This work describes an analytical method using liquid chromatography coupled with tandem-mass spectrometry (LC–MS/MS) with triple quadrupole in selective reaction monitoring mode for the simultaneous determination of ascorbic acid (AA) and dehydroascorbic acid (DHAA) in pepper, tomato, orange and lemon. The negative ion mode of ESI and MS/MS transitions of m/z 173→143 and m/z 173→71 were used for AA and m/z 175→115 and m/z 175→87 for DHAA. All the validation parameters were found to be within the range of acceptance for both compounds. Different matrix effects were observed in the studied vegetables and samples were quantified by the standard addition approach. The proposed method was suitable for simultaneously determining AA and DHAA with no need for oxidation/reduction or derivatisation steps. The main advantages of this method are its simplicity (little sample preparation), speed (total analysis time does not exceed 5 min) and excellent sensitivity (limits of detection (LODs) of 13 and 11 ng/ml for AA and DHAA, respectively). 相似文献

Low fat dry fermented sausages were manufactured using controlled ripening conditions and a slow fermented process. The effect of fat content and ripening time on the chemical, colour, texture parameters and sensory acceptability was studied. The fat reduction in slow fermented sausages produced an increase in the pH decline during the first stage of the process that was favoured by the higher water content of the low fat sausages. Fat reduction did not affect the external appearance and there was an absence of defects but lower fat content resulted in lower sausage lightness. The sausage texture in low fat sausages caused an increase in chewiness and at longer ripening times, an increase in hardness. The sensory acceptability of the fermented sausages analyzed by internal preference mapping depended on the different preference patterns of consumers. A group of consumers preferred sausages with high and medium fat content and high ripening time. The second group of consumers preferred sausages with low ripening time regardless of fat content except for the appearance, for which these consumers preferred sausages of high ripening time. Finally, the limit to produce high acceptability low fat fermented sausages was 16% fat content in the raw mixture that is half the usual content of dry fermented sausages. 相似文献

Modeling fear in animals is a critical approach for identifying the neural mechanisms involved in human disorders such as generalized anxiety and panic. Amygdala kindling has proven useful in this regard because it produces dramatic increases in fearful behavior. The purpose of this experiment was to compare the behavioral effects of kindling in male and female rats. Compared with the sham-stimulated rats, the kindled male and female rats showed similar increases in fearful behavior, with some sex differences in fear-related open-field activity. They also showed decreased immobility in the forced-swim test and increased sucrose consumption. These results suggest that kindling-induced fear is generally similar in male and female rats and that kindling does not appear to induce depression-like behavior. Ion beam mixing has been studied since 1980, and since then a lot of experimental and theoretical work has been done and knowledge has been gathered. Nevertheless, there are still many fundamental aspects that need to be clarified and with that aim many experiments need to be performed. Copper and iron are miscible in the liquid state, while silver and iron are not. However, both systems are thermally immiscible in the solid state. In order to have an insight into the importance of mixing within thermal spikes during ion beam irradiation, we deposited Cu/Fe and Ag/Fe bilayers onto Si substrates and irradiated them at room temperature with 2 MeV Cu and 2.5 MeV Au ions. A combination of Rutherford backscattering spectrometry (RBS) and atomic force microscopy (AFM) was used to analyze the atomic transport at the interface and the morphology changes of the samples. From the element profiles at the interface we conclude a mixing efficiency, which is indeed larger than the prediction of the ballistic model in the Cu/Fe system and smaller in the Ag/Fe system. Since ballistic mixing is expected in any case, we argue that demixing and phase separation in the Ag/Fe system occur in the thermal spike phase of the cascade as a consequence of the positive heat of mixing. Further mixing does occur in the thermal spike in the Cu/Fe system and they remain mixed even at the solid state because of the high cooling rate. In addition, ion irradiation induces a large surface roughening of the Ag and Cu top layers as proven by AFM. This effect is important for the correct interpretation of the results. Furthermore, this recrystallization affects also the interface, producing a rough interface, that appears in the RBS spectra as an atomic ‘diffusion’ at the interface. 相似文献

