Selected physico-chemical properties of pea legumin before and after succinylation have been investigated using isoelectric focusing, PAGE, SDS-PAGE, hydrophobicity measurements, SE-HPLC and RP-HPLC. Exhaustive succinylation shifted the I.P. of legumin from 4.75 to 3.5. The stepwise dissociation of legumin by increasing succinylation has been confirmed both by means of PAGE in a nondenaturing system, and by SE-HPLC. The results of SDS-PAGE provided evidence for the exposure of α-polypeptide chains in the native legumin. High succinylation resulted in a decrease of the surface hydrophobicity (S0) measured by both fluorescence probes (cis-parinaric acid and anilino-naphtalene sulfonic acid). RP-HPLC gave a response both to conformational changes and the introduced succinyl residues.  相似文献

The aim was to select potentially probiotic lactobacilli from 88 strains isolated from unpasteurized milk and cheese products, and to incorporate these bacteria in a viable state into a soft cheese, without changing its quality. The survival of these bacteria was assessed in acidic and bile conditions, after freezing at -80 degrees C. Four strains from unpasteurized Camembert--two Lactobacillus plantarum strains and two Lb. paracasei/casei strains--were identified and typed by PCR and PFGE and were found to display potentially probiotic characteristics in addition to resistance to low pH and bile. These characteristics were resistance to lysozyme, adhesion to CACO-2 cells, antimicrobial effects against common foodborne pathogens (Listeria monocytogenes, Staphylococcus aureus, Salmonella spp., Escherichia coli, innocuity following the ingestion of high doses by mice and appropriate antibiotic susceptibility profiles. The potential of Lb. plantarum strain UCMA 3037 for incorporation into a soft cheese (Pont-l'Eveque registered designation of origin (RDO)) was investigated. This strain grew well and survived in sufficient numbers (more than 10(7) cfu/g throughout the shelf-life of the product) in the cheese. This strain did not change the quality score of the product until the best before date (75 days after manufacture). Thus, unpasteurized Camembert is a natural source of potentially probiotic lactobacilli, which could be used as an additive in the development of potentially probiotic soft cheeses. Further work is required to demonstrate the persistence and efficacy of these strains in the human host upon ingestion.  相似文献

We synthesized and characterized a set of undoped and doped cerium oxide nanoparticles with the following chemical formula: Ce1−xMxO2−(x/2) (M: Y or Gd and x = 0 or 0.15) by different coprecipitation protocols at room temperature. Room temperature photoluminescence spectra were similar whatever the protocols and doping. After hydrothermal ripening at neutral pH, a moderate but significant particle growth was observed for doped particles whereas undoped ones remained the same. Simultaneously, the photoluminescence band at 400 nm was significantly red-shifted and practically vanished for doped particles whereas it was only slightly weaker and shifted for pure ceria. All other bands in the same region were much less affected: the bands at 363 and 378 nm became more intense, the shoulder at 420 nm remained roughly unchanged. These phenomena were observed systematically whatever the coprecipitation protocol. As a consequence, the band at 400 nm can be specifically attributed to surface defects that disappear during growth under hydrothermal conditions.  相似文献

A 10-bit 80-MS/s analog-to-digital converter (ADC) with an area- and power-efficient architecture is described. By sharing an amplifier between two successive pipeline stages, a 10-bit pipeline is realized using just four amplifiers with a separate sample-and-hold block. The proposed feedback signal polarity inverting (FSPI) technique addresses the drawback of the conventional amplifier sharing technique. A wide-swing wide-bandwidth telescopic amplifier and an early comparison technique with a constant delay circuit have been developed to further reduce power consumption. The ADC is implemented in a 0.18-/spl mu/m dual-gate-oxidation CMOS process technology, achieves 72.8-dBc spurious free dynamic range, 57.92-dBc signal-to-noise ratio, 9.29 effective number of bits (ENOB) for a 99-MHz input at full sampling rate, and consumes 69 mW from a 3-V supply. The ADC occupies 1.85 mm/sup 2/.  相似文献

