A polymerase chain reaction (PCR) assay for the rapid and sensitive detection of the most prolific histamine former, Morganella morganii, was developed. 16S rDNA targeted PCR primers were designed, and the primer specificity and sensitivity of the PCR assay were evaluated. The 16S rDNA sequence (1,503 bp) for M. morganii showed 95% identity to those for enteric bacteria, i.e., Enterobacter spp., Klebsiella spp., Citrobacter spp., Hafnia alvei, Proteus spp., and Providencia spp. The unique primers for M. morganii were designed on the basis of the variable regions in the 16S rDNA sequence. The primers showed positive reactions with all M. morganii strains tested. However, PCR amplification was not detected when the primers were tested with other enteric or marine bacteria. When the sensitivity of the assay was evaluated, M. morganii was detected at levels ranging from 10(6) to 10(8) CFU/ml in albacore homogenate after the PCR amplification. The sensitivity of the assay was greatly improved with the enrichment of samples, and 9 CFU of M. morganii per ml of albacore homogenate was detected after 6 h of enrichment at 37 degrees C.  相似文献

An oblique flat surface model is used lo simulate the rotating roll. The flow regions proposed by Landau and Levich is utilized in the analysis. The inertia force terms are included and a more realistic 2-dimensional boundary conditions for the free surface is used in this paper. Also, the solution is obtained by utilizing a refinement of integral method. The results are compared with available experimental data. It is shown that the prediction is sufficiently accurate over a wide range of capillary number, and better than the other existing results. The film thickness increase with respect to increasing capillary number up to a certain higher values, from then this tendency slows down and even decreases due to inertia effects  相似文献

This study was performed in a fluidized-bed reactor (FBR) filled with manganese-coated sand (MCS) to treat copper-contaminated wastewater. The adsorption characteristics of MCS, the adsorption equilibrium of MCS, and the copper removal capacity by MCS in FBR were investigated. In terms of the adsorption characteristics of MCS, the surface of MCS was evaluated using a scanning electron microscope (SEM). Energy dispersive analysis (EDS) of X-rays indicated the composition of MCS, and the quantity of manganese on MCS was determined by means of acid digestion analysis. The experimental results indicated that copper was removed by both sorption (ion exchange and adsorption) and coprecipitation on the surface of MCS in FBR. Copper removal efficiency was highly dependent on the pH and increased with increasing pH from pH 2 to 8. After the copper adsorption by MCS, the pH in solution was decreased. When the MCS concentration was greater than 10 g/l, the copper adsorptivities obtained by FBR were almost the same as that from the shaker and when the MCS concentration reached 40 g/l, the copper adsorptivity in FBR was greater than that from the shaker. The adsorption sites of MCS could be used efficiently by the FBR. A Langmuir adsorption isotherm equation fit the measured adsorption data from the batch equilibrium adsorption test better than the Freundlich adsorption isotherm equation did. An immunoassay system was developed for efficient detection of prohibited meat and bone meal (MBM) in animal feed. Monoclonal antibodies (MAbs) were raised against bovine smooth muscle autoclaved at 130 degrees C for 20 min. Among the 1,500 supernatants of hybridoma cells screened, MAbs 3E1, 1G3, and 3E10 were selected and characterized in this study. The first set of MAbs produced, 3E1 and 1G3, had stronger reactivity against MBM than against smooth muscle that was heat treated at 90 degrees C for 10 min. However, reactivity gradually increased against smooth muscle that was autoclaved at 130 degrees C for up to 1 h. The enzyme-linked immunosorbent assay for detection of MBM in animal feed was optimized with the MAb 3E10 because of its superior performance. MAb 3E10 diluted to 100-fold was used to differentiate bovine MBM from that of other species in ingredients used for commercial animal feeds and could detect down to 0.05% MBM mixed in animal feed.  相似文献

Linear stability theory is used to study the stability characteristics of condensate film flow down a vertical cylinder, taking into account the surface tension and the mass transfer due to phase change at the liquid-vapor interface. The problem is simplified by using the local parallel-flow approximation. Method of perturbation is applied and leads to closed form expressions for the wave amplification rate, the wave velocity, the neutral stability curve, and the critical Reynolds number. When the lateral curvature of the cylinder is taken into account, the surface tension, just as condensation mass transfer rate behaving on the vertical plate, has a dual role in the mechanism inducing instability. Results indicate that the streamwise curvature of the perturbed flow is a stabilizing effect, and the lateral curvature, on the other hand, is a destabilizing effect. The curvature of cylinder intensifies instability of the flow in comparison with the plane flow. The flow on the inner face of a cylinder is found to be more unstable than flow of the same liquid layer on the outer face.  相似文献

