to the model corresponded satisfactorily with experimental data, such as the optical density of cells and the concentration of phenol. Using the model, the stability of steady states in a continuous culture was analytically evaluated based on the eigenvalues. The steady states were separated into three categories: (i) a stable steady state where phenol was consumed, (ii) an unstable steady state where phenol was consumed, and (iii) a washout state where phenol was not consumed. The number, kind, and stability of steady states varied significantly with operational conditions such as dilution rate, feed concentration of phenol, and feed concentration of heavy metal ions. To test the effects of the annual fluctuation of the environmental factors on the starch properties in potato tuber development, starches were isolated from potato cultivars grown for ten consecutive years (2001–2010) on the experimental farm in Memuro, Hokkaido, the northernmost island of Japan. The median granule size, phosphorus content, and rapid visco‐analyzer pasting properties, peak viscosity, breakdown, and pasting temperature of these starches were analyzed. Daily temperature and precipitation records in Memuro were obtained from the Japan Meteorological Agency. Moreover, the correlation coefficients of yearly accumulated temperature and precipitation from flowering to harvesting with all the starch quality parameters were calculated. No relationships of accumulated temperature with median granule size, peak viscosity, and breakdown were found. The low phosphorus content was slightly associated with the high accumulated temperature. The pasting temperature was significantly and positively correlated with the accumulated temperature, suggesting that the pasting temperature strongly responds to the environmental temperature during potato tuber development. In contrast, all the starch quality parameters were almost irrespective of the precipitation during potato tuber development. 相似文献

The concept of context awareness is believed to be a key enabler for the new ubiquitous network service paradigm brought by cloud computing platforms and smartphone OSs. In particular, autonomous context-based service customization is becoming an essential tool in this context because users cannot be expected to pick step by step the appropriate network services by manually and explicitly matching preferences for their current context. In this work, we hence focus on the core problem of how to detect changes of context for network services. In turn, detection of such changes can trigger timely system reconfigurations. We introduce a trigger detection mechanism based on a mixed graph-based representation model able to encode geographical and social relationships among people and social objects like stores, restaurants, and event spots. Our mechanism generates a trigger when a significant change in the graph takes place, and it is able to render significant changes in a geographical relationship that holds among objects socially connected with each other. The main benefits of our method are that (1) it does not require building reference models in advance, and (2) it can deal with different kinds of social objects uniformly once the graph is defined. A computer simulation scenario provides evidence on the expected performance of our method. 相似文献