Amorphous Ge–Sb–Se thin films were fabricated by a rf‐magnetron co‐sputtering technique employing the following cathodes: GeSe2, Sb2Se3, and Ge28Sb12Se60. The influence of the composition, determined by energy‐dispersive X‐ray spectroscopy, on the optical properties was studied. Optical properties were analyzed based on variable angle spectroscopic ellipsometry and UV‐Vis‐NIR spectrophotometry. The results show that the optical bandgap range 1.35‐2.08 eV with corresponding refractive index ranging from 3.33 to 2.36 can be reliably covered. Furthermore, morphological and topographical properties of selenide‐sputtered films studied by scanning electron microscopy and atomic force microscopy showed a good quality of fabricated films. In addition, structure of the films was controlled using Raman scattering spectroscopy. Finally, irreversible photoinduced changes by means of change in optical bandgap energy and refractive index of co‐sputtered films were studied revealing the photobleaching effect in Ge‐rich films when irradiated by near‐bandgap light under Ar atmosphere. The photobleaching effect tends to decrease with increasing antimony content.  相似文献

Local Mutual Information for Dissimilarity-Based Image Segmentation   总被引:1,自引:0,他引:1  
Connective segmentation based on the definition of a dissimilarity measure on pairs of adjacent pixels is an appealing framework to develop new hierarchical segmentation methods. Usually, the dissimilarity is fully determined by the intensity values of the considered pair of adjacent pixels, so that it is independent of the values of the other image pixels. In this paper, we explore dissimilarity measures depending on the overall image content encapsulated in its local mutual information and show its invariance to information preserving transforms. This is investigated in the framework of the connective segmentation and constrained connectivity paradigms and leads to the concept of dependent connectivities. An efficient probability estimator based on depth functions is proposed to handle multi-dimensional images. Experiments conducted on hyper-spectral and multi-angular remote sensing images highlight the robustness of the proposed approach.  相似文献

The thermodynamic and rheological properties of the Pd40Ni40P20 bulk metallic glass are explored by means of an indentation creep technique around the glass transition. We have developed a dedicated instrumented indentation apparatus allowing to assess the mechanical properties at elevated temperatures. The analysis of results is made possible by using the viscoelastic solutions of contact mechanics. We also analyse the thermodynamics of creep around glass transition to estimate the activation free energy changes from the activation free enthalpy changes via the shear modulus – temperature data. The shear viscosity values extracted using this technique allow for the derivation of activation energies (free enthalpy 210 kJ/mol, enthalpy 456 kJ/mol, entropy 410 J/mol/K) for the flow process. All these properties were found to closely match with those obtained using conventional techniques for viscosity measurements. Compared to the latter, the indentation creep technique requires small volumes and samples are easy to prepare. It is therefore expected that such a technique might be employed for the study of glass transition in metallic glasses.  相似文献

In everyday life, people use a large diversity of hand configurations while reaching out to grasp an object. They tend to vary their hands position/orientation around the object and their fingers placement on its surface according to the object properties such as its weight, shape, friction coefficient and the task they need to accomplish. Taking into account these properties, we propose a method for generating such a variety of good grasps that can be used for the accomplishment of many different tasks. Grasp synthesis is formulated as a single constrained optimization problem, generating grasps that are feasible for the hand’s kinematics by minimizing the norm of the joint torque vector of the hand ensuring grasp stability. Given an object and a kinematic hand model, this method can easily be used to build a library of the corresponding object possible grasps. We show that the approach is adapted to different representations of the object surface and different hand kinematic models.  相似文献

Changes in spatio-temporal distribution of bacterial and denitrifying communities were qualitatively studied in a microbial mat from Camargue (France). During a diel and a seasonal cycle, patterns of 16S rRNA and nitrite reductase genes (nirS and nirK) were compared by denaturing gradient gel electrophoresis (DGGE). Statistical analysis of DGGE profiles showed a significant seasonal shift in the community structure of the nirS-containing bacteria with a winter superficial population that extended in summer, whereas the nirK-containing bacteria seemed more affected by vertical gradients rather than by month-to month-changes. Denitrifying activities remained stable during these sampling times. The bacterial community at the surface of the mat also changed according to season, but appeared stable over a day. Finally, during a diel cycle nirK populations were localized in zones with large fluctuations of environmental parameters (oxygen, pH, and sulfur levels) while nirS populations seemed more restricted to the permanent anoxic layer of the microbial mat.  相似文献
