A new technique, microinterferometric optical phase tomography, is introduced for use in measuring small, asymmetric refractive-index differences in the profiles of optical fibers and fiber devices. The method combines microscopy-based fringe-field interferometry with parallel projection-based computed tomography to characterize fiber index profiles. The theory relating interference measurements to the projection set required for tomographic reconstruction is given, and discrete numerical simulations are presented for three test index profiles that establish the technique's ability to characterize fiber with small, asymmetric index differences. An economical phase-transfer method is used to prepare 10-arylaminoartemisinins from DHA and arylamines, and artemether, arteether, and artelinate from the corresponding alcohols. In vivo sc screens against Plasmodium berghei and P. yoelii in mice reveal that the p-fluorophenylamino derivative 5 g is some 13 and 70 times, respectively, more active than artesunate; this reflects the very high sc activity of 10-alkylaminoartemisinins. The establishment of rational frameworks for population-level ecological risk assessment (PLERA) in the context of chemical substances management is an important issue. We illustrate two feasible approaches for establishing predicted-no-effect concentrations (PNECs)for PLERA through a case study of 4-nonylphenol (4-NP) using life-cycle toxicity data for medaka (Oryzias latipes). We first quantified the potential impacts of 4-NP on medaka in terms of reduction of population growth rate (i). An age-classified population matrix model (daily time-step) was developed and used to combine life-cycle survivorship and fecundity data obtained from individual-level responses of medaka expDsed to 4-NP into population-level responses defined by the parameter lambda. Thereafter, from the resulting lambdas, two approaches for establishing population-level PNEC values were proposed and examined. We then derived the PNEC values for population-level impacts, based on (a) the threshold concentration, defined as the chemical concentration at which lambda = 1 as a value with a 95% confidence interval, and (b) the no-observed-effect concentration (NOEC) and the maximum-acceptable-toxic concentration (MATC). The results suggest that PNEC values of 4-NP ranging between 0.82 and 2.10 microg/L affect medaka population growth. Although these approaches have their limitations, current knowledge indicates that they are reasonable and practical for evaluating population-level impacts of chemicals, thereby serving as a case study for establishing PNEC values for PLERA in the context of chemical substances management and decision-making. 相似文献

Diterpene resin acids are major constituents of conifer oleoresin and play important roles in tree defense against insects and microbial pathogens. The tricyclic C-20 carboxylic acids are generally classified into two groups, the abietanes and the pimaranes. The abietanes have conjugated double bonds and exhibit characteristic UV spectra. Here, we report the analysis of abietanes by reversed-phase high-performance liquid chromatography using multiwavelength detection to optimize quantification of underivatized abietic, neoabietic, palustric, levopimaric, and dehydroabietic acids. The utility of the method is demonstrated with methanol extracts of white spruce (Picea glauca) phloem, and representative concentrations are reported. 相似文献

A high-throughput axial MALDI-TOF mass spectrometer utilizing a laser with a 2-kHz pulse repetition-rate was constructed and tested. This fast mass spectrometer provided a data acquisition rate 10 times faster than a commercially available (200 Hz) axial mass spectrometer, while maintaining comparable limits of detection (200 amol of Glu fib peptide). Mass resolution, only slightly less than the commercial instrument (10 000 vs 14 000), was sufficient for baseline resolution of isotopic clusters of peptides with m/z <2700. A new method of mass calibration, which combined a limited number of internal and external standards, provided the same 15 ppm mass accuracy over the entire sample plate on either instrument. Implementing the 2-kHz laser required a faster data acquisition system and high-voltage pulse electronics, together with a novel strategy for rapid sample plate movements during acquisition, to achieve a sample analysis rate of up to 2 spots/s (with 800 shots/spot). The overall performance of the fast MALDI-TOF MS instrument was demonstrated by the acquisition, in 12 min, of an LC-MS data set from a plate of 625 fractions collected during LC separation of an 16O/18O differentially labeled proteomic sample of a tryptic digest of an E. coli lysate. 相似文献

Knowledge of explosives sorption and transformation processes is required to ensure that the proper fate and transport of such contaminants is understood at military ranges and ammunition production sites. Bioremediation of 2,4,6-trinitrotoluene (TNT), hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX), and related nitroaromatic compounds has met with mixed success, which is potentially due to the uncertainty of how energetic compounds are bound to different soil types. This study investigated the dissolution and sorption properties of TNT and RDX explosives associated with six different soil types. Understanding the associations that explosives have with a different soil type assists with the development of conceptual models used for the sequestration process, risk analysis guidelines, and site assessment tools. In three-way systems of crystalline explosives, soil, and water, the maximum explosive solubility was not achieved due to the sorption of the explosive onto the soil particles and observed production of transformation byproducts. Significantly different sorption effects were also observed between sterile (gamma-irradiated) and nonsterile (nonirradiated) soils with the introduction of crystalline TNT and RDX into soil-water systems. 相似文献

A cryogenically cooled linear electrostatic ion beam trap for use in photoelectron-photofragment coincidence (PPC) spectroscopy is described. Using this instrument, anions created in cold, low-duty-cycle sources can be stored for many seconds in a ~20 K environment to cool radiatively, removing energetic uncertainties due to vibrationally excited precursor anions. This apparatus maintains a well-collimated beam necessary for high-resolution fragment imaging and the high experimental duty cycle needed for coincidence experiments. Ion oscillation is bunched and phase-locked to a modelocked laser, ensuring temporal overlap between ion bunches and laser pulses and that ions are intersected by the laser only when travelling in one direction. An electron detector is housed in the field-free center of the trap, allowing PPC experiments to be carried out on ions while they are stored and permitting efficient detection of 3-dimensional electron and neutral recoil trajectories. The effects of trapping parameters on the center-of-mass trajectories in the laser-ion interaction region are explored to optimize neutral particle resolution, and the impact of bunching on ion oscillation is established. Finally, an initial demonstration of radiative cooling is presented. 相似文献
