In this article, we address the problem of adaptive state observation of linear time-varying systems with delayed measurements and unknown parameters. Our new developments extend the results reported in our recently works. The case with known parameters has been studied by many researchers. However in this article we show that the generalized parameter estimation-based observer design provides a very simple solution for the unknown parameter case. Moreover, when this observer design technique is combined with the dynamic regressor extension and mixing estimation procedure the estimated state and parameters converge in fixed-time imposing extremely weak excitation assumptions. 相似文献

A renewable source of carbon black is introduced by the processing of lignin from agro-forestry residues. Lignin side streams are converted into spherical particles by direct aerosolization followed by carbonization. The obtained submicron black carbon is combined with cellulose nanofibers, which act as a binder and rheology modifier, resulting in a new type of colloidal bioink. The bioinks are tested in handwriting and direct ink writing. After consolidation, the black bioinks display total light reflectance (%R) at least three times lower than commercial black inks (reduction from 12 to 4%R). A loading of up to 20% of nanofibers positively affects the cohesion of the dried bioink (1 to 16 MPa), with no significant reduction in light reflectance. This is a result of the superstructuring of the ink components, which disrupts particle packing, intensifies colloidal interactions, introduces light absorption, and non-reflective multiple scattering. 相似文献

The introduction of MINFLUX nanoscopy allows single molecules to be localized with one nanometer precision in as little as one millisecond. However, current applications have so far focused on increasing this precision by optimizing photon collection, rather than minimizing the localization time. Concurrently, commonly used fluorescent switches are specifically designed for stochastic methods (e.g., STORM), optimized for a high photon yield and rather long on-times (tens of milliseconds). Here, accelerated MINFLUX nanoscopy with up to a 30-fold gain in localization speed is presented. The improvement is attained by designing spontaneously blinking fluorescent markers with remarkably fast on-times, down to 1–3 ms, matching the iterative localization process used in a MINFLUX microscope. This design utilizes a silicon rhodamine amide core, shifting the spirocyclization equilibrium toward an uncharged closed form at physiological conditions and imparting intact live cell permeability, modified with a fused (benzo)thiophene spirolactam fragment. The best candidate for MINFLUX microscopy (also suitable for STORM imaging) is selected through detailed characterization of the blinking behavior of single fluorophores, bound to different protein tags. Finally, optimization of the localization routines, customized to the fast blinking times, renders a significant speed improvement on a commercial MINFLUX microscope. 相似文献

Most of the currently known fungal laccases show their maximum activity under acidic environmental conditions. It is known that a decrease in the activity of a typical laccase at neutral or alkaline pH values is the result of an increase in the binding of the hydroxide anion to the T2/T3 copper center, which prevents the transfer of an electron from the T1 Cu to the trinuclear copper center. However, evolutionary pressure has resolved the existing limitations in the catalytic mechanism of laccase, allowing such enzymes to be functionally active under neutral/alkaline pH conditions, thereby giving fungi an advantage for their survival. Combined molecular and biochemical studies, homological modeling, calculation of the electrostatic potential on the Connolly surface at pH 5.0 and 7.0, and structural analysis of the novel alkaliphilic laccase of Myrothecium roridum VKM F-3565 and alkaliphilic and acidophilic fungal laccases with a known structure allowed a new intramolecular channel near the one of the catalytic aspartate residues at T2-copper atom to be found. The amino acid residues of alkaliphilic laccases forming this channel can presumably serve as proton donors for catalytic aspartates under neutral conditions, thus ensuring proper functioning. For the first time for ascomycetous laccases, the production of new trimeric products of phenylpropanoid condensation under neutral conditions has been shown, which could have a potential for use in pharmacology. 相似文献

Furan-based amines are highly valuable compounds which can be directly obtained via reductive amination from easily accessible furfural, 5-(hydroxymethyl)furfural (HMF) and 2,5-diformylfuran (DFF). Herein the biocatalytic amination of these carbonyl derivatives is disclosed using amine transaminases (ATAs) and isopropylamine (IPA) as amine donors. Among the different biocatalysts tested, the ones from Chromobacterium violaceum (Cv-TA), Arthrobacter citreus (ArS-TA), and variants from Arthrobacter sp. (ArRmut11-TA) and Vibrio fluvialis (Vf-mut-TA), afforded high levels of product formation (>80 %) at 100–200 mM aldehyde concentration. The transformations were studied in terms of enzyme and IPA loading. The pH influence was found as a key factor and attributed to the imine/aldehyde equilibrium that can arise from the high reactivity of the carbonyl substrates with a nucleophilic amine such as IPA. 相似文献

The paper presents a new parallel language, mpC, designed specially for programming high‐performance computations on heterogeneous networks of computers, as well as its supportive programming environment. The main idea underlying mpC is that an mpC application explicitly defines an abstract network and distributes data, computations and communications over the network. The mpC programming environment uses, at run time, this information as well as information on any real executing network in order to map the application to the real network in such a way that ensures the efficient execution of the application on this real network.
