The effect of both the type of primary treatment (hydrolitic up-flow sludge blanket (HUSB) reactor and conventional settling) and the flow regime (batch and continuous) on clogging development in subsurface flow constructed wetlands (SSF CWs) was studied. Clogging indicators (such as accumulated solids, hydraulic conductivity and drainable porosity) were determined in an experimental plant with three treatment lines. Correlations were encountered between the solids accumulated and both saturated hydraulic conductivity and drainable porosity reduction over time (74.5% and 89.2% of correlation, respectively). SSF CW implemented with a HUSB reactor accumulated ca. 30% lower sludge (1.9 kg DM/m2) than a system with a settler (2.5-2.8 kg DM/m2). However, no significant differences were recorded among treatment lines concerning hydraulic parameters (such as hydraulic conductivity or porosity). Root system development contributed to clogging. Accordingly, planted wetlands showed between 30% and 40% and 10% lower hydraulic conductivity and porosity reduction, respectively, than non-planted wetlands. 相似文献

Eighty-six water samples were collected in early 2009 from Costa Rican surface water and coastal locations for the analysis of 34 pharmaceutical and personal care product compounds (PPCPs). Sampling sites included areas receiving treated and untreated wastewaters, and urban and rural runoff. PPCPs were analyzed using a combination of solid phase extraction and liquid chromatography tandem mass spectrometry. The five most frequently detected compounds were doxycycline (77%), sulfadimethoxine (43%), salicylic acid (41%), triclosan (34%) and caffeine (29%). Caffeine had the maximum concentration of 1.1 mg L−1, possibly due to coffee bean production facilities upstream. Other compounds found in high concentrations include: doxycycline (74 μg L−1), ibuprofen (37 μg L−1), gemfibrozil (17 μg L−1), acetominophen (13 μg L−1) and ketoprofen (10 μg L−1). The wastewater effluent collected from an oxidation pond had similar detection and concentrations of compounds compared to other studies reported in the literature. Waters receiving runoff from a nearby hospital showed higher concentrations than other areas for many PPCPs. Both caffeine and carbamazepine were found in low frequency compared to other studies, likely due to enhanced degradation and low usage, respectively. Overall concentrations of PPCPs in surface waters of Costa Rica are inline with currently reported occurrence data from around the world, with the exception of doxycycline. 相似文献

The aim of this work was to establish whether Lactococcus lactis strains isolated from spontaneous dairy fermentations exhibited useful milk-processing capabilities and resistance to bacteriophage infection in order to be used as components in starter formulations. The 33 out of 100 isolates of L. lactis, originated from farmhouse cheeses, were found to be resistant to a collection of 34 phages belonging to the c2 and 936 groups. Six of the isolates were discarded as potential starters because they were lysogenic and other five because they produced tyramine. Plasmid and chromosomal profiles of the 22 remaining isolates allowed their classification into 16 different strains. All of these were good lactic acid producers from lactose, moderately proteolytic and, in eight cases, diacetyl production from citrate was observed. The mechanism(s) leading to the phenotype of phage resistance was identified for all the strains used in this study. Inhibition of adsorption was the most frequent one, although genetic determinants for some abortive infection systems were also detected (abiB, abiG and abiI). Frequently, more than one mechanism was present in the same strain. One of the strains, L. lactis IPLA542, was selected as a model starter for pilot fermentations. It clotted milk normally both in the absence and in the presence of phage at concentrations that completely abolished the process when promoted by a phage-susceptible strain. 相似文献

The effect of addition of cereal and fruit dietary fibres on the sensory properties of reduced-fat, dry fermented sausages was studied. Dry fermented sausages with 6 and 10% pork backfat were manufactured, with addition of cereal (wheat and oat) and fruit (peach, apple and orange) dietary fibres, at 1.5 and 3% concentrations. The energy value reduction of the final products was close to 35% and their final fibre contents, after ripening, were 2 and 4%, respectively. The ripening process was monitored by physico-chemical and microbiological analysis. Sensory properties were analyzed using triangular and hedonic tests and, a texture profile analysis was carried out. A correlation principal component analysis was performed. The results showed that the sensory and textural properties of batches with 3% dietary fibre were the worst, due to their hardness and cohesiveness. The best results were obtained with sausages containing 10% pork backfat and 1.5% fruit fibre especially those with orange fibre, which gave organoleptic characteristics similar to conventional high fat products. Thus, reduced fat sausages fortified with dietary fibre can be obtained with an acceptable sensory profile. 相似文献

