Changes in volatile compounds were monitored in whiting (Merlangius merlangus), cod (Gadus morhua) and mackerel (Scomber scombrus) and related to spoilage. Data are presented from headspace/mass spectrometric (HS/MS) analysis and solid‐phase microextraction/gas chromatographic/mass spectrometric (SPME/GC/MS) analysis at two time points (day 0 and day 10) during storage at 4 °C. HS/MS revealed 24 ions that could be used as markers of spoilage. SPME/GC/MS identified 86 compounds, 20 of which could perhaps be used to characterize freshness: principally alcohols, ketones, aldehydes and C2–C11 esters. In the male reproductive organs of mammals, the formation of spermatozoa takes place during two successive phases: differentiation (in the testis) and maturation (in the epididymis). The first phase, spermiogenesis, relies on a unique adherens junction, the apical ectoplasmic specialization linking the epithelial Sertoli cells to immature differentiating spermatids. Vezatin is a transmembrane protein associated with adherens junctions and the actin cytoskeleton in most epithelial cells. We report here the expression profile of vezatin during spermatogenesis. Vezatin is exclusively expressed in haploid germ cells. Immunocytochemical and ultrastructural analyses showed that vezatin intimately coincides, temporally and spatially, with acrosome formation. While vezatin is a transmembrane protein associated with adherens junctions in many epithelial cells, it is not seen at the ectoplasmic specializations, neither at the basal nor at the apical sites, in the seminiferous epithelium. In particular, vezatin does not colocalize with espin and myosin VIIa, two molecular markers of the ectoplasmic specialization. In differentiating spermatids, ultrastructural data indicate that vezatin localizes in the acrosome. In epididymal sperm, vezatin localizes also to the outer acrosomal membrane. Considering its developmental and molecular characteristics, vezatin may be involved in the assembly/stability of this spermatic membrane.  相似文献

利用X射线拉伸实验研究了具有二维残余应力的Cu附着膜的屈服强度与退火温度的关系.结果表明,Cu附着膜的条件屈服点随着退火温度的增高而减小,退火温度在150—300℃间变化时,减小幅度最大,出现明显的拐点.这主要由于Cu附着膜在该温度范围内发生了再结晶,使得原有的大部分组织结构强化因素消失了.Cu附着膜的屈服强度远远高于块体Cu材的屈服强度.  相似文献

This paper describes an in vivo study of the use of encapsulation to alter lipid bio-accessibility in the gastro-intestinal (GI) tract. The hypothesis that encapsulation would delay the accessibility of lipid in the GI tract was tested using subjects (N = 11) who consumed either 30 g of 13C-labelled sunflower oil encapsulated within 70 g alginate gel particles, or unencapsulated ‘free’ sunflower oil and alginate-only particles (control). Lipid accessibility was determined by the appearance of the 13C label in breath carbon-dioxide (CO2), and the particles behaviour in the GI tract was investigated using non-invasive magnetic resonance imaging (MRI).  相似文献

Mycobacteria are waterborne emerging pathogens causing infections in human. Mycobacteria have been previously isolated from wastewater and sludge, but their densities were not estimated due to cultural biases. In order to evaluate the impact of wastewater treatment processes on mycobacteria removal, we used a real time PCR method. First we compared six DNA extraction methods and second we used the more efficient DNA extraction procedure (i.e., enzymatic lysis combined with hexadecyltrimethylammonium bromide-NaCl procedure) in order to quantify Mycobacterium. With the aim to identify parameters that could serve as indicator of mycobacterial behavior, mycobacterial densities were measured in parallel to those of Escherichia coli and enterococci, and to concentrations of chemical parameters usually monitored in wastewater. Mycobacterium reached 5.5 × 10? ± 3.9 × 10? copies/L in the influent, but was not detected in the effluent after decantation and biofiltration. Most mycobacteria (98.6 ± 2.7%, i.e. 2.4 ± 0.7 log??) were removed by the physical-chemical decantation, and the remaining mycobacteria were removed by biofiltration. In contrast, enterococci and E. coli were lightly removed by decantation step and mainly removed by biofiltration. Our results showed that Mycobacterium corresponds to a hydrophobic behavior linked to insoluble compound removal, whereas enterococci and E. coli refer to hydrophilic behaviors linked to soluble compound removals.  相似文献

We reconstructed historical trends in mercury (Hg) accumulation over the past ~ 150 years in nine western Canadian alpine lakes. Recent Hg accumulation rates (fluxes) ranged between ~ 7 and 75 μg m(-2) yr(-1), which were an average of 1.8 times higher than preindustrial (i.e., pre-1850) fluxes. Increased Hg fluxes in these lakes were less than at lower elevation sites, showing that despite the potential for increased deposition, alpine lakes are no more susceptible to Hg accumulation. Unlike other studies, we found that geographic setting, changes in chlorophyll-inferred algal production, and climate were not significant predictors of [Hg] or Hg flux in lakes. Instead, our findings highlight how a combination of atmospheric deposition and site-specific processes, including organic matter supply and catchment weathering, better explain sequestration of Hg in alpine lakes.  相似文献
