Partial glycerides are widely used ingredients in confectionery products that can be produced from natural fats. In a biocatalytic partial hydrolysis of cocoa butter and palm oil fractions, a product mixture containing 1.5% monoglycerides and 5.5% diglycerides intended for the use in confectionery products was created. This study is a proof of principle that shows the feasibility of monitoring the biocatalytic partial hydrolysis of these two natural fats in situ with ATR-FTIR spectroscopy. An economic approach was utilised for calibration since partial glyceride calibration standards are costly and poorly available. The released compounds were quantified by means of chemometric modelling, and the model was validated with gas chromatography. Resulting root mean square errors were in the low per cent range. Additionally, the results indicate that distinction of the released free fatty acids is possible with ATR-FTIR spectroscopy.  相似文献

There is an increasing need for high-throughput analyses of plants and food samples for the presence of specific DNA sequences, e.g. transgenic contaminations. We developed and optimized conditions for the automated isolation of DNA from several maize tissues and various edibles containing maize using the MagNA Pure LC system (Roche Applied Science). Our results show that the system provided is capable of isolating DNA from any tested source. Quantification of an endogenous gene by LightCycler real-time PCR revealed that the DNA is suitable in quality and quantity for multiple PCR analyses.  相似文献

Smokers with smoking-related diseases who are hospitalized in rehabilitation centers should be offered smoking cessation. This is the first study evaluating whether telephone booster sessions after intensive inpatient treatment are an effective strategy. The present study was conducted in 13 rehabilitation centers for somatic disorders as a prospective multicenter study with a randomized treatment-control group design. We compared abstinence rates after hospital discharge from treatment that included a group smoking cessation program with (treatment group) and without telephone booster sessions (control group). Data from 290 smokers were analyzed. After 6 and 12 months the treatment group achieved abstinence rates twice as high as those of the control group. Men profited more from telephone booster sessions than did women. Results indicated that telephone booster sessions were highly effective (even) after an inherently intensive group program during a hospital stay. Further research should focus on the special needs of women receiving telephone counseling.  相似文献

Total and size-segregated Pt and Pd emission factors from on-road vehicles were measured in the Kaisermühlen Tunnel in Vienna, Austria. Aerosol sampling was performed simultaneously inside and outside the tunnel during April and May 2005. Analysis of the acid-digested aerosol samples was performed using a preconcentration procedure with subsequent on-line detection by electro-thermal atomic absorption spectrometry (ETAAS). Inside the tunnel distinctly increased Pt and Pd concentrations were found with highest levels in total suspended particulate matter samples and reduced concentrations in the size-segregated PM10 and PM2.5 samples. Emission factors were calculated from concentration differences between tunnel inside and tunnel outside samples, the distance between tunnel entrance and sampling location, the ventilation rate, and the number of vehicles passing through the tunnel. Emission rates observed for Pt ranged from 38 +/- 5.9 to 146 +/- 13 ng veh(-1) km(-1), whereas the emission factors of Pd varied between 13 +/- 2.1 and 42 +/- 4.1 ng veh(-1) km(-1). Variations in the emission rates were assumed to originate from alterations in traffic conditions. Size-segregated investigations revealed that the major part of Pt and Pd emissions were released in the coarse aerosol mode (size fraction > PM10), nevertheless a considerable fraction (approximately 12% and approximately 22% respectively) was emitted in the inhalable PM2.5 fraction.  相似文献

Deep-water oxygen concentrations in the Baltic Sea are influenced by eutrophication, but also by saltwater inflows from the North Sea. In the last two decades, only two major inflows have been recorded and the lack of major inflows is believed to have resulted in a long-term stagnation of the deepest bottom water. Analyzing data from 1970 to 2000 at the basin scale, we show that the estimated volume of water with oxygen, <2 mL L(-1), was actually at a minimum at the end of the longest so-called stagnation period on record. We also show that annual changes in dissolved inorganic phosphate water pools were positively correlated to the area of bottom covered by hypoxic water, but not to changes in total phosphorus load, thus addressing the legacy of eutrophication on a basinwide scale. The variations in phosphorus pools that have occurred during the past decades do not reflect any human action to reduce inputs. The long residence time and internally controlled variation of the large P pool in the Baltic Sea has important implications for management of both N and P inputs into this eutrophicated enclosed basin.  相似文献

