A nonlinear dynamics framework that has been applied successfully to several laboratory idealizations of rhythmic behaviors was applied to a more naturally occurring behavior, clapping. Inertial loading of limbs and frequency of oscillation were manipulated. Displacement of relative phase from perfectly in phase and the variability of relative phase, both of which are used as indexes of coordination dynamics, increased with greater inertial imbalance between limbs. Increasing frequency exaggerated these effects. These hallmark properties of coupled oscillator dynamics appeared whether or not the hands contacted, albeit with the latter condition revealing a significant asymmetry in the dynamics. Results highlight the generality of the coupled oscillator regime in interlimb coordination as well as its appropriateness for characterizing behaviors that involve contact of limb surfaces and suggest one way in which perceptual information may tune the dynamical regime. Directed evolution of the C25 farnesylgeranyl diphosphate synthase of Aeropyrum pernix (Fgs) was carried out by error-prone PCR with an in vivo color complementation screen utilizing carotenoid biosynthetic pathway enzymes. Screening yielded 12 evolved clones with C20 geranylgeranyl diphosphate synthase activity which were isolated and characterized in order to understand better the chain elongation mechanism of this enzyme. Analysis of these mutants revealed three different mechanisms of product chain length specificity. Two mutants (A64T and A64V) have a single mutation at the 8th amino acid upstream of a conserved first aspartate-rich motif (FARM), which is involved in the mechanism for chain elongation reaction of all prenyl diphosphate synthases. One mutant (A135T) carries a single mutation at the 7th amino acid upstream of another conserved region (141GQ142), which was recently found to be another important region controlling chain elongation of a type III C20 geranylgeranyl diphosphate synthase and Escherichia coli C15 farnesyl diphosphate synthase. Finally, one mutant carrying four mutations (V84I, H88R, I177 M and M191V) is of interest. Molecular modeling, site-directed mutagenesis and in vitro assays of this mutant suggest that product chain-length distribution can be also controlled by a structural change provoked by a cooperative interaction of amino acids. 相似文献

Natural organic matter (NOM) isolated from the eutrophic Sanctuary Pond (Point Pelee National Park, Canada) has an adverse impact on amphipod species (Gammarus tigrinus and Chaetogammarus ischnus from Lake Müggelsee, Germany, and Eulimnogammarus cyaneus, from Lake Baikal, Russia). Increases in amphipod mortality, changes in peroxidase activity and increases of heat shock protein (hsp70) expression were observed upon exposure to NOM. The highest resistance to the adverse impact of NOM was observed with the endemic Baikalian amphipod E. cyaneus. However, the mechanisms behind this finding remains obscure. If differences in the sensitivity of the hsp70 antibody may be excluded, different modes of action may be postulated: because the adverse impact of NOM may be caused by reactive oxygen species (ROS) and the NOM itself, the observed differences may be due to the action of ROS alone (with E. cyaneus) and a combination of both adverse modes of action (European species). 相似文献

Articular cartilage defects do not heal. Biodegradable scaffolds have been studied for cartilage engineering in order to implant autologous chondrocytes and help cartilage repair. We tested some new collagen matrices differing in collagen type, origin, structure and methods of extraction and purification, and compared the behavior of human chondrocytes cultured on them. Human chondrocytes were grown for three weeks on four different equine type I collagen matrices, one type I, III porcine collagen matrix and one porcine type II collagen matrix. After 21 days, samples were subjected to histochemical, immunohistochemical and histomorphometric analysis to study phenotype expression and cell adhesion. At 7, 14 and 21 days cell proliferation was studied by incorporation of [3H]-thymidine. Our data evidence that the collagen type influences cell morphology, adhesion and growth; indeed, cellularity and rate of proliferation were significantly higher and cells were rounder on the collagen II matrix than on either of the collagen I matrices. Among the collagen I matrices, we observed a great variability in terms of cell adhesion and proliferation. The present study allowed us to identify one type I collagen matrix and one type II collagen matrix that could be usefully employed as a scaffold for chondrocyte transplantation. 相似文献

