Herpes simplex virus (HSV) DNA is cleaved from concatemers and packaged into capsids in infected cell nuclei. This process requires seven viral proteins, including UL15 and UL28. UL15 expressed alone displays a nuclear localization, while UL28 remains cytoplasmic. Coexpression with UL15 enables UL28 to enter nuclei, suggesting an interaction between the two proteins. Additionally, UL28 copurified with UL15 from HSV-infected cells after ion-exchange and DNA affinity chromatography, and the complex sedimented as a 1:1 heterodimer upon sucrose gradient centrifugation. These findings are evidence of a physical interaction of UL15 and UL28 and a functional role for UL15 in directing UL28 to the nucleus. 相似文献

Evaluation of the fit of a data set to an algebraic model often relies on the squared correlation coefficient (r-2). Algebraic models commonly prescribe functions between two variables that are monotonic. We created data sets that are monotonic but otherwise random and computed their r-2 values. These values are shown to far exceed those for data sets not constrained to be monotonic. We tabulated selected cumulants of the distribution of r-2 for monotone data fit to two common models, linear and power functions, for several conditions of randomness for data sets comprising 3, 9, and 15 points. Random monotone data fit all these functions well. The common experimental practices of averaging several sets of data and using regular spacing in the values of the independent variable both tend to produce slight additional improvements in fit. Consideration of these results reveals that correlation alone is inadequate to test the fit of monotone data to algebraic models. Some helpful auxiliary techniques are recommended. Social housing in Britain is managed by a large grouping of ‘housing managers’, an occupational label which is viewed as unproblematic by most housing academics, and therefore, is largely taken for granted. Urban sociologists posit that a collective identity is crucial to professional status, and hence have been sceptical of housing managers' prospects of furthering their aims collectively. The paper departs from this focus on the collective endeavours of housing management as a profession to examine how housing managers themselves create and present self-identities. Specifically, it argues that housing managers use the lack of a collective identity as an opportunity to manage the ‘spoiled identity’ of being in an ‘invisible’ and marginalised profession. In doing so, they construct individualised occupational trajectories which bear little relation to the domain professionalism of more established occupations. 相似文献
Dominant follicles are those that continue to develop and have the potential to ovulate while subordinate follicles regress. Characteristics of dominant follicles include a larger diameter, higher intrafollicular estradiol, and lower IGF-binding protein (IGFBP)-4 concentrations compared with other cohort follicles. Follicle development is regulated by endocrine hormones that act via intracellular signaling pathways. Here, we show the differences in Akt, Erk, c-Jun N-terminal protein kinase, and p-38 signaling pathways between dominant and subordinate follicles at the dominance stage of the follicle wave. However, earlier in the follicle wave (dominant follicle selection), there were only differences in the levels of Akt and Erk signal transduction proteins among dominant and subordinate follicles. Using this profile of Akt and Erk protein expression in granulosa and theca cells of selected dominant follicles compared with subordinate follicles, we suggest a predictive model to identify future dominant and subordinate follicles from the pool of otherwise similar cohort follicles at the time of follicle wave emergence. We conclude that the Erk and Akt signal transduction pathways are important for dominant follicle selection and development and, furthermore, that the observed differences in these pathways mark the future dominant follicle from subordinate follicles before differences in follicular diameter, follicular fluid estradiol, and IGFBP-4 concentrations are apparent. 相似文献
A yeast shipment method suitable (depending on vessel size) for 1 to 20 litres of slurried yeast is described. The method was investigated primarily assuming transit (storage) temperature at approximately 21°C. Under these conditions, the contained yeast was stable over 116 h at temperatures well under 16°C, and maintained its viability, fermentability and vigour to reinitiate new fermentations. Under more vigorous climatic conditions, shipment time would have to be reduced, but adequate protection time was afforded for all but the most extreme sub-zero situations. 相似文献
Capillary electrophoresis/electrospray ionization-mass spectrometry (CE/ESI-MS) was applied to the analysis of underivatized amino acids and the separation of their D/L-enantiomers. Under full-scan mode, all standard protein amino acids were separated and detected at low-femtomole levels using a 130-cm-long, 20-microm-i.d., 150-microm-o.d. underivatized fused-silica capillary with 1 M formic acid as the background electrolyte. The CE/ESI-MS technique was also applied to the separation of L-arginine from L-canavanine (a close analogue of arginine where the terminal methylene linked to the guanidine group of arginine is replaced by an oxygen atom) in a complex mixture containing all standard protein amino acids. The utility of CE/ESI-MS in the analysis of real-world samples was demonstrated by the identification of two metabolic diseases (PKU and tyrosinemia) through blood analysis with minimal sample preparation. In addition, the on-line separation of 11 underivatized L-amino acids from their D-enantiomers was achieved by using a 30 mM solution of (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid as the background electrolyte. 相似文献
In this work, two medium Mn steels (5.8 and 5.7 wt pct Mn) were subjected to a quenching and partitioning (Q&P) treatment employing a partitioning temperature which corresponded to the start of austenite reverse transformation (ART). The influence of a 1.6 wt pct Ni addition in one of the steels and cycle parameters on austenite stability and mechanical properties was also studied. High contents of retained austenite were obtained in the lower quenching temperature (QT) condition, which at the same time resulted in a finer microstructure. The addition of Ni was effective in stabilizing higher contents of austenite. The partitioning of Mn and Ni from martensite into austenite was observed by TEM–EDS. The partitioning behaviour of Mn depended on the QT condition. The lower QT condition facilitated Mn enrichment of austenite laths during partitioning and stabilization of a higher content of austenite. The medium Mn steel containing Ni showed outstanding values of the product of tensile strength (TS) and total elongation (TEL) in the lower QT condition and a higher mechanical stability of the austenite.
Polycomb group (PcG) proteins are epigenetic regulators that facilitate both embryonic development and cancer progression. PcG proteins form Polycomb repressive complexes 1 and 2 (PRC1 and PRC2). PRC2 trimethylates histone H3 lysine 27 (H3K27me3), a histone mark recognized by the N-terminal chromodomain (ChD) of the CBX subunit of canonical PRC1. There are five PcG CBX paralogs in humans. CBX2 in particular is upregulated in a variety of cancers, particularly in advanced prostate cancers. Using CBX2 inhibitors to understand and target CBX2 in prostate cancer is highly desirable; however, high structural similarity among the CBX ChDs has been challenging for developing selective CBX ChD inhibitors. Here, we utilize selections of focused DNA encoded libraries (DELs) for the discovery of a selective CBX2 chromodomain probe, SW2_152F. SW2_152F binds to CBX2 ChD with a Kd of 80 nM and displays 24-1000-fold selectivity for CBX2 ChD over other CBX paralogs in vitro. SW2_152F is cell permeable, selectively inhibits CBX2 chromatin binding in cells, and blocks neuroendocrine differentiation of prostate cancer cell lines in response to androgen deprivation. 相似文献
