Research in the field of neurobiology and neurochemistry has seen a rapid expansion in the last several years due to advances in technologies and instrumentation, facilitating the detection of biomolecules critical to the complex signaling of neurons. Part of this growth has been due to the development and implementation of high-resolution Fourier transform (FT) mass spectrometry (MS), as is offered by FT ion cyclotron resonance (FTICR) and Orbitrap mass analyzers, which improves the accuracy of measurements and helps resolve the complex biological mixtures often analyzed in the nervous system. The coupling of matrix-assisted laser desorption/ionization (MALDI) with high-resolution MS has drastically expanded the information that can be obtained with these complex samples. This review discusses notable technical developments in MALDI-FTICR and MALDI-Orbitrap platforms and their applications toward molecules in the nervous system, including sequence elucidation and profiling with de novo sequencing, analysis of post-translational modifications, in situ analysis, key advances in sample preparation and handling, quantitation, and imaging. Notable novel applications are also discussed to highlight key developments critical to advancing our understanding of neurobiology and providing insight into the exciting future of this field. © 2020 John Wiley & Sons Ltd. Bragg gratings used as input-output couplers in polymeric waveguides have been demonstrated at infrared wavelengths. These Bragg grating couplers were holographically formed volume phase gratings with a near-45 degrees fringe slant angle embedded directly into a waveguide layer. A photopolymer was used for both producing a planar waveguide and constructing the embedded Bragg grating coupler. A coupling efficiency of 23% for input and 5% for output has been achieved at 850 nm. The output-coupling beam profiles are also discussed. 相似文献

Novel biological vascular conduits, such as decellularized tissue engineered vascular grafts (TEVGs) are hindered by high thrombogenicity. To mimic the antithrombogenic surface of native vessels with a continuous glycosaminoglycan layer that is present on endothelial cells (ECs), a hyaluronic acid (HA) modified surface is established, to effectively shield blood platelets from collagen‐triggered activation. Using the amine groups present on 4 mm diameter decellularized TEVGs, a continuous HA hydrogel coating is built via a bifunctional thiol‐reactive cross‐linker, thereby avoiding nonspecific collagen matrix cross‐linking. The HA hydrogel layer recreates a luminal wall, “hiding” exposed collagen from the bloodstream. In vitro blood tests show that adhered platelets, fibrinogen absorption, and fibrin formation on HA‐coated decellularized TEVGs are significantly lower than on uncoated decellularized TEVGs. The HA surface also inhibits macrophage adhesion in vitro. HA‐coated decellularized syngeneic rat aortae (≈1.5 mm diameter), and TEVGs in rat and canine models, respectively, are protected from aggressive thrombus formation, and preserve normal blood flow. Re‐endothelialization is also observed. HA‐coated TEVGs may be an off‐the‐shelf small‐diameter vascular graft with dual benefits: antithrombogenic protection and promotion of endothelium. 相似文献

This study explored the relationships between eye tracking and traditional usability testing data in the context of analyzing the usability of Algebra Nation?, an online system for learning mathematics used by hundreds of thousands of students. Thirty-five undergraduate students (20 females) completed seven usability tasks in the Algebra Nation? online learning environment. The participants were asked to log in, select an instructor for the instructional video, post a question on the collaborative wall, search for an explanation of a mathematics concept on the wall, find information relating to Karma Points (an incentive for engagement and learning), and watch two instructional videos of varied content difficulty. Participants’ eye movements (fixations and saccades) were simultaneously recorded by an eye tracker. Usability testing software was used to capture all participants’ interactions with the system, task completion time, and task difficulty ratings. Upon finishing the usability tasks, participants completed the System Usability Scale. Important relationships were identified between the eye movement metrics and traditional usability testing metrics such as task difficulty rating and completion time. Eye tracking data were investigated quantitatively using aggregated fixation maps, and qualitative examination was performed on video replay of participants’ fixation behavior. Augmenting the traditional usability testing methods, eye movement analysis provided additional insights regarding revisions to the interface elements associated with these usability tasks. 相似文献

The objective of this work was to develop and demonstrate a probabilistic life prediction method for the prediction of minimum fatigue lives that are typically used in the design of fracture critical rotating turbine engine components. A Monte Carlo analysis was used to predict the variability in fatigue lives based on the distribution of microstructural features that lead to early crack initiation as well as the variability in small fatigue crack growth rates. Two titanium alloys, both with bimodal microstructures, were tested and analysed in this study. The distribution of critical microstructural features was calibrated based on test results and understanding of microstructure neighbourhood effects. Testing was conducted on both alloys and included both smooth and notched specimens. The predictions are presented and compared with the data for smooth and notch geometries for the various loading conditions. A parametric study was performed to identify the importance of several model inputs and to identify areas for future improvement. 相似文献

