Filter media of E12 filters were exposed to NaCl particles and high relative humidity. Afterwards the particle separation efficiency of DEHS (DEHS: di(2‐ethylhexyl) sebacate) and pressure drop were measured according to DIN EN 1822 part 3. The results of new filter media and those at different loadings were compared to determine the influence of relative humidity or water‐soluble impurities like salt. In addition, scanning electron microscope pictures were taken to achieve a better understanding of the particle behavior inside the filters. The results are compared to filter media aged in real applications.  相似文献

Directed evolution of the C25 farnesylgeranyl diphosphate synthase of Aeropyrum pernix (Fgs) was carried out by error-prone PCR with an in vivo color complementation screen utilizing carotenoid biosynthetic pathway enzymes. Screening yielded 12 evolved clones with C20 geranylgeranyl diphosphate synthase activity which were isolated and characterized in order to understand better the chain elongation mechanism of this enzyme. Analysis of these mutants revealed three different mechanisms of product chain length specificity. Two mutants (A64T and A64V) have a single mutation at the 8th amino acid upstream of a conserved first aspartate-rich motif (FARM), which is involved in the mechanism for chain elongation reaction of all prenyl diphosphate synthases. One mutant (A135T) carries a single mutation at the 7th amino acid upstream of another conserved region (141GQ142), which was recently found to be another important region controlling chain elongation of a type III C20 geranylgeranyl diphosphate synthase and Escherichia coli C15 farnesyl diphosphate synthase. Finally, one mutant carrying four mutations (V84I, H88R, I177 M and M191V) is of interest. Molecular modeling, site-directed mutagenesis and in vitro assays of this mutant suggest that product chain-length distribution can be also controlled by a structural change provoked by a cooperative interaction of amino acids.  相似文献

By depositing scent marks on flowers, bees reduce both the search time and the time spent with the handling of nonrewarding flowers. They thereby improve the efficiency of foraging. Whereas in honey bees the source of these scent marks is unknown, it is assumed to be the tarsal glands in bumble bees. According to histological studies, however, the tarsal glands lack any openings to the outside. Foragers of the stingless bee Melipona seminigra have previously been shown to deposit an attractant pheromone at sugar solution feeders, which is secreted at the tips of their tarsi. Here we show that the claw retractor tendons have specialized glandular epithelia within the femur and tibia of all legs that produce this pheromone. The secretion accumulates within the hollow tendon, which also serves as the duct to the outside, and is released from an opening at the base of the unguitractor plate. In choice experiments, M. seminigra was attracted by feeders baited with pentane extracts of the claw retractor tendons in the same way as it was attracted by feeders previously scent marked by foragers. Our results resolve the seeming contradiction between the importance of foot print secretions and the lack of openings of the tarsal glands.  相似文献

Oxidoreductases are attractive biocatalysts that convert achiral substrates into products of higher value, but they are also for the most part dependent on nicotinamide cofactors. Recently, biomimetic nicotinamide derivatives have received attention as less costly alternatives to natural cofactors. However, recycling of biomimetics is still challenging because there are only limited opportunities. Here, we have characterized various biomimetic cofactors with regard to stability and redox potentials to find the best alternative to natural cofactors. Further, the cofactor spectrum of NADH oxidase from Lactobacillus pentosus (LpNox) could be expanded, and the enzymatic activity was also compared to activities with different small‐molecule catalysts. As a result, we succeeded in identifying several strategies for regeneration of oxidized biomimetics.  相似文献

The oxidative dehydrogenation of a 1‐butene/trans‐butene (1:1) mixture to 1,3‐butadiene was carried out in a two‐zone fluidized bed reactor using a Mo‐V‐MgO and a γ‐Bi2MoO6 catalyst. The significant operating conditions temperature, oxygen/butene molar ratio, butene inlet height, and flow velocity were varied to gain high 1,3‐butadiene selectivity and yield. Furthermore, axial concentration profiles were measured inside the fluidized bed to gain insight into the reaction network in the two zones. For optimized conditions and with a suitable catalyst, the two‐zone fluidized bed reactor makes catalyst regeneration and catalytic reaction possible in a single vessel. In the lower part of the fluidized bed, the oxidation of coke deposits on the catalyst as well as the filling of oxygen vacancies in the lattice can occur. The oxidative dehydrogenation reaction takes place in the upper zone. Glycosphingolipids are involved in a number of physiological and pathophysiological processes, and they serve as receptors for a variety of bacterial toxins and viruses. To investigate their function in lipid membranes, fluorescently labeled glycosphingolipids are highly desirable. Herein, a synthetic route to access Gb3 glycosphingolipids with fluorescently labeled fatty acids, consisting of pentaene and hexaene moieties either at the terminus or in the middle of the acyl chain, has been developed. The fluorescent properties of the Gb3 derivatives were investigated in small unilamellar vesicles composed of a raft-like mixture. Phase-separated giant unilamellar vesicles (GUVs) allowed the quantification of the apparent partitioning coefficients of the Gb3 compounds by means of confocal fluorescence laser scanning microscopy. The determined partition coefficients demonstrate that the Gb3 derivatives are preferentially localized in the liquid-disordered (ld) phase. To analyze whether the compounds behave like their physiological counterparts, Cy3-labeled (Cy: cyanine) Shiga toxin B subunits (STxB) were specifically bound to Gb3-doped GUVs. However, the protein was favorably localized in the ld phase, in contrast to results reported for STxB bound to naturally occurring Gb3, which is discussed in terms of the packing density of the lipids in the liquid-ordered (lo) phase.  相似文献