N2O emissions from a fertilized humid grassland near Cork, Ireland were continuously measured during 2003 using an eddy covariance system. For most of the year emissions were close to zero and 60% of the emissions occurred in eight major events of 2–20 days’ duration. Two hundred and seven kg ha−1 of synthetic N and 130 kg ha−1 organic N were applied over the year and the total measured annual N2O emission was 11.6 kg N ha−1. The flux data were used to test the prediction of N2O emissions by the DNDC (DeNitrification – DeComposition) model. The model predicted total emissions of 15.4 kg N ha−1, 32 % more than the observed emissions. On this basis the model was further used to simulate (a) background (non-anthropogenic) N2O emissions and (b) the effect on N2O emissions of future climate perturbations based on the Hadley Center model output of the IS92a scenario for Ireland. DNDC predicts 1.7 kg N ha−1 year−1 of background N2O emissions, accounting for 15% of the observed emissions. Climate shifts will increase total annual modeled N2O emissions from 15.4 kg N ha−1 to 22.4 kg N ha−1 if current levels of N applications are maintained, or to 21.2 kg N ha−1 if synthetic N applications are reduced to 170 kg N ha−1 to comply with recent EU water quality legislation. Thus the projected increase in N2O emissions due to climate change is far larger than the decrease expected from reduced fertilizer applications.  相似文献

Molecular dynamics simulations and quantum transport theory are employed to study the temperature-dependent electrical properties of individual (12,0) zigzag and (5,5) armchair carbon nanotubes deposited on silicon substrates. The results demonstrate that the magnitude of the leakage current depends on the length of the nanotube. Furthermore, the leakage current is generated periodically along the length of the nanotube. Finally, the results indicate that given an appropriate value of the applied bias voltage, the induced current varies linearly with the temperature over specific temperature ranges. As a result, the temperature can be inversely derived from the measured current signal. Overall, the results show that the (12,0) zigzag and (5,5) armchair carbon nanotubes are suitable for temperature sensing applications over temperature ranges of 200-420?K and 300-440?K, respectively.  相似文献

Interaction of Escherichia coli O157:H7/pGFP with hydroponically grown lettuce plants was evaluated in this study. Lettuce seedlings were planted in contaminated Hoagland's nutrient solution and thereafter subjected to gamma radiation at 0.25, 0.5, and 0.75 kGy, and aqueous chlorine at 200 ppm. There was no trace of E. coli O157:H7/pGFP in lettuce leaves harvested from noncontaminated nutrient solution (control); however, for plants grown in contaminated nutrient solution, the pathogen was recovered from the leaves disinfected with 80% ethanol and 0.1% mercuric chloride. Most of the lettuce seedlings grown in contaminated nutrient solution tested negative for E. coli O157:H7/pGFP under controlled conditions. Gamma radiation at 0.25 and 0.5 kGy, and aqueous chlorine at 200 ppm failed to eliminate E. coli O157:H7/pGFP in lettuce tissue completely; however, the bacteria were not detected in 0.75-kGy treated plants. The presence of E. coli O157:H7/pGFP in lettuce leaves is an indication that the pathogen migrated from the contaminated hydroponic system through the roots to the internal locations of lettuce tissue. Due to inaccessibility and limited penetrating power, aqueous chlorine could not eliminate the bacteria localized in the internal tissue. Findings from this study suggest that gamma irradiation was more efficacious than was aqueous chlorine to control internal contamination in hydroponically grown lettuce. Gamma irradiation is a process that processors can use to inactivate E. coli O157:H7 and therefore, consumers benefit from a safer food product [corrected]  相似文献

This paper describes a novel approach in simulating laminar to turbulent transition by using two-equation models. The Total Stresses Limitation (TSL) concept is used to make the two-equation model capable of predicting high-Reynolds-number transitional flow. In order to handle the transition triggered by laminar separation at a low Reynolds number location, which commonly occurs in high speed flow, a sensor is introduced to detect separation and trigger transition in the separated zone. Test cases include the classical flat-plate turbulent boundary flow, and low-pressure turbine cascade flows at design and off-design conditions. © 1998 John Wiley & Sons, Ltd.  相似文献
