ADP-ribosyl cyclase is a multi-functional enzyme that catalyzes the formation of two Ca2+ signaling molecules, cyclic ADP-ribose (cADPR) and nicotinic acid adenine dinucleotide phosphate (NAADP). X-ray crystallography of three different crystal forms shows that it is a non-covalent dimer. Chemical cross-linking and dynamic light scattering were used in this study to determine if the cyclase is also a non-covalent dimer in solution. Treatment of the cyclase in dilute solution (0.05 mg/ml) with dimethylsuberimidate resulted in complete conversion to a species with molecular weight about twice that of the monomeric cyclase. Prolonged cross-linking of the cyclase at four times higher concentration produced also only the covalently linked dimers and no multimer formation was observed. The cross-linked dimer retained full enzymatic activity and readily catalyzed the formation of cADPR from NAD, NAADP from NADP, cyclic ADP-ribose phosphate from NADP, and cyclic GDP-ribose from nicotinamide guanine dinucleotide. Analysis of the autocorrelation functions obtained from dynamic light scattering measurements indicated the cyclase solution (2 mg/ml) was composed of a single molecular species and its diffusion coefficient was measured to be 7. 4x10-7 cm2/s. Computer modeling using the crystallographic dimensions of the non-covalent cyclase dimer, a donut shaped molecule with a central cavity and overall dimensions of 7x6x3 nm, gave a value for the diffusion coefficient essentially the same as that measured. These results indicate the cyclase is a non-covalent dimer in solution.  相似文献

Several acoustic transmission and reflection technique measurements were carried out to determine mechanical properties (acoustic attenuation and velocity) versus frequency of polyvinylidene-fluoride (PVDF) and six other polymers. Acoustic measurements (0.5 to 12 MHz) included time-delay spectrometry (TDS; in which separate transmitting and receiving transducers utilize a swept frequency signal) and two pulse-echo methods (short tone burst echoes utilizing transducers with different center frequencies and Fourier analysis of echoes sent and received by damped transducers operating in the broadband pulse mode). Electrical impedance measurements of piezoelectric thin films of PVDF and P(VDF/TrFE) yielded comparable high frequency mechanical parameters. Of the seven acoustically examined polymers, PVDF had the greatest acoustic impedance, lowest acoustic velocity, and greatest mechanical loss (13.4 dB/cm per MHz). Polymethyl-methacrylate (PMMA; lucite) and polydimethyl-pentane (TPX) had the lowest loss. PMMA had the highest acoustic velocity, and TPX had the lowest acoustic impedance and a velocity almost identical to that of PVDF. These data are useful in the design of backing, matching, and lens materials to be used in association with PVDF transducers.  相似文献

Recently, transmission Raman spectroscopy has been shown to be a valuable tool in the volumetric quantification of pharmaceutical formulations. In this work a Monte Carlo simulation and experimental study are performed to elucidate the dependence of the Raman signal on depth from the viewpoint of probing pharmaceutical tablets and powders in this experimental configuration. The transmission Raman signal is shown to exhibit a moderate bias toward the center of the tablets and this can be considerably reduced by using a recently developed Raman signal-enhancing concept, the "photon diode." The enhancing element not only reduces the bias but also increases the overall Raman signal intensity and consequently improves the signal-to-noise ratio of the measured spectrum. Overall, its implementation with appropriately chosen reflectivity results in a more uniform volumetric sampling across the half of the tablet where the photon diode is used (or across the tablet's entire depth if two photon diodes are used on each side of tablet) and enhanced overall sensitivity. These findings are substantiated experimentally on a segmented tablet by inserting a poly(ethelyne terephthalate) (PET) film doped with TiO(2) at different depths and monitoring its contribution to the overall transmission Raman signal from the segmented tablet. The numerical simulations also indicate considerable sensitivity of the overall Raman signal to the absorption of the sample, which is in line with large migration distances traversed by photons in these measurements. The presence of sample absorption was shown numerically to reduce the signal enhancement effect while the overall depth-dependence profile remained broadly unchanged. The absorption was also shown to produce a depth profile with the photon diode similar to that without it, although with a reduced absolute intensity of Raman signals and diminished enhancement effect.  相似文献

The acoustic frequency ranges in birdsongs provide important absolute pitch cues for the recognition of conspecifics. Black-capped chickadees (Poecile atricapillus), mountain chickadees (Poecile gambeli), and zebra finches (Taeniopygia guttata) were trained to sort tones contiguous in frequency into 8 ranges on the basis of associations between response to the tones in each range and reward. All 3 species acquired accurate frequency-range discriminations, but zebra finches acquired the discrimination in fewer trials and to a higher standard than black-capped or mountain chickadees, which did not differ appreciably in the discrimination. Chickadees' relatively poorer accuracy was traced to poorer discrimination of tones in the higher frequency ranges. During transfer tests, the discrimination generalized to novel tones when the training tones were included, but not when they were omitted.
