The effect of Fe addition on the microstructural properties and the corrosion resistance of Al–Zn–Mg alloys submitted to different heat treatments (cast, annealed and aged), has been studied in chloride solutions using optical microscopy (OM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy-dispersive X-ray (EDX), cyclic polarization (CP) and open circuit potential (o.c.p.) measurements. The presence of 0.3% Fe in the alloy limited the growth of the MgZn2 precipitates, both in the annealed and in the quenched specimens. No effect of Cr on the grain size in the presence of Fe was found because of the accumulation of Cr in the Fe-rich particles. Fe in the Al–Zn–Mg alloys also made them more susceptible to pitting. Pitting occurred mainly near the Fe-rich particles both, under o.c.p. conditions in O2-saturated solutions and during the CP.  相似文献

Gait usually presents an excellent improvement after total knee replacement. Nevertheless, some abnormalities persist even after a long period of time. The abnormal knee patterns have been attributed to several possible causes, such as implant geometry and surgical technique, posterior cruciate ligament sparing/sacrificing, preoperative "stiff-knee" pattern due to pain and altered biomechanics, weakness of the extensor muscles, preoperative arthritic pattern, proprioceptive deficiency, and multijoint degenerative involvement. Cocontraction of the knee flexors and extensors is a common strategy adopted to reduce strain and shear forces at the joint, but it increases compressive forces and joint loading. Even in patients with an excellent functional score, the duration of the implant may be compromised by an altered neuromuscular control of the knee. In this paper, we report a single case study carried out over two years on a patient that underwent total knee replacement. The aim of this work is to show that quantitative gait analysis is essential to augment the understanding of the mechanisms underlying gait, thus enabling clinicians to adapt the rehabilitation program to the specific patient. Although the limits of single case reports are obvious, we believe that this evaluation methodology could be beneficial for assessing the effectiveness of rehabilitation programs aimed at achieving an active control of the knee during gait through a correct muscular activation pattern.  相似文献

Branched nanoparticles are one of the most promising nanoparticle catalysts as their branch sizes and surfaces can be tuned to enable both high activity and stability. Understanding how the crystallinity and surface facets of branched nanoparticles affect their catalytic performance is vital for further catalyst development. In this work, a synthesis is developed to form highly branched ruthenium (Ru) nanoparticles with control of crystallinity. It is shown that faceted Ru branched nanoparticles have improved stability and activity in the oxygen evolution reaction (OER) compared with polycrystalline Ru nanoparticles. This work achieves a low 180 mV overpotential at 10 mA cm?2 for hours, demonstrating that record‐high stability for Ru nanocrystals can be achieved while retaining high activity for OER. The superior electrocatalytic performance of faceted Ru branched nanoparticles is ascribed to the lower Ru dissolution rate under OER conditions due to low‐index facets on the branch surfaces.  相似文献

Aspartic protease (PR) of HIV-1 virus represents a valid therapeutic target for the design of antiviral agents suitable for treatment of AIDS. We have designed peptidomimetic PR inhibitors containing a novel dihydroxyethylenediamine -Phe-Psi[CHOH-CHOH]-Pro- core using molecular modelling approach that predicts the inhibitory potencies (IC(50)(pre)) in terms of computed relative enzyme-inhibitor complexation Gibbs free energies (Delta Delta G(comp)). The modelling approach considers not only the enzyme-inhibitor interactions, but includes also the solvent and entropic effects affecting the enzyme inhibition. The objectives of this study were to optimize the number and type of flanking residues that occupy the S(3), S(2) and S(2'), S(3') positions in the PR binding pocket and to select potent lead candidates, which display also favourable ADME-related properties. The structure-based design was combined with a synthetic strategy used to prepare a training set of 10 analogues sharing the -Phe Psi Pro- core. This strategy couples stereochemical control with full flexibility in the choice of the flanking residues and in vitro activity assays. A QSAR model correlating calculated Delta Delta G(comp) with the measured IC(50)(exp) values for the training set was prepared and confirmed that our computational approach can serve for reliable prediction of PR inhibitory potencies of peptidomimetics. The appropriate choice of the flanking residues allowed us to design virtual lead compounds, such as FP14, FP23 and FP76, with reduced molecular weight, predicted inhibitory potencies in the picomolar range, promising ADME profiles and a potential to escape drug resistance due to favourable interactions with the PR backbone.  相似文献

