Type 2 diabetes mellitus (T2D) is one of the prominent risk factors for the development and progression of calcific aortic valve disease. Nevertheless, little is known about molecular mechanisms of how T2D affects aortic valve (AV) remodeling. In this study, the influence of hyperinsulinemia and hyperglycemia on degenerative processes in valvular tissue is analyzed in intact AV exposed to an either static or dynamic 3D environment, respectively. The complex native dynamic environment of AV is simulated using a software-governed bioreactor system with controlled pulsatile flow. Dynamic cultivation resulted in significantly stronger fibrosis in AV tissue compared to static cultivation, while hyperinsulinemia and hyperglycemia had no impact on fibrosis. The expression of key differentiation markers and proteoglycans were altered by diabetic conditions in an environment-dependent manner. Furthermore, hyperinsulinemia and hyperglycemia affect insulin-signaling pathways. Western blot analysis showed increased phosphorylation level of protein kinase B (AKT) after acute insulin stimulation, which was lost in AV under hyperinsulinemia, indicating acquired insulin resistance of the AV tissue in response to elevated insulin levels. These data underline a complex interplay of diabetic conditions on one hand and biomechanical 3D environment on the other hand that possesses an impact on AV tissue remodeling.  相似文献

The fluorescent modification of proteins (with genetically encoded low-molecular-mass fluorophores, affinity probes, or other chemically active species) is extraordinarily useful for monitoring and controlling protein functions in vitro, as well as in cell cultures and tissues. The large sizes of some fluorescent tags, such as fluorescent proteins, often perturb normal activity and localization of the protein of interest, as well as other effects. Of the many fluorescent-labeling strategies applied to in vitro and in vivo studies, one is very promising. This requires a very short (6- to 12-residue), appropriately spaced, tetracysteine sequence (-CCXXCC-); this is either placed at a protein terminus, within flexible loops, or incorporated into secondary structure elements. Proteins that contain the tetracysteine motif become highly fluorescent upon labeling with a nonluminescent biarsenical probe, and form very stable covalent complexes. We focus on the development, growth, and multiple applications of this protein research methodology, both in vitro and in vivo. Its application is not limited to intact-cell protein visualization; it has tremendous potential in other protein research disciplines, such as protein purification and activity control, electron microscopy imaging of cells or tissue, protein-protein interaction studies, protein stability, and aggregation studies.  相似文献

This paper presents a novel scheme to address the challenge of identifying failing scan cells from production test responses in the presence of scan compression. The scheme is based on a very simple test response compactor employing orthogonal—spatial and time—signatures. The advantage of this scheme as compared to previous work in this field is the simple and incremental nature of the compaction hardware required. The ability of the scheme to accurately identify failing scan cells from compacted responses has been measured on production fail data from five industrial designs and is reported herein.  相似文献

The existence of localized vibrational modes both at the positive and at the negative LiNbO3 (0001) surface is demonstrated by means of first-principles calculations and Raman spectroscopy measurements. First, the phonon modes of the crystal bulk and of the (0001) surface are calculated within the density functional theory. In a second step, the Raman spectra of LiNbO(3) bulk and of the two surfaces are measured. The phonon modes localized at the two surfaces are found to be substantially different, and are also found to differ from the bulk modes. The calculated and measured frequencies are in agreement within the error of the method. Raman spectroscopy is shown to be sensitive to differences between bulk and surface and between positive and negative surface. It represents therefore an alternative method to determine the surface polarity, which does not exploit the pyroelectric or piezoelectric properties of the material.  相似文献

More than a decade after the first report of single-molecule conductance, it remains a challenging goal to prove the exact nature of the transport through single molecules, including the number of transport channels and the origin of these channels from a molecular orbital point of view. We demonstrate for the archetypical organic molecule, benzenedithiol (BDT), incorporated into a mechanically controllable break junction at low temperature, how this information can be deduced from studies of the elastic and inelastic current contributions. We are able to tune the molecular conformation and thus the transport properties by displacing the nanogap electrodes. We observe stable contacts with low conductance in the order of 10(-3) conductance quanta as well as with high conductance values above ~0.5 quanta. Our observations show unambiguously that the conductance of BDT is carried by a single transport channel provided by the same molecular level, which is coupled to the metallic electrodes, through the whole conductance range. This makes BDT particularly interesting for applications as a broad range coherent molecular conductor with tunable conductance.  相似文献

Analysis of a purified fraction from acetone extracts of olive pulp and pomace by electrospray ionisation tandem mass spectrometry (ESI‐MSn) showed the presence of oleuropein oligomers, whose occurrence has not been reported previously in the literature. The main ionic species (m/z 1613) in the ESI‐MS spectrum was an oleuropein trimer containing three linkages through the hydroxytyrosol backbone. In both samples, oleuropein dimers (m/z 1075), trimers comprising two hydroxytyrosol linkages (m/z 1615), tetramers (m/z 2153) and pentamers (m/z 2691) were also detected by MS. The occurrence of oleuropein oligomers was also observed by nuclear magnetic resonance (NMR). 13C, 13C distortionless enhancement by polarisation transfer DEPT 90, 13C DEPT 135, gHSQC (heteronuclear single quantum coherence) and gHMBC (heteronuclear multiple bond coherence) spectra showed all carbon and proton resonances of oleuropein with the exception of the low‐mobility and asymmetric signals of the aromatic rings. Since mature olives were used in this study, it is possible that the disappearance of oleuropein that has been described to occur with the olive fruit maturation, could be associated with the formation of phenolic oligomers together with lower‐molecular‐weight compounds resulting from its degradation. An electrospinning technology have been developed to obtain zinc oxide nanofibres doped with aluminum. Properties of the obtained nanostructures can be controlled by both the composition of a precursor and subsequent annealing treatment. The gas sensors manufactured with the use of ZnO:Al nanofibres exhibit good response to NO2 at relatively low operating temperatures. For some samples it was observed that interaction with ambient NO2 gas causes the change of conductivity from n-type to p-type at higher operating temperatures. This phenomenon was not observed for the samples annealed at higher temperature.  相似文献

Motris, an integrated system for model-based tracking research, has been designed modularly to study the effects of algorithmic variations on tracking results. Motris attempts to avoid introducing bias into the relative assessment of alternative approaches. Such a bias may be caused by differences of implementation and parameterization if the component approaches are evaluated in separate testing environments. Tracking results are evaluated automatically on a significant test sample in order to quantify the effects of different combinations of alternatives. The Motris system environment thus allows an in-depth comparison between the so-called ‘Edge-Element Association’ approach documented in Haag and Nagel (1999) and the more recent ‘Expectation-Maximization’ approach reported by Pece and Worrall (2002).  相似文献

Using pilot-scale test plant, the interfacial area per unit volume was investigated in different gasliquid reactors, i.e. packed column, bubble column and free jet reactor. The interfacial area was studied as a function of liquid viscosity and of operating parameters. As a rule, identical test conditions were maintained in all the measurements, in order to obtain comparable results. The interfacial area was determined by chemical means using the sulphite system (a solution of sodium sulphite in water as model liquid and air as gaseous medium). The viscosity of the solution can be increased by adding carboxymethyl cellulose without significantly affecting the reaction kinetics. The addition of a surfactant to the sulphite system allowed comparative measurements at reduced surface tension. Based on a large number of measurements, the correlations of the interfacial area are expressed as power laws. The inclusion of experiments with a jet tube reactor and a stirring vessel allows an extensive comparison of all reactors. All tests were carried out with the same material system and the same method was used to determine the interfacial area per unit volume. Therefore, a comparison with respect to mass transfer is possible.  相似文献