To determine when the calcium-binding protein parvalbumin appears during development, neurons in the chick Edinger Westphal nucleus were examined for parvalbumin immunoreactivity at a variety of embryonic stages. Parvalbumin immunoreactivity appeared on embryonic day 14 (E14, Hamburger and Hamilton stage 40) in predominantly lateral Edinger Westphal neurons. Cytochrome oxidase activity within the nucleus was examined throughout development, as an indicator of physiological activity, and expression of cytochrome oxidase was compared with that of parvalbumin. Cytochrome oxidase activity was found to be uniformly high in all parts of the Edinger Westphal nucleus throughout development. Either the Edinger Westphal nucleus in physiologically active quite early in its development or other energy demands mask the correlation of cytochrome oxidase with electrical activity. Cytochrome oxidase was expressed well before parvalbumin immunoreactivity appeared. Voltage-activated calcium currents were characterized in E12 Edinger Westphal neurons. In both amplitude and composition, E12 calcium currents resemble those of E16 neurons, excluding the possibility that calcium currents appear de novo during or just prior to the appearance of parvalbumin. Both cytochrome oxidase activity and calcium currents are observed in Edinger Westphal neurons well before the appearance of parvalbumin during development. These findings do not exclude the possibility that physiological activity affects the expression of parvalbumin since other factors such as changing patterns of synaptic activity or the appearance of calcium conducting NMDA receptors have yet to be examined. However, they raise the possibility that additional factors such as an intrinsic developmental program or a change in the neuron's basal intracellular calcium requirements may also be involved. 相似文献

We present a novel CWDM metropolitan multiple-access ring network based on optical switching of packets according to their
wavelength. Each node within the MAN is identified by a combination of wavelength and numerical address. Hence, nodes are
able to drop packets presenting a particular wavelength and numerical address, but can insert packets in any wavelength into
the ring. This configuration allows wavelength sharing, as several nodes are identified by the same wavelength (but different
numerical addresses), and simplifies switching requirements since the set of numerical addresses is reduced. We analyze the
viability and scalability of such a network, determining the number of nodes supported by the network under different traffic
scenarios and wavelength resources. The impact of switching time on network performance is also analyzed in order to determine
which switching technology should be employed when implementing the network.

Experiment 1 used Pavlovian conditioning procedures to show that rats formed distinct memorial representations of 2 (peanut oil and sucrose pellets) unconditioned stimuli (USs) that could be activated by 2 different conditioned stimuli (CSs). After training in Experiment 2, rats injected with the lipid antimetabolite Na-2-mercaptoacetate (MA) responded more to the CS for oil than to the CS for sucrose. This pattern was not shown by rats that received isotonic saline or systemic 2-deoxy-d-glucose (a glucose antimetabolite). By contrast, intracerebroventricular infusion of the glucose antimetabolite 5-thioglucose selectively promoted responding to the CS for sucrose (Experiment 4). Thus, lipoprivic and glucoprivic treatments selectively promoted the activation of the memories of fat and carbohydrate USs, respectively. In Experiment 3, the capacity of MA to augment responding to a CS for oil was abolished for rats that received subdiaphragmatic vagal deafferentation. This indicates that the capacity of lipoprivic signals to selectively activate the representations of fat USs may depend on vagal afferent fibers. Multicultural training at internship sites is a critical component in the preparation of doctoral-level psychologists, so the quality of this training is of the utmost importance. In the following study, the authors examine multicultural training from the perspective of predoctoral interns and training directors at counseling center sites that offer a major or minor rotation in multicultural therapy. Results suggest that perspectives vary between interns and training directors and that there is a great difference in the type of criteria used by each site as evidence of a major or minor rotation. The need for more standardized criteria to define major and minor rotations and suggestions for the Association of Psychology Postdoctoral and Internship Centers, internship training directors, and prospective interns are discussed. In this paper we develop and test a model of the associations between major project managers' personal attributes and project success in the context of the Australian Defence industry. In our model, emotional intelligence, cognitive flexibility and systemic thinking were hypothesised to relate to project success, mediated by internal and external stakeholder relationships. The model was tested in an online survey with 373 major project managers. Emotional intelligence and cognitive flexibility were found to be related to the development, quality and effectiveness of major project managers' relationships with both internal and external stakeholders; and these in turn were associated with their ratings of project success. Systemic thinking, however, had no relationship with either stakeholder relationships or project success. Additional research is needed to examine the contribution of a wider range of personal attributes to stakeholder relationships and project success, and to assess whether this model is applicable in other industries and types of projects. 相似文献
