Peptidases occupy a central position in the enzyme market because of their importance in many areas, such as for physiological processes, foods, and detergents, as well as in the pharmaceutical, leather, and biotechnology industries. Microbial production is among the major sources of peptidases because it presents many advantages when compared with other methods. In this study, the metallopeptidases produced by the fungus Eupenicillium javanicum under a solid-state fermentation bioprocess were spray-dried. The enzymatic extract was dried using drying adjuvants, and optimal conditions for preserving enzymatic activity were studied following a Box-Behnken experimental design. The spray process factors studied were the air-drying temperature, enzyme feed flow rate, and the proportion of enzyme/additive. The responses analyzed were the dry extract yield and enzymatic activity after spray drying. Additionally, the stability of the dry extracts was assessed during 180 days at 4°C and 25°C. The results revealed extract yields of up to 66.12% and good enzymatic activity for intermediate values of temperature and adjuvant proportions. Furthermore, the dried enzymatic extracts showed potential for future commercial applications because of their stability at 25°C for 180 days.  相似文献

Polylactide (PLA) is the most used biodegradable and biobased food packaging polymer for rigid containers and films. However, its low ductility is a hurdle for increasing its applications in flexible food packaging. A solution is the use of additives. Palm oil deodorizer distillate (PODC) is revealed to be an excellent additive promoting PLA ductility. PODC is a by‐product of vegetable oil refining, which is available in stable quality and in sufficient amounts. Amorphous PLA/PODC blends had an elongation at break of around 130% and that of semi‐crystalline blends was still around 55% compared to the initial 5% of neat PLA. At the same time the PLA rigidity and high glass transition temperatures were kept. PODC was also a very efficient processing aid, allowing for film blow extrusion. The blends were stable in properties during six months without exudation. They complied with legal norms of Food Contact Materials (EU 10/2011) and induced no sensorial alteration of packed food. Therefore PODC is a very interesting alternative to common plasticizers for the production of flexible PLA packaging films. © 2016 Society of Chemical Industry  相似文献

Many techniques have been used to coat metallic substrate with bioceramics. The aim of this study was to study the physical-chemical characteristics of polyvinylidene fluoride (α-PVDF)/hydroxyapatite (HA) composite coating, obtained by casting method, on commercially pure titanium (α-CP Ti) substrate surface modified by laser beam irradiation. The preparation of coating was done for mixing α-PVDF pellets shape dissolved in dimethylacetamide (DMA) with HA/DMA emulsion. The mixture was poured onto the α-CP Ti sample and left to dry in an oven. CP Ti plates were coated with α-PVDF/HA composite film, in proportions of 100/00 and 60/40 in weight, and characterized by particle size analysis, scanning electron microscopy, energy dispersive spectroscopy (EDS), X-ray diffractometry, thickness measurement and contact angle. Uniform coating with a small thickness variation along the coated surface was successfully obtained.  相似文献

We present a novel comprehensive optimization model for designing reconfigurable machining lines. Due to the proposed fine mathematical modelling, it is possible to optimize simultaneously the whole set of machines and machining modules as well as their cutting parameters, their configuration that will be used for processing of each part and part position at each machine. The experimental results show that the proposed optimization approach substantially outperforms the existing heuristic design method and therefore it can be used by the designers in order to reduce the total system cost and improve the efficiency of reconfigurable machining lines.  相似文献

The kinetic model, established in a previous article (François‐Heude et al., J. Appl. Polym. Sci., in press) to predict the homogeneous oxidation in iPP films typically thinner than 100 µm, is now extended to simulate the oxidation profiles in thicker plates by coupling the oxygen diffusion and its consumption by the chemical reactions. In this perspective, oxygen transport properties (namely oxygen solubility, diffusivity, and permeability) are measured by permeametry on a reference iPP. These values are compared with an exhaustive compilation of literature data to evaluate their variability among the whole iPP family, which one has been reasonably ascribed to initial differences in polymer morphology, but also to evaluate their consistency, especially their temperature dependence between 20 and 140°C. Failing to simulate oxidation profiles, the kinetic model is then used as an inverse resolution method for estimating more satisfactory values of oxygen transport properties. It is thus evidenced that the crystallinity changes induced by thermal oxidation largely explains the dramatic decrease in oxygen penetration toward the sample core just after the induction period. A strategy aimed for introducing the relationship between the polymer crystalline morphology and oxygen transport properties into the kinetic model is given in the graphical abstract, although the effect of polymer polarity remains to be established prior to this implementation. © 2014 Wiley Periodicals, Inc. We aimed at investigating oxidative stability and changes in fatty acid and tocopherol composition of extra virgin olive oil (EVOO) in comparison with refined seed oils during short‐term deep‐frying of French fries, and changes in the composition of the French fries deep‐fried in EVOO. EVOO samples from Spain, Brazil, and Portugal, and refined seed oils of soybean and sunflower were studied. Oil samples were used for deep‐frying of French fries at 180 °C, for up to 75 min of successive frying. Tocopherol and fatty acid composition were determined in fresh and spent vegetable oils. Tocopherol, fatty acid, and volatile composition (by SPME–GC–MS) were also determined in French fries deep‐fried in EVOO. Oil oxidation was monitored by peroxide, acid, and p‐anisidine values, and by Rancimat after deep‐frying. Differential scanning calorimetry (DSC) analysis was used as a proxy of the quality of the spent oils. EVOOs presented the lowest degree of oleic and linoleic acids losses, low formation of free fatty acids and carbonyl compounds, and were highly stable after deep‐frying. In addition, oleic acid, tocopherols, and flavor compounds were transferred from EVOO into the French fries. In conclusion, EVOOs were more stable than refined seed oils during short‐term deep‐frying of French fries and also contributed to enhance the nutritional value, and possibly improve the flavor, of the fries prepared in EVOO.  相似文献

Intra-articular drug delivery systems (DDSs) are envisaged as interesting alternative to locally release non-steroidal anti-inflammatory drugs (NSAIDs) such as ibuprofen to reduce pain in patients with osteoarthritis. The present study examines the efficacy of S-(+)-ibuprofen on cartilage degradation as drug candidate for DDS loading. Humeral cartilage and joint capsule explants were collected from healthy sheep shoulder joints and they were cultured in mono- or in co-culture for 13?days with LPS in combination with S-(+)-ibuprofen at 50?µM and 1?mM. S-(+)-ibuprofen (50?µM) blocked prostaglandins production in LPS-activated explants but did not reduce cartilage degradation. By contrast, 1?mM S-(+)-ibuprofen treatment of cartilage explants reduced nitric oxide synthesis by 51% (p?=?0.0072), proteoglycans degradation by 35% (p?=?0.0114) and expression of serum amyloid protein – the main protein induced upon LPS challenge – by 44% (p?相似文献

This study is composed of three essential parts. The first part describes an indirect semi-experimental method which is used to reconstruct the excitation force of an operating diesel engine from the acceleration data measured at the mounting points. These internal forces can not be directly measured with force sensors; they have to be derived from the dynamic deformation of the engine support, so a theoretical analysis is carried out to derive the equations for the force re-construction.The second part deals with prevention of low frequency vibration of the powertrain from spreading to the rest of the vehicle. Three uncoupling techniques are used to minimize these vibrations. The first technique reduces the non-diagonal elements of stiffness matrix. The second technique uses the elastic axes decoupling criterion. The third technique uncouple the torque roll axis (TRA) by using the previously determined excitation efforts.In the third part, numerical and experimental results are discussed. The solicitations deducted and the positions of mounts allowing decoupling of the powertrain are presented.  相似文献
