Filtered olive oil samples spiked with three different concentrations of λDNA were stored at 25 °C under a 12 h photoperiod for up to a year. These samples were used for DNA extraction and PCR amplification of λDNA amplicons of 107, 415 and 691 bp length. The amplification signal was gradually decreased with longer storage periods, while the strength of the signal was related to the initial concentration of spiking λDNA particularly during longer storage periods. The 107 bp amplicon was the only one successfully amplified from all the samples, regardless of both concentration of spiking λDNA and storage period. The amplification of 415 and 691 bp amplicons was not successful for samples stored longer than a threshold period of 20 and 10 days, respectively. These results suggest that detection of polymorphic markers requiring DNA templates shorter than 100 bp might have a wider range of applications in DNA fingerprinting of olive oil. In addition, the DNA extracts were tested for the presence of inhibitors in PCR amplification reactions of yeast DNA amplicons. The inhibitory effect of olive DNA extracts was partial and gradually increased with the storage period of the olive oil samples used for the DNA extraction.  相似文献

Zusammenfassung Im Rohrleitungsbau und insbesondere in der nuklearen Kraftwerkstechnik sind die Rohrb?gen die am st?rksten beanspruchten Konstruktionselemente. Biegemomente, die durch thermische Belastung, schnelles Schlie?en von Sicherheitsventilen, Wasserhammer usw. entstehen, belasten die Rohrleitung, wobei die Rohrb?gen ihre ursprünglich kreisrunden Querschnitte ?ndern (Ovalisierung). Diese Eigenschaft der Krümmerquerschnitte ist der Grund für ihre h?here Flexibilit?t und die zus?tzlichen Spannungen in den Rohrbogenbereichen. Der vorliegende Beitrag beschreibt eine einfache N?herungsmethode, welche die Quantifizierung der Ovalisierungseinflüsse in der Rohrleitung auf Grund von Dehnungsmessungen erm?glicht. Dies erlaubt die Behandlung relevanter Fragen wie die experimentelle Verifizierung von FE-Programmen und- modellen und der entsprechenden Regelwerte für Rohrb?gen (Korrekturfaktoren für Flexibilit?t und Spannung).  相似文献

Indoor exposure to air contaminants penetrating from the outdoor environment depends on a number of key processes and parameters such as the ventilation rate, the geometric characteristics of the indoor environment, the outdoor concentration and the indoor removal mechanisms. In this study two alternative methods are used, an analytical and a numerical one, in order to study the time lag and the reduction of the variances of the indoor concentrations, and to estimate the deposition rate of the air contaminants in the indoor environment employing both indoor and outdoor measurements of air contaminants. The analytical method is based on a solution of the mass balance equation involving an outdoor concentration pulse which varies sinusoidally with the time, while the numerical method involves the application of the MIAQ indoor air quality model assuming a triangular pulse. The ratio of the fluctuation of the indoor concentrations to the outdoor ones and the time lag were estimated for different values of the deposition velocity, the ventilation rate and the duration of the outdoor pulse. Results have showed that the time lag between the indoor and outdoor concentrations is inversely proportional to the deposition and ventilation rates, while is proportional to the duration of the outdoor pulse. The decrease of the ventilation and the deposition rate results in a rapid decrement of the variance ratio of indoor to outdoor concentrations and to an increment of the variance ratio, respectively. The methods presented here can be applied for gaseous species as well as for particulate matter. The nomograms and theoretical relationships that resulted from the simulation results and the analytical methods respectively were used in order to study indoor air phenomena. In particular they were used for the estimation of SO2 deposition rate. Implications of the studied parameters to exposure studies were estimated by calculating the ratio of the indoor exposure to the exposure outdoors. Limitations of the methods were explored by testing various scenarios which are usually met in the indoor environment. Strong indoor emissions, intense chemistry and varying ventilation rates (opening and closing of the windows) were found to radically influence the time lag and fluctuation ratios.  相似文献

In the present work, fat reduction of Greek strained yoghurt ice cream (YIC) was carried out in three proportional milkfat levels i.e. 30, 50 and 70% using three types of oligosaccharides namely long-chain inulin, oligofructose and maltodextrin 12 DE. Greek strained yoghurt was blended with ice cream mixes in ratios of 1:3 and 1:1. The physico-chemical, textural and thermal characteristics of the YIC mixes and their obtained frozen end products were determined. The sensory modalities (olfactory, gustatory, tactile and oro-tactile) of the YIC were monitored following 2 and 16 weeks of quiescent frozen storage at ?25 °C. Milkfat reduction impaired significantly (p?<?0.05) the perceived creaminess and mouthcoating sensation stimuli, whist it intensified the oral tissue friction associated sense stimuli such as astringency, wateriness and coarseness. Long-chain inulin- and maltodextrin-based samples received the highest scores for creaminess, mouthcoating, gumminess, hardness and iciness. The increase of the yoghurt to ice cream mix ratio escalated the friction/recrystallization-associated sensations e.g. astringency, sourness, coarseness and wateriness. Notwithstanding yoghurt supplementation reinforced the pseudoplasticity and macroviscosity of the ice cream mixes, it suppressed their aeration capacity leading to heavy-bodied ice creams. However, no significant effects of yoghurt supplementation level on the colligative and meltdown rate of the YIC formulations were identified. Partial least squares coupled discriminant analysis (PLS-DA) revealed that fat reformulation of YICs using oligosaccharides results in a substantially diversified sensory profile. Generally, a 50% fat reduction of YICs using long-chain oligosaccharides appears to be a technologically tangible solution.  相似文献

