Summary Variations in low-molecular-weight phenolic compounds in different parts (must, skin, and seeds) of berries ofV. vinifera, variety Cencibel during ripening were studied using HPLC andTLC. Twenty-five different compounds were identified. The differences were chiefly quantitative, although some quantitative differences were also recorded.

The influence of spatial attention on lexical decisions to lateralized target letter-strings appearing either along with a distractor (Experiment 1) or in an otherwise empty field (Experiments 2–6) was examined. Attentional orienting was controlled by peripheral (Experiments 1, 2, 3, and 6) and central (Experiments 4–5) cuing methods. Manipulations of spatial attention, including cue validity and cue–target stimulus onset asynchrony, were combined with manipulations of word frequency in Experiments 3-6. All the attentional manipulations were effective, but they did not modify the right visual field advantage in word performance, In addition, the attentional effects did not interact with either the presence or absence of distractors or with stimulus familiarity. Implications of these results regarding the influence of spatial attention (the posterior attention system) on word processing are discussed. The aging of the population is giving rise to a large number of studies aimed at improving the quality of life, considering diet as a major contributing factor in this life stage. The objectives of the present study were to evaluate the nutritional status of a group of institutionalized elderly in Murcia, Spain, and to relate the diet to anthropometric and biochemical indices, paying special attention to total antioxidant status. The dietary evaluation showed excessive intakes of proteins and lipids, and deficiencies in those of carbohydrates, dietary fiber, zinc, iodine and vitamins A, E and D. The high proportion of overweight observed were not significantly correlated to the energy intakes. Although total plasma antioxidant status was low (0.62 mM eq Trolox and 0.98 mM eq Fe(II)) compared with reference values, estimated oxidative risk in this group of elderly was low according to plasma levels of vitamins C, A & E. 相似文献

The occurrence and evolution of Enterobacteriaceae, coliforms, Escherichia coli, and enterococci in 252 samples of six types of Mediterranean dry-ripened sausages during maturation was investigated. The changes of the pH values were also recorded. Samples were analyzed in three steps of the ripening process: fresh product, first drying stage, and finished product. In all six sausages, Enterobacteriaceae, coliforms, and E. coli counts were characterized by high initial concentrations that significantly decreased during ripening. However, the initial counts of enterococci remained stable throughout the experiments in all sausages types. The average pH values of finished sausages ranged from 4.54 to 5.31. 相似文献

We investigate the use of laser reflectometry near the critical angle to monitor particle adsorption onto a flat glass surface. Experimental results show that positive particles are adsorbed onto the glass surface and that their adsorption kinetics depend strongly on the volume fraction occupied by the particles in suspension but not appreciably on the particle size. The reflectance near the critical angle is dominated by the particles on the surface, with the contribution of the particles in suspension being very low. We compare the reflectance change near the critical angle with the change in reflectance near the Brewster angle when particles are adsorbed onto the glass surface. We find that reflectometry near the critical angle is 3000 times more sensitive than it is near the Brewster angle. Some optical images are presented to validate our results. 相似文献

Effective preharvest strategies to eliminate aflatoxin accumulation in crops are not presently available. The molecular biology of aflatoxin biosynthesis has been extensively studied, and genetic and molecular tools such as reporter gene systems for the measurement of fungal growth have been developed. A reporter construct containing the Aspergillus flavus beta-tubulin gene promoter fused to Escherichia coli beta-glucuronidase (GUS) has been shown to be a reliable tool for the indirect measurement of fungal growth in maize kernels. Since cost-saving alternative methods for the direct measurement of aflatoxin levels are needed to facilitate more widespread field and laboratory screening of maize lines, a new reporter gene construct involving the promoter region of the omtA gene of the aflatoxin biosynthetic pathway was constructed and tested. Expression of GUS activity by this construct (omtA::GUS) was correlated with aflatoxin accumulation in culture. In the fungal transformant GAP26-1, which harbors this construct, aflatoxin production and GUS expression on sucrose-containing medium showed the same temporal pattern of toxin induction. Furthermore, GUS expression by GAP26-1 was shown to be associated with aflatoxin accumulation in maize kernels inoculated with this strain. Our results suggest that this and other reporter gene pathway promoter constructs may provide superior alternatives to direct aflatoxin quantification with respect to time, labor, and materials for the screening of maize lines for resistance to aflatoxin accumulation. 相似文献