Volatile compounds produced in cheese by five Pseudomonas fragi strains isolated from 1-day-old raw milk cheeses were investigated. Each strain was representative of a different biochemical group of isolates of identical phenotypic characteristics, according to identification with API 20 NE strips. The five strains were ascribed to the species P. fragi after 16S rRNA sequencing because of their high degree of coincidence with P. fragi ATCC 4973. In each of two experiments, carried out on different days, five cheeses were made at laboratory scale from pasteurized milk separately inoculated with approximately 10(5) CFU/ml of each P. fragi strain. After 12 days at 10 degrees C, mean counts of P. fragi strains were close to 10(10) CFU/g in the outer part of cheeses and close to 10(8) CFU/g in the inner part. A total of 131 volatile compounds, 49 of which were further characterized, were identified in cheeses by gas chromatography-mass spectrometry after extraction with a purge and trap apparatus. Abundances of compounds were generally higher in the outer part of cheeses. Production of volatile compounds was clearly strain dependent. Only two strains produced ethyl esters, and three produced nonethyl esters. Ethyl acetate, ethyl butyrate, ethyl caproate, methyl acetate, isopropyl acetate, and propyl tiglate were the major esters, and ethanol, 2-propanol, and 3-methyl butanol were the major alcohols. Undecene was the major hydrocarbon, dimethyl sulfide and methyl thiocyanate the major sulfur compounds, and 2-pentanone the major ketone. In this study four cultivars of lentil originating from Spain were examined: cv Paula, cv Agueda, cv Almar and cv Alcor. Since consumption of these seeds after heat treatment and as sprouts has been popularised, the impact of cooking (up to 30 min) and germination process (in dark, at 25 degrees C, for up to 4 days) on peroxyl radical-trapping capacity (PRTC) and Trolox-equivalent antioxidant capacity (TEAC) of the processed seeds was addressed. Also, changes in the content of low-molecular-weight antioxidants (LMWA) and soluble proteins in the course of cooking and germination were studied. The analyzed LMWAwere: total phenolics, tocopherols (alpha-T, beta-T, gamma-T, delta-T), reduced glutathione, and L-ascorbic acid. On the basis of the results obtained, the contribution of LMWA and soluble proteins to the PRTC and TEAC of raw, cooked, and germinated lentil seeds was calculated by multiple mean values for the content of investigated compounds and their relative potential with respect to Trolox. The results showed avery high molar percentage contribution of phenolic compounds and low contribution of tocopherols, glutathione, soluble proteins, and ascorbate (only in germinated seeds) to the total TEAC and total PRTC calculated as a sum of data provided for phosphate-buffered and 80% methanolic extracts of raw and processed lentil seeds. 相似文献

Ghrelin, the endogenous ligand of GH secretagogue receptor type 1a, has emerged as a pleiotropic modulator of diverse biological functions, including energy homeostasis and, lately reproduction. Here, we review recent reports evaluating the reproductive effects and sites of action of ghrelin, with particular emphasis regarding its role as a molecule integrating reproductive function and energy status. Data gleaned from rodent studies clearly show that besides having direct gonadal effects, ghrelin may participate in the regulation of gonadotropin secretion and it may influence the timing of puberty. In addition, experimental data showing that ghrelin and/or its receptor are expressed in normal human ovary and testis as well as in human ovarian and testicular tumors raise the possibility that the ghrelin system may be involved in the control of cell proliferation in these tumors. We propose that ghrelin either acting as an endocrine and/or paracrine signal may play a major role in the endocrine network that integrates energy balance and reproduction. 相似文献

The relationship between membrane permeabilization and loss of viability by pulsed electric fields (PEF) depending on the treatment intensity and the treatment media pH in two gram-positive (Lactobacillus plantarum, Listeria monocytogenes) and two gram-negative (Escherichia coli, Salmonella senftenberg 775W) bacterial species has been investigated. Loss of membrane integrity was measured as increased uptake of the fluorescent dye propidium iodide (PI). Non-permanent/reversible permeabilization was detected when cells stained with PI during PEF resulted in higher fluorescence than that measured in cells stained after PEF. Whereas loss of viability of the two gram-negative bacteria was correlated with the sum of non-permanent and permanent membrane permeabilization when treated at pH 7.0, in the case of the two gram-positives, loss of viability was correlated with a permanent loss of membrane integrity. At pH 7.0, the four bacteria exhibited reversible permeabilization. However, whereas the gram-positives capable of reversing permeabilization survived, the gram-negative cells died, despite their capacity to reverse permeabilization immediately after PEF. Thus, resealing is not necessarily related to the survival of PEF-treated cells. In contrast, when cells were PEF-treated at pH 4.0 a more complicated picture emerged. Whereas loss of viability was correlated with a permanent loss of membrane integrity in L. monocytogenes cells, in L. plantarum the degree of permeabilization was higher, and in the gram-negative strains, much lower than the percentage of inactivated cells. These results support the view that membrane permeabilization is involved in the mechanism of bacterial inactivation by PEF, but the nature of membrane damage and its relationship with cell death depends on the bacterial species and the treatment medium pH. 相似文献