This paper describes the POESIA approach to systematic composition of Web services. This pragmatic approach is strongly centered in the use of domain-specific multidimensional ontologies. Inspired by applications needs and founded on ontologies, workflows, and activity models, POESIA provides well-defined operations (aggregation, specialization, and instantiation) to support the composition of Web services. POESIA complements current proposals for Web services definition and composition by providing a higher degree of abstraction with verifiable consistency properties. Event structures, Game Semantics strategies and Linear Logic proof-nets arise in different domains (concurrency, semantics, proof-theory) but can all be described by means of directed acyclic graphs (dag's). Fluoroelastomer can be used as a sealing material for different purposes. The aim of this work is the evaluation of the effects of the ionizing radiation of an electron beam (EB) on the mechanical, thermal and surface properties of a commercial fluoroelastomer containing carbon black and inorganic fillers. The material was irradiated with overall doses between 10 and 250 kGy. Tensile strength (stress and strain at break), hardness (Shore A) and compression set were evaluated. Thermal behavior was evaluated by thermogravimetric analysis and differential scanning calorimetry. Surface modifications were inspected using scanning electron microscopy (SEM) and optical microscopy. The experiments have shown that EB irradiation promotes beneficial changes in the fluoroelastomer tensile strength behavior while compression set remain constant and the glass transition temperature increases. The SEM micrographs have shown compactness in the irradiated samples, although optical observations showed no surface morphology changes. 相似文献

For testing the integration of the remote sensing related technologies into the water quality monitoring programs of Lake Garda (the largest Italian lake), the spatial and spectral resolutions of Hyperion and the capability of physics-based approaches were considered highly suitable. Hyperion data were acquired on 22nd July 2003 and water quality was assessed (i) defining a bio-optical model, (ii) converting the Hyperion at-sensor radiances into subsurface irradiance reflectances, and (iii) adopting a bio-optical model inversion technique. The bio-optical model was parameterised using specific inherent optical properties of the lake and light field variables derived from a radiative transfer numerical model. A MODTRAN-based atmospheric correction code, complemented with an air/water interface correction was used to convert Hyperion at-sensor radiances into subsurface irradiance reflectance values. These reflectance values were comparable to in situ reflectance spectra measured during the Hyperion overpass, except at longer wavelengths (beyond 700 nm), where reflectance values were contaminated by severe atmospheric adjacency effects. Chlorophyll-a and tripton concentrations were retrieved by inverting two Hyperion bands selected using a sensitivity analysis applied to the bio-optical model. The sensitivity analysis indicated that the assessment of coloured dissolved organic matter was not achievable in this study due to the limited coloured dissolved organic matter concentration range of the lake, resulting in reflectance differences below the environmental measurement noise of Hyperion. The chlorophyll-a and tripton image-products were compared to in situ data collected during the Hyperion overpass, both by traditional sampling techniques (8 points) and by continuous flow-through systems (32 km). For chlorophyll-a the correlation coefficient between in situ point stations and Hyperion-inferred concentrations was 0.77 (data range from 1.30 to 2.16 mg m− 3). The Hyperion-derived chlorophyll-a concentrations also match most of the flow-through transect data. For tripton, the validation was constrained by variable re-suspension phenomena. The correlation coefficient between in situ point stations and Hyperion-derived concentrations increased from 0.48 to 0.75 (data range from 0.95 to 2.13 g m− 3) if the sampling data from the re-suspension zone was avoided. The comparison of Hyperion-derived tripton concentrations and flow-through transect data exhibited a similar mismatch. The results of this research suggest further studies to address compatibilities of validation methods for water body features with a high rate of change, and to reduce the contamination by atmospheric adjacency effects on Hyperion data at longer wavelengths in Alpine environment. The transferability of the presented method to other sensors and the ability to assess water quality independent from in situ water quality data, suggest that management relevant applications for Lake Garda (and other subalpine lakes) could be supported by remote sensing. 相似文献