The presence of drug residues in ewe's milk samples can be determined by microbial assays. The main limitation of these tests is the large number of false-positive results associated with them. False-positive results can be explained by the interaction of certain substances naturally existing in ewe's milk with the growth of the microorganism used in the test. In this study, milk chemical composition (fat, protein, lactose, total solids), somatic cell counts (SCCs), free fatty acid concentrations, and lactoperoxidase system components were determined in order to investigate their influence on the rate of false-positive results for the BRT and Delvotest microbiological inhibitor tests. Milk samples were obtained after morning milking of Manchega ewes at 15, 30, 45, 60, 75, 90, 105, 120, and 135 days after parturition. The animals did not receive any kind of treatment or medicated feed throughout the experiment. The false-positive rates for BRT and Delvotest were 3.75 and 2.4%, respectively. When the logistic regression model was applied, the percentages of total solids for positive samples were significantly different from those for negative samples (16.90 versus 18.42% for BRT, 16.05 versus 18.45% for Delvotest), while the SCC logarithmic transformation was significantly higher for the positive samples than for the negative samples (5.38 versus 5.11 log units for BRT, 5.32 versus 5.11 log units for Delvotest). Moreover, Delvotest-positive samples exhibited thiocyanate concentrations higher than those of Delvotest-negative samples (8.18 mg/liter versus 6.85 mg/liter). Further analyses are needed to confirm the possible presence of antimicrobial residues in this particular type of milk sample. 相似文献

A new microbiological predictive model was developed, relating pH, NaCl concentration and temperature (T) with the decimal reduction coefficient D as dependent variable, using data previously reported by [1]. The model was obtained by multiple linear regression using only three predictor variables (T 2, pH2 and NaCl), easily computed from the basic variables pH, NaCl concentration and temperature (T). The fitted model is robust, stable and satisfies all the basic hypotheses of the regression models. This is the first study to analyze variations in time estimation during 60 h of sleep deprivation and the relation between time estimation performance and the activation measures of skin resistance level, body temperature, and Stanford Sleepiness Scale (SSS) scores. Among 30 healthy participants 18 to 24 years of age, for a 10-s interval using the production method, we found a lengthening in time estimations that was modulated by circadian oscillations. No differences in gender were found in the time estimation task during sleep deprivation. The variations in time estimation correlated significantly with body temperature, skin resistance level, and SSS throughout the sleep deprivation period. When body temperature is elevated, indicating a high level of activation, the interval tends to be underestimated, and vice versa. When the skin resistance level or SSS is elevated (low activation), time estimation is lengthened, and vice versa. This lengthening is important because many everyday situations involve duration estimation under moderate to severe sleep loss. Actual or potential applications of this research include transportation systems, emergency response work, sporting activities, and industrial settings in which accuracy in anticipation or coincidence timing is important for safety or efficiency. 相似文献

Phosphorylation is the predominant posttranslational modification involved in regulating enzymatic activity and mediating signal transduction in prokaryotic and eukaryotic cells. Selective enrichment of phosphorylated peptides prior to mass spectrometric analysis facilitates identification of phosphorylated proteins, determination of specific phosphorylated residues, and characterization of the conditions under which phosphorylation occurs. Such protocols have been established for peptides containing residues that form phosphoesters, such as serine and threonine, using immobilized metal-ion affinity chromatography. Despite the importance of histidine phosphorylation in two-component signal transduction pathways, similar protocols for peptides containing phosphorylated histidine (P-His) residues have proven elusive, due to the instability of these modifications and the propensity of unphosphorylated histidines to interact with immobilized metals ions. We describe a method for the selective extraction of a P-His-containing peptide using immobilized copper(II) ions and disposable metal-chelating pipet tips (ZipTipMC, Millipore). The method is contingent upon pH-dependent interactions between the phosphate group and immobilized copper(II) ions. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry with postsource decay confirms the identity and phosphorylation state of the extracted peptide. Peptides containing unphosphorylated histidine residues or other phosphorylated amino acids are not retained, demonstrating the specificity of the method for P-His-containing peptides. 相似文献
