Respiratory syncytial virus (RSV) is the leading cause of lower respiratory tract infection in children and infants. To date, there is no effective vaccine available against RSV. Heparan sulfate is a type of glycosaminoglycan that aids in the attachment of the RSV to the host cell membrane via the G protein. In the present study, the effect of amino acid substitution on the structure and stability of the ectodomain G protein was studied. Further, it was investigated whether mutation (K117A) in the CX3C motif of G protein alters the binding with heparan sulfate. The point mutation significantly affects the conformational stability of the G protein. The mutant protein showed a low binding affinity with heparan sulfate as compared to the wild-type G protein, as determined by fluorescence quenching, isothermal titration calorimetry (ITC), and molecular docking studies. The low binding affinity and decreased stability suggested that this mutation may play an important role in prevention of attachment of virion to the host cell receptors. Collectively, this investigation suggests that mutation in the CX3C motif of G protein may likely improve the efficacy and safety of the RSV vaccine.  相似文献

In vivo and in situ trials were conducted to investigate the utilization of alfalfa protein concentrate as a protein supplement for lactating dairy cows. In the in vivo trial, four Holstein cows fitted with ruminal, duodenal, and ileal cannulae were used in a crossover experiment to measure site and extent of nutrient digestion. Isonitrogenous and isoenergetic diets containing alfalfa protein concentrate or soybean meal were fed four times daily. Sixty percent of dietary total N was provided by the test proteins. Ruminal ammonia and total VFA concentrations were lower in cows fed alfalfa protein concentrate. Fractional digestion coefficient of organic matter (52 vs. 43%) and degradation of dietary protein (57 vs. 47%) in the rumen were slightly lower in cows fed alfalfa protein concentrate. The ratio of amino acid flow to the small intestine to dietary intake of amino acid was higher in cows fed alfalfa protein concentrate. In the in situ trial, rate of N disappearance (2.4 vs. 9.5%.h-1) and potential protein degradability (35 vs. 68%) were lower for alfalfa protein concentrate. Results suggest that alfalfa protein concentrate may be more resistant to ruminal degradation than soybean meal and could be utilized as a relatively undegradable protein source for milk production.  相似文献

Four lactating Holstein cows fitted with rumen cannulae and T-type cannulae in proximal duodenum and terminal ileum were used to measure protein degradation in the rumen and amino acid flow and absorption in the small intestine. Soybean meal, whole soybeans, and whole soybeans extruded at 132 and 149 degrees C provided 50% of the protein in diets that contained 51% grain, 36% corn silage, and 13% alfalfa hay (dry matter). Spot samples of digesta were collected from duodenum and ileum during 96 h, and lanthanum was an indigestible marker to estimate flow and digestibility of nutrients. With diaminopimelic acid as a microbial marker, apparent degradations of dietary crude protein in the rumen were 73, 80, 66, and 60% for diets containing soybean meal, whole soybeans, and whole soybeans extruded at 132 and 149 degrees C. Because of the extensive degradation of protein with the raw soybean diet, less total amino acids reached the duodenum, and because of decrease of availability, absorption from the small intestine (g/day) was lowest with this diet. Feeding diets containing extruded whole soybeans increased availability of total essential amino acids in the small intestine compared with diets containing soybean meal and whole soybeans. Absorption from the small intestine (g/day and percent entering) of individual amino acids was generally higher for extruded whole soybean diets.  相似文献

Staggered arrays of dimples imprinted on opposite surfaces of an internal flow channel have been formulated numerically to enhance turbulent heat transfer compromising with pressure drop. The channel is simulated with the help of three-dimensional Reynoldsaveraged Navier-Stokes (RANS) analysis. Three nondimensional design variables based on dimple size and channel dimensions and two objectives related to heat transfer and pressure drag have been considered for shape optimization. A weighted-sum method for multi-objective optimization is applied to integrate multiple objectives into a single objective and polynomial response surface approximation (RSA) coupling with a gradient based search algorithm has been implemented as optimization technique. By the present effort, heat transfer rate is increased much higher than pressure drop and the thermal performance also has shown improvement for the optimum design as compared to the reference one. The optimum design produces lower channel height, wider dimple spacing, and deeper dimple as compared to the reference one.  相似文献

This study evaluated the effect of coarse or fine grinding of three forms of corn on the performance of lactating cows. Six diets, fed as total mixed rations, were identical except for the corn portion of the diet. Corn treatments were dry shelled corn, high moisture ensiled ear corn, and high moisture ensiled shelled corn, either coarsely or finely ground. The experimental design was a 6 x 6 Latin square with 36 cows. Eighteen cows were assigned to the six different treatments and were fed once daily. Within this group of 18 cows, six had a ruminal cannula and were used to evaluate nutrient digestibilities and ruminal fermentation. The remaining 18 cows, six of which were ruminally cannulated, were similarly assigned, except they were fed twice daily. In the group fed once daily, milk production and composition were not affected by treatment. Starch digestibility was greater with the high moisture and with the finely ground corn treatments. In addition, the high moisture ensiled corn treatments had reduced ruminal ammonia concentrations. In the group that was fed twice daily, milk production and protein yield were greatest for the finely ground high moisture ensiled shelled corn treatment. Starch utilization was improved by fine grinding. Lower ruminal ammonia concentrations were obtained with the high moisture ensiled corn treatments, and there was a tendency for reduced ammonia concentration with fine grinding. Results indicate that high moisture ensiled corn as well as fine grinding improved nitrogen and starch utilization.  相似文献