ABSTRACT: A simple method for the fabrication of porous silicon (Si) by metal-assisted etching was developed using gold nanoparticles as catalytic sites. Etching masks were prepared by spin-coating of colloidal gold nanoparticles onto Si. Appropriate functionalization of the gold nanoparticle surface prior to the deposition step enabled the formation of quasi-hexagonally ordered arrays by self-assembly which were translated into an array of pores by subsequent etching in a HF solution containing H2O2. The quality of the pattern transfer depended on the chosen preparation conditions for the gold nanoparticle etching mask. The influence of the Si surface properties were investigated by using either hydrophilic or hydrophobic Si substrates resulting from piranha solution or HF treatment, respectively. The polymer coated gold nanoparticles had to be thermally treated in order to provide direct contact at the metal/Si interface which is required for the following metal-assisted etching. Plasma-treatment as well as flame annealing were successfully applied. Best results were obtained for Si substrates which were treated with HF prior to spin-coating and flame annealed in order to remove the polymer matrix. The presented method opens up new resources for the fabrication of porous silicon by metal-assisted etching. Here, the vast variety of metal nanoparticles accessible by well-established wet-chemical synthesis can be employed for the fabrication of the etching masks.  相似文献

Successful drug design requires not only the detailed knowledge of the pharmacokinetic and pharmacodynamic profiles of the drug candidate portfolio but also a thorough documentation of the possible toxic effects on humans and the environment. Thus, experimental and computational strategies able to measure or predict specific profiles of designed compounds related to their potential toxicity are highly desired. Moreover, a strategy to avoid toxic effects thus enhancing the potential efficacy of drug candidates is of great interest. To fulfil this aim, the pharmacochemistry research unit at the EPGL has recently developed and improved methodologies that detect the potential human health and environmental hazards of compounds active against neurodegeneration at an early stage. A three-step strategy is presented herein. In particular, i) an alternative index to model the bioconcentration of chemicals in the environment was determined; ii) the antioxidant activity of chemical species against free radicals was evaluated. Moreover, since antioxidants play a key role in both toxicity prevention and neuroprotection, iii) the potential interaction of such compounds with enzymatic targets involved in the neurodegenerative cascade was investigated in silico.  相似文献

Three novel magnetic adsorbents were synthesized through the immobilization of di-, tri-, and tetraamine onto the surface of silica coated magnetite nanoparticles. The adsorbents were characterized by XRD patterns, FTIR spectroscopy, elemental and thermogravimetric analysis, magnetic measurements, SEM/TEM, EDX spectroscopy, and N2 adsorption/desorption isotherms. Their capacity to remove copper ions from aqueous solutions was investigated and discussed comparatively. The equilibrium data were analyzed using Langmuir and Freundlich isotherms. The kinetics was evaluated using the pseudo-first-order, pseudo-second-order, and intra-particle diffusion models. The best interpretation for the equilibrium data was given by the Langmuir isotherm for the tri- and tetraamine functionalized adsorbents, while for the diamine functionalized adsorbent the Freundlich model seemed to be better. The kinetic data were well fitted to the pseudo-second-order model. The overall rate of adsorption was significantly influenced by external mass transfer and intraparticle diffusion. It was observed that the adsorption capacity at room temperature decreased as the length of polyamine chain immobilized on the adsorbent surface increased, the maximum adsorption capacities being 52.3 mg g?1 for 1,3-diaminopropan functionalized adsorbent, 44.2 mg g?1 for diethylenetriamine functionalized adsorbent, and 39.2 mg g?1 for triethylenetetramine functionalized adsorbent. The sorption process proved to be highly dependent of pH. The results of the present work recommend these materials as potential candidates for copper removal from aqueous solutions.  相似文献
