The metalloprotease ADAM17 is a key regulator of the TNFα, IL-6R and EGFR signaling pathways. The maturation and function of ADAM17 is controlled by the seven-membrane-spanning proteins iRhoms1 and 2. The functional properties of the ADAM17/iRhom1 and ADAM17/iRhom2 complexes differ, in that stimulated shedding of most ADAM17 substrates tested to date can be supported by iRhom2, whereas iRhom1 can only support stimulated shedding of very few ADAM17 substrates, such as TGFα. The first transmembrane domain (TMD1) of iRhom2 and the sole TMD of ADAM17 are important for the stimulated shedding of ADAM17 substrates by iRhom2. However, little is currently known about how the iRhoms interact with different substrates to control their stimulated shedding by ADAM17. To provide new insights into this topic, we tested how various chimeras between iRhom1 and iRhom2 affect the stimulated processing of the EGFR-ligands TGFα (iRhom1- or 2-dependent) and EREG (iRhom2-selective) by ADAM17. This uncovered an important role for the TMD7 of the iRhoms in determining their substrate selectivity. Computational methods utilized to characterize the iRhom1/2/substrate interactions suggest that the substrate selectivity is determined, at least in part, by a distinct accessibility of the substrate cleavage site to stimulated ADAM17. These studies not only provide new insights into why the substrate selectivity of stimulated iRhom2/ADAM17 differs from that of iRhom1/ADAM17, but also suggest new approaches for targeting the release of specific ADAM17 substrates.  相似文献

During 2001-2005, 210 Salmonella enterica strains were isolated from seafood samples imported into US. Strains of S. enterica serovar Weltevreden were the most predominantly found among the 64 different serovars isolated. A total of 37 Salmonella Weltevreden isolates were characterized by pulsed-field gel electrophoresis (PFGE), plasmid profiles and antibiotic susceptibility to assess genetic diversity. Our results showed a low frequency of antibiotic resistance; 35 of the 37 isolates were sensitive to ampicillin, tetracycline, chloramphenicol, gentamicin, sulfisoxazole, streptomycin and kanamycin. Only two isolates, from samples originating in the Philippines and India, showed resistance to ampicillin and tetracycline and to streptomycin, sulfisoxazole and tetracycline, respectively. Of the 37 isolates, two isolates did not carry any plasmid and 35 isolates harbored several small and mega-plasmids. These isolates were differentiated into 10 distinct types based on plasmid profiles. Four different PFGE clusters were obtained with a genetic similarity of 66-76%. Four groups of isolates (formed by two or three isolates each) showed 100% similarity in the PFGE profiles. One of these groups included strains isolated in Vietnam in 2003, 2004 and 2005 from fish and shrimp. The other groups included strains isolated in Vietnam, Indonesia and Thailand in 2000, 2004 and 2005 from snail, shrimp and fish. Our findings show genetic diversity and temporal persistence of S. enterica serovar Weltevreden in recently monitored seafood imports.  相似文献

Lead (Pb) and lead–bismuth eutectic (44Pb–56Bi) have been the two primary candidate liquid metal target materials for the production of spallation neutrons. Selection of a container material for the liquid metal target will greatly affect the lifetime and safety of the target subsystem. For the liquid lead target, niobium–1 wt% zirconium (Nb–1Zr) is a candidate containment material for liquid lead, but its poor oxidation resistance has been a major concern. In this paper, the oxidation rate of Nb–1Zr was studied based on the calculations of thickness loss resulting from oxidation. According to these calculations, it appeared that uncoated Nb–1Zr may be used for a 1-year operation at 900°C at PO2=1×10–6 Torr, but the same material may not be used in argon with 5-ppm oxygen. Coating technologies to reduce the oxidation of Nb–1Zr are reviewed, as are other candidate refractory metals such as molybdenum, tantalum, and tungsten. For the liquid lead–bismuth eutectic target, three candidate containment materials are suggested, based on a literature survey of the materials’ compatibility and proton irradiation tests: Croloy 2-1/4, modified 9Cr–1Mo, and 12Cr–1Mo (HT-9) steel. These materials seem to be used only if the lead–bismuth is thoroughly deoxidized and treated with zirconium and magnesium.  相似文献