While traditional approaches for disease management in the era of modern medicine have saved countless lives and enhanced patient well-being, it is clear that there is significant room to improve upon the current status quo. For infectious diseases, the steady rise of antibiotic resistance has resulted in super pathogens that do not respond to most approved drugs. In the field of cancer treatment, the idea of a cure-all silver bullet has long been abandoned. As a result of the challenges facing current treatment and prevention paradigms in the clinic, there is an increasing push for personalized therapeutics, where plans for medical care are established on a patient-by-patient basis. Along these lines, vaccines, both against bacteria and tumors, are a clinical modality that could benefit significantly from personalization. Effective vaccination strategies could help to address many challenging disease conditions, but current vaccines are limited by factors such as a lack of potency and antigenic breadth. Recently, researchers have turned toward the use of biomimetic nanotechnology as a means of addressing these hurdles. Recent progress in the development of biomimetic nanovaccines for antibacterial and anticancer applications is discussed, with an emphasis on their potential for personalized medicine. 相似文献

Lake Erie has experienced dramatic changes in water quality over the past several decades requiring extensive monitoring to assess effectiveness of adaptive management strategies. Remote sensing offers a unique potential to provide synoptic monitoring at daily time scales complementing in-situ sampling activities occurring in Lake Erie. Bio-optical remote sensing algorithms require knowledge about the inherent optical properties (IOPs) of the water for parameterization to produce robust water quality products. This study reports new IOP and apparent optical property (AOP) datasets for western Lake Erie that encapsulate the May–October period for 2015 and 2016 at weekly sampling intervals. Previously reported IOP and AOP observations have been temporally limited and have not assessed statistical differences between IOPs over spatial and temporal gradients. The objective of this study is to assess trends in IOPs over variable spatial and temporal scales. Large spatio-temporal variability in IOPs was observed between 2015 and 2016 likely due to the difference in the extent and duration of mid-summer cyanobacteria blooms. Differences in the seasonal trends of the specific phytoplankton absorption coefficient between 2015 and 2016 suggest differing algal assemblages between the years. Other IOP variables, including chromophoric, dissolved organic matter (CDOM) and beam attenuation spectral slopes, suggest variability is influenced by river discharge and sediment re-suspension. The datasets presented in this study show how these IOPs and AOPs change over a season and between years, and are useful in advancing the applicability and robustness of remote sensing methods to retrieve water quality information in western Lake Erie. 相似文献

Pacific salmon were introduced to the Great Lakes in the 1960s and now support major recreational fishery. Population declines resulting from invasive species have prompted agencies to consider diversifying sport fisheries through stocking. Atlantic salmon are native to Lake Ontario, but a small fishery has developed in northern Lake Huron since the 1990s that appears suited to the Lake Huron food web leading to requests for increased stocking by anglers and consideration by agencies. However, no study has evaluated the trophic ecology of Atlantic salmon in relation to other salmonine predators in northern Lake Huron. In this study, we used stable isotopes of carbon (δ13C) and nitrogen (δ15N), along with mercury (Hg) concentrations to assess resource use, niche overlap, and contaminant accumulation in Atlantic salmon compared to select Lake Huron predators. Atlantic salmon exhibited considerable niche overlap with Chinook and coho salmon but were strongly differentiated from lake trout. In addition, we observed that Atlantic salmon had similar Hg concentrations as coho but were lower than both Chinook salmon and lake trout. Based upon the relationship between fish size, δ15N, and Hg, Atlantic salmon bioaccumulate Hg similarly to Pacific salmon but likely have lower consumptive demands than Chinook salmon. Continued attention should be placed on understanding how Atlantic salmon fit into the current Lake Huron food web in order to evaluate the long-term efficacy of the Atlantic salmon stocking program. 相似文献

The strong hydrogen bonding ability of 2-pyridones were exploited to build nanotrains on surfaces. Carborane wheels on axles
difunctionalized with 2-pyridone hydrogen bonding units were synthesized and displayed spontaneous formation of linear nanotrains
by self-assembly on SiO2 or mica surfaces. Imaging using atomic force microscopy confirmed linear formations with lengths up to 5 μm and heights within
the range of the molecular height of the carborance-tipped axles.
