Polyethersulphone (PES) is an aromatic thermoplastic, at low environmental impact, evaluated in this work as a promising candidate for new polymer electrolytes in the PEMFCs technology. A sulfonation procedure has been tuned in order to graft sulfonic acid groups on the polymer chains (sPES) and to make it hydrophilic. Homogeneous membranes with different polymer's sulfonation degrees (SD%) have demonstrated excellent mechanical properties and very low permeability toward methanol (important in the DMFCs), even if low proton conductivity. Nanocomposite sPES membranes were prepared by dispersion of highly hydrophilic lamellar particles such as layered double hydroxide (LDH) in the polymer. Deep investigations performed by a combination of PFG-NMR, EIS, XRD, DMA, and scanning electron microscopy have evidenced the exfoliation of the lamellae in polymer matrix. However, a certain anisotropy was evidenced both in the morphology and molecular diffusion, favored in the longitudinal direction (parallel to surface), while completely inhibited in the cross-section. The three GxxxG repeating motifs from the C-terminal region of β-amyloid (Aβ) peptide play a significant role in regulating the aggregation kinetics of the peptide. Mutation of these glycine residues to leucine greatly accelerates the fibrillation process but generates a varied toxicity profile. Using an array of biophysical techniques, we demonstrated the uniqueness of the composite glycine residues in these structural repeats. We used solvent relaxation NMR spectroscopy to investigate the role played by the surrounding water molecules in determining the corresponding aggregation pathway. Notably, the conformational changes induced by Gly33 and Gly37 mutations result in significantly decreased toxicity in a neuronal cell line. Our results indicate that G33xxxG37 is the primary motif responsible for Aβ neurotoxicity, hence providing a direct structure–function correlation. Targeting this motif, therefore, can be a promising strategy to prevent neuronal cell death associated with Alzheimer's and other related diseases, such as type II diabetes and Parkinson's. 相似文献

Fining experiments have been conducted on fino sherry wine from the Jerez-Xérès-Sherry region (southern Spain), in which the combined use of activated charcoal with proteinaceous fining agents (casein, potassium caseinate, albumin, and gelatin) has been studied. The effect of these fining agents on the polyphenolic content, the aromatic profile, and the resistance to browning of the treated wine has been determined. The polyphenolic content suffers significant decreases following the use of activated charcoal; these decreases are only increased slightly by the subsequent use of the other fining agents. The aromatic profile was not found to be altered by the clarification agents used. Despite the reduction in the polyphenolic content, the treated wines show a tendency to suffer browning similar to that observed in non-clarified wine. 相似文献

In this research I investigated whether the use of relevant affective outcomes influences depressed and nondepressed subjects' judgment of contingency. Similar to previous studies (Alloy & Abramson, 1979, Experiments 1 and 2), Experiments 1 and 2 confirmed that when the outcome is affectively neutral (i.e., the onset of a light) depressed subjects make accurate judgments of contingency, whereas nondepressed subjects show (in noncontingent situations) a significant illusion of control. In Experiments 3 and 4 (a contingency situation and a noncontingency situation, respectively) different types of sentences (negative self-referent, negative other-referent, positive self-referent, positive other-referent) were used as outcomes. Although depressed subjects were more reluctant to show biased judgments than were the nondepressed subjects, in noncontingency situations depressed subjects made overestimated judgments of contingency when the outcomes were negative self-referent sentences. The present work proposes the use of a fast analytical platform for the mass spectrometric (MS) profiling of canine mammary tissues in their native form for the building of a predictive statistical model. The latter could be used as a novel diagnostic tool for the real-time identification of different cellular alterations in order to improve tissue resection during veterinary surgery, as previously validated in human oncology. Specifically, Rapid Evaporative Ionization Mass Spectrometry (REIMS) coupled with surgical electrocautery (intelligent knife—iKnife) was used to collect MS data from histologically processed mammary samples, classified into healthy, hyperplastic/dysplastic, mastitis and tumors. Differences in the lipid composition enabled tissue discrimination with an accuracy greater than 90%. The recognition capability of REIMS was tested on unknown mammary samples, and all of them were correctly identified with a correctness score of 98–100%. Triglyceride identification was increased in healthy mammary tissues, while the abundance of phospholipids was observed in altered tissues, reflecting morpho-functional changes in cell membranes, and oxidized species were also tentatively identified as discriminant features. The obtained lipidomic profiles represented unique fingerprints of the samples, suggesting that the iKnife technique is capable of differentiating mammary tissues following chemical changes in cellular metabolism. 相似文献

We propose a new organ-conditioning strategy based on mesenchymal stromal cell (MSCs)/extracellular vesicle (EVs) delivery during hypothermic perfusion. MSCs/EVs marker CD73 is present on renal proximal tubular cells, and it protects against renal ischemia-reperfusion injury by converting adenosine monophosphate into adenosine (ADO). In this study, after checking if CD73-silenced EVs (EVsi) would impact in vitro tubular-cell proliferation, we perfused kidneys of a rat model of donation after circulatory death, with Belzer solution (BS) alone, BS supplemented with MSCs, EVs, or EVsi. The ADO and ATP levels were measured in the effluents and tissues. Global renal ischemic damage score (GRS), and tubular cell proliferation index (IPT) were evaluated in the tissue. EVsi did not induce cell proliferation in vitro. Ex vivo kidneys perfused with BS or BS + EVsi showed the worst GRS and higher effluent ADO levels than the MSC- and EV-perfused kidneys. In the EV-perfused kidneys, the tissue and effluent ATP levels and IPT were the highest, but not if CD73 was silenced. Tissue ATP content was positively correlated with tissue ADO content and negatively correlated with effluent ADO level in all groups. Despite of the development of advanced segmentation techniques, achieving accurate and reproducible gross tumor volume (GTV) segmentation results is still an important challenge in neuro-radiosurgery. Nowadays, magnetic resonance imaging (MRI) is the most prominent modality in radiation therapy for soft-tissue anatomical districts. Gamma Knife stereotactic neuro-radiosurgery is a minimally invasive technology for dealing with inaccessible or insufficiently treated tumors with traditional surgery or radiotherapy. During a treatment planning phase, the GTV is generally contoured by experienced neurosurgeons and radiation oncologists using fully manual segmentation procedures on MR images. Unfortunately, this operative methodology is definitely time-expensive and operator-dependent. Delineation result repeatability, in terms of both intra- and inter-operator reliability, can be achieved only by using computer-assisted approaches. In this paper a novel semi-automatic seeded image segmentation method, based on a cellular automata model, for MRI brain cancer detection and delineation is proposed. This approach, called GTVcut, employs an adaptive seed selection strategy and helps to segment the GTV, by identifying the target volume to be treated using the Gamma Knife device. The accuracy of GTVcut was evaluated on a dataset composed of 32 brain cancers, using both spatial overlap-based and distance-based metrics. The achieved experimental results are very reproducible, showing the effectiveness and the clinical feasibility of the proposed approach. 相似文献
