Over the years, natural-based scaffolds have presented impressive results for bone tissue engineering (BTE) application. Further, outstanding interactions have been observed during the interaction of graphene oxide (GO)-reinforced biomaterials with both specific cell cultures and injured bone during in vivo experimental conditions. This research hereby addresses the potential of fish gelatin/chitosan (GCs) hybrids reinforced with GO to support in vitro osteogenic differentiation and, further, to investigate its behavior when implanted ectopically. Standard GCs formulation was referenced against genipin (Gp) crosslinked blend and 0.5 wt.% additivated GO composite (GCsGp/GO 0.5 wt.%). Pre-osteoblasts were put in contact with these composites and induced to differentiate in vitro towards mature osteoblasts for 28 days. Specific bone makers were investigated by qPCR and immunolabeling. Next, CD1 mice models were used to assess de novo osteogenic potential by ectopic implantation in the subcutaneous dorsum pocket of the animals. After 4 weeks, alkaline phosphate (ALP) and calcium deposits together with collagen synthesis were investigated by biochemical analysis and histology, respectively. Further, ex vivo materials were studied after surgery regarding biomineralization and morphological changes by means of qualitative and quantitative methods. Furthermore, X-ray diffraction and Fourier-transform infrared spectroscopy underlined the newly fashioned material structuration by virtue of mineralized extracellular matrix. Specific bone markers determination stressed the osteogenic phenotype of the cells populating the material in vitro and successfully differentiated towards mature bone cells. In vivo results of specific histological staining assays highlighted collagen formation and calcium deposits, which were further validated by micro-CT. It was observed that the addition of 0.5 wt.% GO had an overall significant positive effect on both in vitro differentiation and in vivo bone cell recruitment in the subcutaneous region. These data support the GO bioactivity in osteogenesis mechanisms as being self-sufficient to elevate osteoblast differentiation and bone formation in ectopic sites while lacking the most common osteoinductive agents.  相似文献

Drying Related Strain Development in Restrained Wood   总被引:2,自引:0,他引:2  
The objective of this study was to investigate elastic, viscoelastic, mechanosorptive, and plastic strains developed by the drying process in perpendicular-to-grain restrained western hemlock specimens. Drying tests were performed on small, clear wood specimens sawn in such a manner that either the tangential or the radial direction was parallel to their length, where stress driven deformation rates were determined using resistive transducers. Different mechanical restraint schemes were designed by using weights for partly and a load cell for totally restrained specimens. The difference from previous research done in this domain was the restraining technique, which allowed stress to be induced by the drying process, thus, wood was forced to lift the weight as it reached the fiber saturation point. Drying rates were determined gravimetrically on free specimens having similar wood structure. Experiments with free and restrained specimens were carried out simultaneously while drying at 40, 60, and 80°C, down to 17, 12, and 6% target moisture contents. Overall, the magnitude of restrained shrinkage was successfully quantified and separated in free, combined viscoelastic and mechanosorptive and plastic strains. In all these preliminary experiments the dimensional change started at high overall moisture contents, temperature and environmental conditions being key factors. The results can be used to assess the stress indirectly by studying the restrained strain together with moisture content value.  相似文献

Despite intensive research, the pathophysiology of Alzheimer’s disease (AD) is still not fully understood, and currently there are no effective treatments. Therefore, there is an unmet need for reliable biomarkers and animal models of AD to develop innovative therapeutic strategies addressing early pathologic events such as neuroinflammation and redox disturbances. The study aims to identify inflammatory and redox dysregulations in the context of AD-specific neuronal cell death and DNA damage, using the APPV717I× TAUP301L (AT) mouse model of AD. The expression of 84 inflammatory and 84 redox genes in the hippocampus and peripheral blood of double transgenic AT mice was evaluated against age-matched controls. A distinctive gene expression profile in the hippocampus and the blood of AT mice was identified, addressing DNA damage, apoptosis and thrombosis, complemented by inflammatory factors and receptors, along with ROS producers and antioxidants. Gene expression dysregulations that are common to AT mice and AD patients guided the final selection of candidate biomarkers. The identified inflammation and redox genes, common to AD patients and AT mice, might be valuable candidate biomarkers for preclinical drug development that could be readily translated to clinical trials.  相似文献

Cardiovascular diseases (CVD) are one of the leading causes of high mortality in patients with systemic lupus erythematosus (SLE). The Framingham risk score and other traditional risk factors do not fully reflect the CVD risk in SLE patients. Therefore, in order to stratify these high-risk patients, additional biomarkers for subclinical CVD are needed. The mechanisms of atherogenesis in SLE are still being investigated. During the past decades, many reports recognized that inflammation plays a crucial role in the development of atherosclerosis. The aim of this report is to present novel proinflammatory and pro-atherosclerotic risk factors that are closely related to SLE inflammation and which determine an increased risk for the occurrence of early cardiovascular events.  相似文献