A simple approach is presented for the modeling of complex oscillatory thermal-fluid systems capable of converting low grade heat into useful work. This approach is applied to the NIFTE, a novel low temperature difference heat utilization technology currently under development. Starting from a first-order linear dynamic model of the NIFTE that consists of a network of interconnected spatially lumped components, the effects of various device parameters (geometric and other) on the thermodynamic efficiencies of the device are investigated parametrically. Critical components are highlighted that require careful design for the optimization of the device, namely the feedback valve, the power cylinder, the adiabatic volume and the thermal resistance in the heat exchangers. An efficient NIFTE design would feature a lower feedback valve resistance, with a shorter connection length and larger connection diameter; a smaller diameter but taller power cylinder; a larger (time-mean) combined vapor volume at the top part of the device; as well as improved heat transfer behavior (i.e. reduced thermal resistance) in the hot and cold heat exchanger blocks. These modifications have the potential of increasing the relevant form of the second law efficiency of the device by 50% points, corresponding to a 3.8% point increase in thermal efficiency.  相似文献

In this paper, a promising heat engine technology capable of utilizing low grade heat is examined. Based on a two-phase thermofluidic oscillator concept, the novelty and advantage of this particular system lie in its use of phase change and its lack of reliance on inertia to sustain oscillations, though it is recognized that inertia will always be present in any physical manifestation of the engine. The system is analysed using lumped linearized one-dimensional network models, both with and without inertia, based on thermoacoustic principles and extending these to account for phase change. The gain (temperature difference between source and sink heat exchangers) and frequency at which marginal stability (desirable continuous oscillations) can be achieved is calculated. The effects of the load resistance (fluid drag) and fluid inertia, as well as of the flow resistance due the feedback valve on the marginal stability gain, frequency and exergetic efficiency of the system are investigated. It is found that an increase in feedback resistance leads to a need for a higher gain for oscillatory behaviour to be achieved. In addition, even though an increase in either the resistance or inertia in the load, or the feedback resistance at low values of these variables has almost no effect on the required gain and the oscillation frequency of the system, an increase in these variables can lead at higher values to increased gains and reduced frequencies. A reduced feedback resistance and greater load inertia can also lead to considerably higher efficiencies, while increasing the load resistance allows for an increase in efficiency until a maximum is reached, after which the efficiency decreases again. The validity of certain approximations made previously is considered, and it is shown that these must be made with care. The results from this study can be used for the improved design and optimization of such oscillators, and similar systems.  相似文献

The degradation of herbicides 4-chlorophenoxyacetic acid (4-CPA), 4-chloro-2-methylphenoxyacetic acid (MCPA), 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) in aqueous medium of pH 3.0 has been comparatively studied by anodic oxidation and electro-Fenton using a boron-doped diamond (BDD) anode. All solutions are totally mineralized by electro-Fenton, even at low current, being the process more efficient with 1 mM Fe2+ as catalyst. This is due to the production of large amounts of oxidant hydroxyl radical (OH) on the BDD surface by water oxidation and from Fenton’s reaction between added Fe2+ and H2O2 electrogenerated at the O2-diffusion cathode. The herbicide solutions are also completely depolluted by anodic oxidation. Although a quicker degradation is found at the first stages of electro-Fenton, similar times are required for achieving overall mineralization in both methods. The decay kinetics of all herbicides always follows a pseudo first-order reaction. Reversed-phase chromatography allows detecting 4-chlorophenol, 4-chloro-o-cresol, 2,4-dichlorophenol and 2,4,5-trichlorophenol as primary aromatic intermediates of 4-CPA, MCPA, 2,4-D and 2,4,5-T, respectively. Dechlorination of these products gives Cl, which is slowly oxidized on BDD. Ion-exclusion chromatography reveals the presence of persistent oxalic acid in electro-Fenton by formation of Fe3+-oxalato complexes, which are slowly destroyed by OH adsorbed on BDD. In anodic oxidation, oxalic acid is mineralized practically at the same rate as generated.  相似文献

Main objective of the presented research is the life time prediction of self-supporting flame-sprayed Al2O3- and Al2O3 -ZrO2 -TiO2-materials under constant load. The characteristic life time and its scatter were derived from stable crack growth tests and Weibull-statistics including the four-point-bending and ball-on-three-balls method. The potential life time was estimated in order to assess room-temperature handling and long term storage of self-supporting flame-sprayed alumina components. In terms of flexural strength, energy release, and subcritical crack growth parameters, distinct differences between both materials were shown. In turn, the characteristic life times only barely deviated from each other. From that the conclusion was drawn that life time performance under constant load application is governed by the characteristic flame-spray microstructure. However, advantages in the flame-spray processing of Al2O3 - ZrO2 -TiO2 are still given, attributed to its lower melting temperature.  相似文献

This white paper is the result of discussions during the FIPSE-4 conference ( http://fi-in-pse.org ) in June 2018. It aims to highlight open problems and provide directions for future research in the area of water with emphasis on its agricultural usages. Some of the open problems discussed are: (a) the use of ecosystems as unit operations to understand their role in providing freshwater and in cleaning polluted water; (b) consideration of interactions and independencies between flows of water and other resources, such as food, energy, materials, ecosystem services, and environmental emissions; (c) challenges in modeling, sensing, and closed-loop control in precision irrigation. In particular, the development of agro-hydrological models that balance computing speed versus solution details and accuracy: (d) The use of state and parameter estimation approaches, through field measurements, to obtain soil moisture levels accurately; and (e) decision support systems to administer water and nutrient needs for optimum yields of agricultural products.  相似文献