A commercially available PCR kit (AnDiaTec Salmonella sp. PCR-ELISA) was developed and evaluated for the detection of Salmonella sp. in food samples. The test is based on PCR amplification and hybridization of the amplified DNA to a microtiter plate followed by the detection of PCR product in the manner of an enzyme-linked immunosorbent assay test. The sensitivity and specificity were evaluated first with Salmonella pure cultures and artificially contaminated food samples, including food types for which an inhibition of the PCR reaction was expected. Both experiments proved a very good sensitivity, specificity, and reliability of the test with a very broad range of food types. In a second evaluation study, more than 1,100 food samples of different types were tested in parallel with the PCR method and with the International Standardization Organization 6579 bacteriological reference method. The results of this evaluation study and the results from other experiments on dilutions of artificially contaminated food samples led to the establishment of a positive-negative cutoff value (optical density at 450 nm of more than 0.9) with respect to the conventional bacteriological method. Using this positive-negative cutoff, 98% agreement to the bacteriological method was obtained.  相似文献

The contamination of honey with hepatotoxic pyrrolizidine alkaloids (PAs) is a well-known hazard for food safety. While management strategies and controls of the honey industry aim to reduce the PA levels, uncertainties remain with regard to the safety of regionally produced and marketed honey. In addition, a previous study showed large differences of results obtained after various periods of storage and apparent differences between the analytical results of different laboratories. Therefore, this study aimed at examining these uncertainties by monitoring the impact of storage on the PA and PA N-oxide (PANO) content of two freshly harvested honeys and on possible demixing effects caused by pollen settling. Additionally, three analytical approaches – target analysis with matrix-matched calibration or standard addition and a sum parameter method – were applied for a comparative analysis of 20 honeys harvested in summer 2016. All samples originated from Schleswig-Holstein in Northern Germany where the PA plant Jacobaea vulgaris is currently observed on a massive scale. The results of the time series analyses showed that PANO levels markedly decreased within a few weeks and practically reached the LOD 16 weeks after harvest. Tertiary PAs, by contrast, remained stable and did not increase as a consequence of PANO decrease. The experiments on a putative demixing, which may result in a heterogeneous distribution of PAs/PANOs, revealed that there was no such effect during storage of up to 12 weeks. A comparison of the PA/PANO levels obtained by different analytical approaches showed that in some cases the sum parameter method yielded much higher levels than the target approaches, whereas in other cases, the target analysis with standard addition found higher levels than the other two methods. In summary, the results of this study highlight uncertainties regarding the validity and comparability of analytical results and consequently regarding health risk assessment.  相似文献

A new identification and indexing for the phase BaAl2Ti5O14 were accomplished using an X-ray diffraction technique. The new lattice parameters for the tetragonal lattice structure are: a0=9.990 × 10-10 m and c0=12.264 × 10-10 m, with a corresponding volume 1.224 × 10-27 m3. The data provided by the Joint Committee on Powder Diffraction Standards are inconsistent both in lattice parameter values and Miller indices. The X-ray powder diffraction pattern of BaAl2Ti5O14 was indexed using the LSUCR (least-squares unit cell refinement) computer program.  相似文献

Ozone and hydroxyl radical (*OH) reaction kinetics were measured for 14 antibacterial compounds from nine structural families, to determine whether municipal wastewater ozonation is likely to result in selective oxidation of these compounds' biochemically essential moieties. Each substrate is oxidized by ozone with an apparent second-order rate constant, k'(O3,app) > 1 x 10(3) M(-1) s(-1), at pH 7, with the exception of N(4)-acetylsulfamethoxazole (K'(O3,app) is 2.5 x 102 M(-1) s(-1)). k'(O3,app) values (pH 7) for macrolides, sulfamethoxazole, trimethoprim, tetracycline, vancomycin, and amikacin appear to correspond directly to oxidation of biochemically essential moieties. Initial reactions of ozone with N(4)-acetylsulfamethoxazole, fluoroquinolones, lincomycin, and beta-lactams do not lead to appreciable oxidation of biochemically essential moieties. However, ozone oxidizes these moieties within fluoroquinolones and lincomycin via slower reactions. Measured k'(O3,app) values and second-order *OH rate constants, k'(*OH,app) were utilized to characterize pollutant losses during ozonation of secondary municipal wastewater effluent. These losses were dependent on k'(O3,app), but independent of k'(*OH,app). Ozone doses > or =3 mg/L yielded > or =99% depletion of fast-reacting substrates (K'(O3,app) > 5 x 10(4) M(-1) s(-1)) at pH 7.7. Ten substrates reacted predominantly with ozone; only four were oxidized predominantly by .OH. These results indicate that many antibacterial compounds will be oxidized in wastewater via moiety-specific reactions with ozone.  相似文献