A magnetic resonance (MR) compatible positron emission tomography (PET) insert has been developed in our laboratory for simultaneous small animal PET/MR imaging. This system is based on lutetium oxyorthosilicate (LSO) scintillator arrays with position-sensitive avalanche photodiode (PSAPD) photodetectors. The PET performance of this insert has been measured. The average reconstructed image spatial resolution was 1.51 mm. The sensitivity at the center of the field of view (CFOV) was 0.35%, which is comparable to the simulation predictions of 0.40%. The average photopeak energy resolution was 25%. The scatter fraction inside the MRI scanner with a line source was 12% (with a mouse-sized phantom and standard 35 mm Bruker 1H RF coil), 7% (with RF coil only) and 5% (without phantom or RF coil) for an energy window of 350-650 keV. The front-end electronics had a dead time of 390 ns, and a trigger extension dead time of 7.32 μs that degraded counting rate performance for injected doses above ~0.75 mCi (28 MBq). The peak noise-equivalent count rate (NECR) of 1.27 kcps was achieved at 290 μCi (10.7 MBq). The system showed good imaging performance inside a 7-T animal MRI system; however improvements in data acquisition electronics and reduction of the coincidence timing window are needed to realize improved NECR performance.  相似文献

One of the aims of this work is the study of the geometry of a micro‐cogenerator using a Stirling engine with four double effect pistons. The complex geometry of the heat exchangers was determined by optical measurements. Results of three thermodynamic models: Direct Method from Finite Speed Thermodynamics (FST), isothermal model (Schmidt), and adiabatic model (Finkelstein) are confronted to experimental ones. Direct Method consists of the study and the evaluation of the irreversibilities of thermal machines by analyzing the cycle, step by step, and directly integrating the equation of the First Law for processes with finite speed combined with Second Law of Thermodynamics, for each process of the cycle. The expression of efficiency and power, depending on the speed of the processes and geometric and functional parameters, is then obtained in a straightforward manner. The isothermal and adiabatic models are based on the division of Stirling engine in 3, respectively 5 control volumes, for which the ideal gas law and the equations of mass and energy balance are applied. Analysis of the process of heat transfer and flow of the working gas, taking place in the Stirling engine, is carried out taking into account instantaneous representation of the working fluid volume in the engine. A system of differential equations is solved by iteration using Matlab/Simulink software. The theoretical results are compared to experimental ones. This comparison allows to point out a good accuracy of the Direct Method and the Adiabatic Model, for the thermal operating parameters of the system, noting the different assumptions of each analysis. In this work, different types of honey were characterized based on their palynological and biochemical characteristics. A mellisopalynological analysis was performed to authenticate the botanical origin of the honey samples. According to this method, the honey types were classified in: acacia (n = 10), linden (n = 10), and rape (n = 10). This article also reports the phenols content, the flavonoids content, and the antioxidant activity of honey samples. The highest level of antioxidant activity was recorded for linden honeys and the lowest for acacia honeys. The multivariate analysis demonstrated to be an important tool in classification and discrimination between different honeys concerning the palynological and biochemical properties.  相似文献

The paper presents a characterisation of amorphous silicon carbide films deposited in plasma-enhanced chemical vapour deposition (PECVD) reactors for MEMS applications. The main parameter was optimised in order to achieve a low stress and high deposition rate. We noticed that the high frequency mode (13.56 MHz) gives a low stress value which can be tuned from tensile to compressive by selecting the correct power. The low frequency mode (380 kHz) generates high compressive stress (around 500 MPa) due to ion bombardment and, as a result, densification of the layer achieved. Temperature can decrease the compressive value of the stress (due to annealing effect). A low etching rate of the amorphous silicon carbide layer was noticed for wet etching in KOH 30% at 80 °C (around 13 A/min) while in HF 49% the layer is practically inert. A very slow etching rate of amorphous silicon carbide layer in XeF2 -7 A/min- was observed. The paper presents an example of this application: PECVD-amorphous silicon carbide cantilevers fabricated using surface micromachining by dry-released technique in XeF2.  相似文献

The use of cooling systems based on heat pipe principle to control operation temperature of electronic components is very efficient. They have an excellent miniaturizing capacity and this fact creates adaptability for more practical situations. Starting from the observation that these cooling systems are not precisely characterized from the thermal efficiency point of view, the present paper proposes a methodology of data acquisition for their thermal characterization. An experimental set-up and a data processing algorithm are shown to describe the cooling of a heat generating electronic device using heat pipes. A Thermalright SI-97 PC cooling system is employed as a case-study to determine the heat transfer characteristics of a fins cooler.  相似文献
