Influenza neuraminidases hydrolyze the ketosidic linkage between N-acetylneuraminic acid and its adjacent galactose residue in sialosides. This enzyme is a tetrameric protein that plays a critical role in the release of progeny virions. Several methods have been described for the determination of neuraminidase activity, usually based on colorimetric, fluorescent, or chemiluminescent detection. However, only a few of these tests allow discrimination of the sialyl-linkage specificity (i.e., α2-3- versus α2-6-linked sialyllactosides) of the neuraminidase. Herein we report a glycoarray-based assay and a MALDI-TOF study for assessing the activity and specificity of two influenza neuraminidases on whole viruses. The human A(H3N2) and avian A(H5N2) neuraminidase activities were investigated. The results from both approaches demonstrated that α2-3 sialyllactoside was a better substrate than α2-6 sialyllactoside for both viruses and that H5N2 virus had a lower hydrolytic activity than H3N2.  相似文献

A comparative study was made of sorted semi-conducting single walled carbon nanotube (SWCNT) films and unsorted SWCNT films for gas sensing applications. The transmission line method is used to monitor separately the SWCNTs film resistance and the contact resistance between electrodes and the SWCNTs, thus revealing that the sensing mechanism mainly relies on a modification of the tube conductivity during gas exposure. The fabricated sensors demonstrate a detection limit of 20 ppb NO2 and 600 ppb NH3 mainly attributed to experimental setup limitations. Moreover, semi-conducting nanotubes happened to be 2.5 times more sensitive to NH3 than unsorted ones, thus proving that selectivity can be improved by sorting the SWCNTs. The temperature dependence of the sensor sensitivity was studied, and a good agreement was found between experimental results and the Langmuir adsorption model.  相似文献

The concept of modelling using a rules -based approach is introduced and developed for die prediction of the quality degradation of grains during hot air drying. It is men validated on two typical examples of quality degradation during drying: the head kernel yield of paddy rice and the wet-milling quality of maize. The linguistic approach gives a representation of die experts understanding of phenomena and leads to a good kinetic model after identification of die parameters using genetic algorithms. This study also points out die robustness of die model prediction, even if noisy data are used, and its ability to predict transient behaviours under certain conditions.  相似文献

The influence of various conditions of Inconel 738 superalloy welding or deposition welding has been studied in order to shed light on the coupling between thermal, metallurgical and mechanical states in the heat affected zone (HAZ) in which cracking may occur particularly during welding and post-weld heat treatment. Predominant crack controlling factors have been highlighted thanks to different pre-weld and post-weld heat treatments, in addition to various welding rates and pre-heating prior to welding. These factors are mainly the material ductility related to the morphology and volume fraction of intermetallic precipitates and thermally induced residual stress. It has appeared that reducing thermally induced residual stress could be more effective for preventing cracking than controlling the material ductility in the related zones thanks to adjustment of pre-weld and post-weld heat treatments. With the objective of cracking remediation, welding on preheated parts leads to lower weld power, to reduce significantly thermal gradients, decreases thermally induced stress and impedes cracks formation despite some localized and temporary decrease in alloy ductility.  相似文献

The sinterability of two industrial mullite powders, in the presence of MgO as a sintering aid, was investigated. A glassy phase, which was generated during preparation, was present in both powders; this glassy phase had a strong influence on sintering, depending on its content, composition, and spatial distribution. MgO promoted sintering in the presence of a liquid phase, both in the as-received materials and in samples washed with HF, in which most of the pre-existing glassy phase was eliminated. Investigations using transmission electron microscopy, coupled with energy-dispersive spectroscopy, as well as dilatometric measurements and X-ray diffraction data, on washed and unwashed materials and on quenched and slow-cooled samples allowed a better understanding of the influence of MgO and the glassy phase on the sintering behavior and the formation of new phases. Most of the phases, in fact, can be explained by using the MgO–Al2O3–SiO2 phase diagram, even in such complex systems.  相似文献

The effects of growth temperature and salinity on the cellular fatty acids were investigated on Aeromonas caviae, Aeromonas hydrophila and Aeromonas sobria. Under optimal growth conditions, fatty acids patterns were dominated by even-numbered chains C(16:0), C(16:1cis9), C(18:1cis11), C(12:0) and C(14:0). Growth temperature modifications induced, in the three Aeromonas species, important changes in fatty acid (i) unsaturation, (ii) branching and (iii) chain length. An important decrease in the C(18:1cis11) fatty acid content was observed for the three species below 15 degrees C and above 25 degrees C. The evolution of C(18:1cis11) and C(16:0) showed a mirror image for the three Aeromonas species. Low NaCl concentrations did not elicit significant changes in the fatty acids content of the three Aeromonas species. However, for high NaCl concentration in the medium, the growth ability was related to an important decrease of the unsaturated to saturated fatty acids ratio indicating a membrane rigidification. Thermal and salinity adaptations were branched fatty acid-dependent for A. caviae, whereas this phenomenon was less significant for A. hydrophila and A. sobria.  相似文献

Carbonic anhydrase IX (CAIX) is a tumor-specific and hypoxia-induced biomarker for the molecular imaging of solid malignancies. The nuclear- and optical-imaging of CAIX-expressing tumors have received great attention due to their potential for clinical applications. Nuclear imaging is a powerful tool for the non-invasive diagnosis of primary and metastatic CAIX-positive tumors and for the assessment of responses to antineoplastic treatment. Intraoperative optical fluorescence imaging provides improved visualization for surgeons to increase the discrimination of tumor lesions, allowing for safer surgical treatment. Over the past decades, many CAIX-targeted molecular imaging probes, based on monoclonal antibodies, antibody fragments, peptides, and small molecules, have been reported. In this review, we outline the recent development of CAIX-targeted probes for single-photon emission computerized tomography (SPECT), positron emission tomography (PET), and near-infrared fluorescence imaging (NIRF), and we discuss issues yet to be addressed.  相似文献