A promising energy storage material containing magnesium, nickel and aluminum metals has been synthesized using conventional coprecipitation method. The physio-chemical properties, morphology and microstructure of hydrotalcite based mixed oxides (MNAM) have been characterized using ICP-MS, XRD, BET, FESEM and TEM techniques. The characterizations revealed that the adsorbent are highly dispersed, poly crystalline, homogenously distributed and interactive. Temperature programmed techniques have been used to explore the sorption capacities of mixed oxides. Nano-crystalline mesoporous and interactive reduced mixed oxides exhibit a 3.9 wt% H2 adsorption capacity at ambient conditions. Temperature programmed desorption study predicted a desorption capacity of 1.9 wt% H2. Different hydrogen adsorbed phases of magnesium and nickel have been characterized using HRTEM, Raman and FT-IR techniques.  相似文献

A new self-consistent technique is proposed to simultaneously extract the density of interface traps (Dit) and flat-band voltages of MOS structures fabricated on technologically relevant high-mobility semiconductors with arbitrary combination of gate stacks. The technique is based on novel analysis of the low-frequency C-V measurement. The two major problems associated with the existing low-frequency C-V technique for arbitrary substrate/oxide combinations are resolved by (i) accurate calculation of the ideal semiconductor capacitance using a self-consistent, quantum-mechanical model including wave function penetration effect, and (ii) accurate determination of the flat-band voltage utilizing an iterative scheme. The proposed technique has been applied to extract Dit profiles of a number of MOS structures fabricated on III-V semiconductors like InGaAs (with ALD grown Al2O3 gate dielectric) and elemental semiconductors like Ge (with GeON gate dielectric). The advantages of the proposed technique have been demonstrated by comparing with Dit profiles extracted from other capacitor-based extraction methods.  相似文献

Sixty-four Holstein cows (32 primiparous) were assigned randomly at parturition to one of two diets differing in forage:grain ratio and were further assigned to either control or treatment with recombinant bST. A full lactation study was conducted. Total mixed rations consisted of alfalfa silage, high moisture ear corn, soybean meal, and vitamin and mineral supplements. Percentages of forage in the diet (dry basis) for the high and medium energy diets were wk 1 to 12, 38.2 and 58.2; wk 13 to 26, 48.2 and 68.2; and wk 27 to 44, 68.2 and 88.2. Injection of a saline placebo or 20.6 mg/d per cow of bST started at wk 13 and stopped at wk 43 of lactation. Administration of bST increased 305-d milk yield by 19.9 and 18.3% for multiparous cows fed high and medium grain diets and by 13.0 and 5.9% for primiparous cows fed the same diets. Body weight gain during lactation tended to be lower for cows treated with bST, and body condition scores were significantly lower for those treated with bST and fed the medium energy diets. Daily injection of bST did not change concentrations of glucose or urea in blood or beta-hydroxybutyrate in plasma. Multiparous cows had a transitory increase in plasma FFA concentration immediately after bST treatment started. Milk composition, health, and reproductive parameters observed were not adversely affected by bST administration. Cows fed low energy diets during bST administration in the first lactation did not produce as much milk as controls when compared during the first 10 wk of their second lactation.  相似文献

Conjugated linoleic acid in milk was determined from cows fed different diets. In Experiment 1, cows were fed either normal or high oil corn and corn silage. Conjugated linoleic acid was 3.8 and 3.9 mg/g of milk fatty acids in normal and high oil treatments, respectively. In Experiment 2, cows consumed one-third, two-thirds, or their entire feed from a permanent pasture. Alfalfa hay and concentrates supplied the balance of feed for the one-third and two-third pasture treatments. Conjugated linoleic acid was 8.9, 14.3, and 22.1 mg/g of milk fatty acids in the one-third, two-third, and all pasture treatments, respectively. Cows grazing pasture and receiving no supplemental feed had 500% more conjugated linoleic acid in milk fat than cows fed typical dairy diets (Experiment 1). In Experiment 3, cows were fed either a control diet containing 55% alfalfa silage and 45% grain, or similar diets supplemented with 3% fish meal, or 250 g of monensin/cow/per day, or fish meal and monensin together. Conjugated linoleic acid was 5.3, 8.6, 6.8, and 8.9 mg/g of milk fatty acids in the control, fish meal, monensin, and fish meal plus monensin treatments, respectively. In Experiment 4, cows were fed either finely chopped alfalfa hay (Treatment 1), or coarsely chopped alfalfa hay (Treatment 2) in a 50% forage and 50% grain diet, or 66.6% grass hay and 33.4% grain (Treatment 3), or 98.2% grass hay (Treatment 4). Conjugated linoleic acid was 7.3, 8.3, 9.0, and 7.9 mg/g of milk fatty acids in treatments 1 through 4, respectively.  相似文献

A method for approximating spherical topology digital shapes by rational Gaussian (RaG) surfaces is presented. Points in a shape are parametrized by approximating the shape with a triangular mesh, determining parameter coordinates at mesh vertices, and finding parameter coordinates at shape points from interpolation of parameter coordinates at mesh vertices. Knowing the locations and parameter coordinates of the shape points, the control points of a RaG surface are determined to approximate the shape with a required accuracy. The process starts from a small set of control points and gradually increases the control points until the error between the surface and the digital shape reduces to a required tolerance. Both triangulation and surface approximation proceed from coarse to fine. Therefore, the method is particularly suitable for multiresolution creation and transmission of digital shapes over the Internet. Application of the proposed method in editing of 3D shapes is demonstrated.  相似文献