Activated Sludge Models are widely used for simulation-based evaluation of wastewater treatment plant (WWTP) performance. However, due to the high workload and cost of a measuring campaign on a full-scale WWTP, many simulation studies suffer from lack of sufficiently long influent flow rate and concentration time series representing realistic wastewater influent dynamics. In this paper, a simple phenomenological modelling approach is proposed as an alternative to generate dynamic influent pollutant disturbance scenarios. The presented set of models is constructed following the principles of parsimony (limiting the number of parameters as much as possible), transparency (using parameters with physical meaning where possible) and flexibility (easily extendable to other applications where long dynamic influent time series are needed). The proposed approach is sub-divided in four main model blocks: 1) model block for flow rate generation, 2) model block for pollutants generation (carbon, nitrogen and phosphorus), 3) model block for temperature generation and 4) model block for transport of water and pollutants. The paper is illustrated with the results obtained during the development of the dynamic influent of the Benchmark Simulation Model no. 2 (BSM2). The series of simulations show that it is possible to generate a dry weather influent describing diurnal flow rate dynamics (low rate at night, high rate during day time), weekend effects (with different flow rate during weekends, compared to weekdays), holiday effects (where the wastewater production is assumed to be different for a number of weeks) and seasonal effects (with variations in the infiltration and thus also the flow rate to the WWTP). In addition, the dry weather model can be extended with a rain and storm weather generator, where the proposed phenomenological model can also mimic the “first flush” effect from the sewer network and the influent dilution phenomena that are typically observed at full-scale WWTPs following a rain event. Finally, the extension of the sewer system can be incorporated in the influent dynamics as well: the larger the simulated sewer network, the smoother the simulated diurnal flow rate and concentration variations. In the discussion, it is pointed out how the proposed phenomenological models can be expanded to other applications, for example to represent heavy metal or organic micro-pollutant loads entering the treatment plant.  相似文献

Poly(vinylidene fluoride) (PVDF) samples, obtained by casting from tetrahydrofuran solutions and submitted to various thermal treatments, have been examined by Fourier transform–infrared microspectroscopy (FTIR-M) and differential scanning calorimetry (DSC). This kind of analysis allowed us to examine microdomains of samples with different morphological characteristics and to obtain an indication of the polymorphism of PVDF. In some cases the simultaneous presence of two or three forms has been evidenced thanks to the comparison of FTIR-M spectra and DSC traces. Vibrational spectra of single crystalline forms can be recorded by FTIR-M on phase homogeneous microdomains.  相似文献

WC–12Co nanopowders have been densified by means of an innovative apparatus belonging to the Field Assisted Sintering Techniques (FAST), named Pressure Assisted Fast Electric Sintering (PAFES). The technique allows to control the electrical parameters in terms of waveforms and frequencies of the applied currents. By means of AC currents different sintering cycles for the consolidation of cemented carbides have been investigated and optimised. The effect of the different sintering cycles on the microstructure and mechanical properties of the final bulks has been analysed. Fully dense WC–12Co has been obtained, characterised by ultrafine grained microstructure and very high hardness combined with good fracture toughness values.  相似文献

HP1 integrase promotes site-specific recombination of the HP1 genome into that of Haemophilus influenzae. The isolated C-terminal domain (residues 165-337) of the protein interacts with the recombination site and contains the four catalytic residues conserved in the integrase family. This domain represents a novel fold consisting principally of well-packed alpha helices, a surface beta sheet, and an ordered 17-residue C-terminal tail. The conserved triad of basic residues and the active-site tyrosine are contributed by a single monomer and occupy fixed positions in a defined active-site cleft. Dimers are formed by mutual interactions of the tail of one monomer with an adjacent monomer; this orients active-site clefts antiparallel to each other.  相似文献

Recent evidence indicates the presence of p21 Ras and of a protein with characteristics similar to mitogen-activated protein kinases (MAPKs), also known as extracellular signal-regulated kinases (ERKs), in mammalian spermatozoa, suggesting the occurrence of the Ras/ERK cascade in these cells. In the present study we investigated the subcellular localization of ERKs and their biological functions in human spermatozoa. Immunohistochemistry, immunofluorescence, confocal microscopy, and immunoelectron microscopy demonstrated localization of ERKs in the postacrosomal region of spermatozoa. After stimulation of acrosome reaction with the calcium ionophore A23187 and progesterone, ERKs were mostly localized at the level of the equatorial region, indicating redistribution of these proteins in acrosome-reacted spermatozoa. Two proteins of 42 and 44 kDa that are tyrosine phosphorylated in a time-dependent manner during in vitro capacitation were identified as p42 (ERK-2) and p44 (ERK-1) by means of specific antibodies. The increase in tyrosine phosphorylation of these proteins during capacitation was accompanied by increased kinase activity, as determined by the ability of ERK-1 and ERK-2 to phosphorylate the substrate myelin basic protein. The role of this activity in the occurrence of sperm capacitation was also investigated by using PD098059, an inhibitor of the MAPK cascade. The presence of this compound during in vitro capacitation inhibits ERK activation and significantly reduces the ability of spermatozoa to undergo the acrosome reaction in response to progesterone. Since only capacitated spermatozoa are able to respond to progesterone, these data strongly indicate that ERKs are involved in the regulation of capacitation. In summary, our data demonstrate the presence of functional ERKs in human spermatozoa and indicate that these enzymes are involved in activation of these cells during capacitation, providing new insight in clarifying the molecular mechanisms and the signal transduction pathways of this process.  相似文献
