Androstenone (5α-androst-16-en-3-one) is a volatile steroid which is synthesized in boar testes and stored in high amounts in fat thus leading to an urine-like odor in pork. Whereas microtiter assays (MTA) exist for fat determination, measurement in blood with MTA was not yet possible. The system reported here is based on a specific antiserum and a horse radish peroxidase conjugate as tracer. For coating bovine serum albumin was avoided which otherwise would lead to unspecific binding. Blood plasma was extracted with n-hexane, solvent was evaporated in a vacuum centrifuge and the extract was taken up in buffer with 10% methanol. Assay sensitivity was 2.5pg/well (0.025ng/mL), specificity was confirmed by GC-MS (r=0.96; n=50) and inter- and intraassay variation were 4.9% and 5.9%, respectively. Thus an assay system is available for studies in pig production and in wildlife research due to pheromone activity also in other species.  相似文献

5-Fluoroanthranilic acid (FAA)-resistant mutants were selected in homothallic diploids of three Saccharomyces species, taking care to isolate mutants of independent origin. Mutations were assigned to complementation groups by interspecific complementation with S. cerevisiae tester strains. In all three species, trp3, trp4 and trp5 mutants were recovered. trp1 mutants were also recovered if the selection was imposed on a haploid strain. Thus, FAA selection may be more generally applicable than was previously described.  相似文献

This paper answers the research question: Can the contactless induced energy supply from a novel inductive floor be used to navigate omnidirectional automated guided vehicles (AGVs)?

In contrast to existing systems a novel inductive floor enables AGVs traveling through production without charging breaks. This floor consists of tiles with inductive modules, which supply the AGV with energy. In addition to supplying power to the AGV, the inductive modules are also intended to guide the vehicle through production. To enable such a guidance sensors placed in the AGV measure the induced voltages of the floor. To answer the research question these voltages are calculated with the help of an electromagnetic simulation of the AGV’s travel on the inductive tiles. To estimate the position as well as rotation of the AGV depending on the simulated voltages as inputs a novel algorithm is presented. During the travel the AGV is able to move in arbitrary directions independently of its orientation. To control the omnidrectional AGV consistently without singularities, a transformation in Omni-Curve-Parameters (OCP) is proposed. As simulation case study a four wheeled steering- and velocity controlled AGV is introduced. For the evaluation a novel motion model depending on the input OCP is presented. This model is compared to the estimation of the position to verify the accuracy and the reproducibility of the algorithm.

Consistent with a positive psychology perspective, this longitudinal study investigated relations between positive and negative nonwork experiences (i.e., feeling recovered, thinking about the positive and negative aspects of one’s work during leisure time) with different job performance dimensions. In total, 358 employees working with persons with special needs responded to two questionnaires at an interval of 6 months. Results from hierarchical regression analyses showed that feeling recovered during leisure time predicted an increase in task performance after 6 months. This relation was mediated by occupational self-efficacy. Positive work reflection was found to predict an increase in proactive behavior (personal initiative, creativity) and organizational citizenship behavior. Negative work reflection was unrelated to job performance. Our results emphasize the role of positive nonwork experiences for employees’ job performance. Oxidative stress is a central mechanism by which the receptor for advanced glycation endproducts (RAGE) mediates its pathological effects. Multiple experimental inquiries in RAGE-expressing cultured cells have demonstrated that ligand-RAGE interaction mediates generation of reactive oxygen species (ROS) and consequent downstream signal transduction and regulation of gene expression. The primary mechanism by which RAGE generates oxidative stress is via activation of NADPH oxidase; amplification mechanisms in the mitochondria may further drive ROS production. Recent studies indicating that the cytoplasmic domain of RAGE binds to the formin mDia1 provide further support for the critical roles of this pathway in oxidative stress; mDia1 was required for activation of rac1 and NADPH oxidase in primary murine aortic smooth muscle cells treated with RAGE ligand S100B. In vivo, in multiple distinct disease models in animals, RAGE action generates oxidative stress and modulates cellular/tissue fate in range of disorders, such as in myocardial ischemia, atherosclerosis, and aneurysm formation. Blockade or genetic deletion of RAGE was shown to be protective in these settings. Indeed, beyond cardiovascular disease, evidence is accruing in human subjects linking levels of RAGE ligands and soluble RAGE to oxidative stress in disorders such as doxorubicin toxicity, acetaminophen toxicity, neurodegeneration, hyperlipidemia, diabetes, preeclampsia, rheumatoid arthritis and pulmonary fibrosis. Blockade of RAGE signal transduction may be a key strategy for the prevention of the deleterious consequences of oxidative stress, particularly in chronic disease.  相似文献

Interaction between insect herbivores and host plants can be modulated by endogenous and exogenous compounds present in the source of food and might be successfully exploited in Colorado potato beetle (CPB) pest management. Feeding tests with CPB larvae reared on three solanaceous plants (potato, eggplant and tomato) resulted in variable larval growth rates and differential susceptibility to Bacillus thuringiensis Cry3Aa toxin as a function of the host plant. An inverse correlation with toxicity was observed in Cry3Aa proteolytic patterns generated by CPB midgut brush-border membrane vesicles (BBMV) from Solanaceae-fed larvae, being the toxin most extensively proteolyzed on potato, followed by eggplant and tomato. We found that CPB cysteine proteases intestains may interact with Cry3Aa toxin and, in CPB BBMV from larvae fed all three Solanaceae, the toxin was able to compete for the hydrolysis of a papain substrate. In response to treatment with the JA-dependent plant inducer Hexanoic acid (Hx), we showed that eggplant reduced OPDA basal levels and both, potato and eggplant induced JA-Ile. CPB larvae feeding on Hx-induced plants exhibited enhanced Cry3Aa toxicity, which correlated with altered papain activity. Results indicated host-mediated effects on B. thuringiensis efficacy against CPB that can be enhanced in combination with Hx plant induction.  相似文献

In view of recent reports of a strong multivalent effect in glycosidase inhibition, a library of β‐CD‐based multivalent iminosugars has been efficiently synthesized by way of CuI‐catalyzed azide–alkyne cycloaddition (CuAAC). In combination with the first application of isothermal titration calorimetry (ITC) experiments to the study of multivalent iminosugar–enzyme interactions, the inhibition properties of these click clusters were evaluated on a panel of glycosidases. The structural parameters that were varied include valency, peripheral ligand structure, and topology. The inhibition results obtained with the iminosugar clusters further highlight the importance of multivalency in the inhibition of α‐mannosidase. Generally, the evaluated multivalent iminosugars displayed comparable thermodynamic signatures of binding towards α‐mannosidase (Jack bean): that is, large negative enthalpies of complexation coupled with small entropies of either sign. In addition, the enthalpy–entropy compensation observed in all tested cases may be attributed to a common mechanism of dissociation for the enzyme–multivalent iminosugar interactions. The measured binding stoichiometries indicated that each iminosugar cluster interacts with no more than one protein molecule.  相似文献
