The Staphylococci populations in different types of Iberian dry fermented sausages from central-west Spain were identified. A simple electrophoretic method of whole-cell proteins and extracellular protein profiling was evaluated for speed of identification. This study was correlated with a 16S rRNA gene sequence analysis and biochemical identification by API Staph. A total of 81 isolates were identified by SDS-PAGE of the whole-cell proteins. These showed stable profiles in the range 99-14kDa that were clearly different for the different species, and were grouped into clusters together with the profiles of the eight reference strains. SDS-PAGE of the extracellular protein extracts provided additional characteristic banding patterns for the characterization of the Staphylococcus species present. The whole-cell SDS-PAGE showed that the predominant species was Staphylococcus saprophyticus (61.7%) followed by Staphylococcus aureus (19.7%). The identifications were confirmed by the 16S rRNA gene sequencing and by a BLAST search of the GenBank database. However, the API Staph biochemical identifications were frequently erroneous at the species level. In sum, SDS-PAGE analysis showed itself to be rapid and accurate in identifying the most commonly encountered Staphylococcus isolates in dry fermented sausages.  相似文献

Red wine has been reported to exert beneficial effects in preventing cardiovascular diseases probably due to their polyphenols constituents. The vasorelaxant capacity and phenolic composition of four monovarietal young red wines Merlot, Tempranillo, Garnacha and Cabernet-Sauvignon obtained from grapes from 2004 vintage, cultivated under the same climatic and agricultural conditions, wine-making technology and storage conditions, were determined using vascular reactivity assay in rat aortic and high-performance liquid chromatography, respectively. Primary cultures of vascular smooth muscle cells from Wistar rats were also used to evaluate their survival prevention activity as well. Every studied wine present vasorelaxation effect, but the higher value corresponds to the analyzed Merlot wine, especially rich in phenolic compounds, mainly catechins and oligomeric proanthocyanidins and anthocyanin glycosides. This study further demonstrated previous investigations about the phenolic composition of wines relation with their vasorelaxation activity.  相似文献

Manufacturers are deeply involved in the development of new pomegranate‐based products, which have acquired great prestige due to many studies proving their potential health benefits. Commercial pomegranate products including capsules and supplements and juices and nectars were assayed. The contents of the key phytochemical compounds (punicalagin, ellagic acid, and total polyphenolic content) and the associated antioxidant capacity (DPPH?, ABTS?+, and FRAP) were analyzed. The experimental ranges of punicalagin and ellagic acid contents were 0.96 to 308 mg/g and 0.09 to 13.1 mg/g, respectively. Punicalagin content was positively correlated (< 0.001) with DPPH? and total polyphenolic content values. No significant (P > 0.05) correlation was observed among ellagic acid with the antioxidant capacity. The labeling standardization of these products is necessary due to the wide variability among “theoretically” similar pomegranate‐based products.  相似文献

A method to derive reflectance spectra and colour coordinates of meat by measurement of reflectance at only three wavelengths, 480, 570 and 620 nm has been developed, by treating the spectra of a meat sample as a linear combination of the spectra of the three major myoglobin derivatives (reduced myoglobin, oxymyoglobin and metmyoglobin). Applied to 300 samples of Semitendinosus and Longissimus dorsi veal muscles at different times post mortem (24 h to 9 d), colour differences ΔE (?) between the derived spectra and the real ones were about 1.5 units. The total pigment content, with high correlation with the extraction method (P < 0.02), and the relative proportions of the derivatives can also be estimated.  相似文献

A polystyrene (PS)/poly(butyl acrylate) (PBA) composite emulsion was produced by seeded emulsion polymerization of butyl acrylate (BA) with PS seed particles which were prepared by emulsifier‐free polymerization of styrene with potassium persulfate (KPS) under a nitrogen atmosphere at 70°C for 24 h with stirring at 60 rpm and swelled with the BA monomer in an ethanol/water medium. The structure of the PS/PBA composite particles was confirmed by the presence of the characteristic absorption band attributed to PS and PBA from FTIR spectra. The particles for pure PS and PS/PBA with a low content of the BA monomer were almost spherical and regular. As the BA monomer content was increased, the particle size of the PS/PBA composite particles became larger, and more golf ball‐like particles were produced. The surface morphology of the PS/PBA composite particles was investigated by AFM and SEM. The Tg's attributed to PS and PBA in the PS/PBA composite particles were found at 110 and ?49°C, respectively. The thermal degradation of the pure PS and PS/PBA composite particles occurred in one and two steps, respectively. With an increasing amount of PBA, the initial thermal decomposition temperature increased. Three new regulated deficit irrigation (RDI) treatments were applied to “Arbequina” olive orchards during pit hardening. Oil quality was determined by measuring analytical parameters for olive oil grading, antioxidant activity, total phenol content, fatty acid profile, volatile compounds profile, and sensory analysis. Oils from RDI were classified as “extra virgin olive oil” and their quality was improved due to their higher antioxidant potential (ABTS+ [increased ~75%] and DPPH˙ [increased ~25%] assays) and phenols (increased ~53%) than control. Concentration of total volatile compounds decreased (~27%) but RDI olive oils showed a more balanced profile (alcohols, aldehydes, and esters). Monounsaturated fatty acid content increased (~5%) and atherogenic and thrombogenic indexes decreased (~8.5%) in RDI olive oil. Regarding sensory analysis, RDI provided more balanced oils with higher fruit aroma than control. Other benefits of RDI olive oil, when compared with oil from full irrigated orchards are reduced use of water and improved functional and sensory quality.  相似文献

The esterification of oleic acid and methanol using sulfuric acid as a homogeneous catalyst is studied in reactive‐separation systems. The conversion of the free fatty acid was investigated in two different experiments with the molar ratio of methanol/oleic acid, amount of catalyst, temperature, and reaction time as variables. The conversion of the free fatty acid was found to depend strongly on the molar ratio of methanol/oleic acid. The reaction time had a direct effect on the conversion of the free fatty acid, and this conversion decreased with higher temperature. These results were valuable for a preliminary study on biodiesel production, using an acid homogeneous catalyst in a reactive dividing‐wall distillation column.  相似文献

Stabilized tetragonal Bi2O3 nanoparticles (β-Bi2O3) were obtained by annealing treatments of amorphous Bi-based precursors, obtained by chemical precipitations, at temperatures between 350 and 450?°C. The formation of the stabilized β-Bi2O3 phase was possible by using (BiO)4CO3(OH)2 while other precursors such as amorphous bismuth carbonate ((BiO)2CO3) and amorphous basic bismuth nitrate (Bi6O6(OH)2(NO3)4·2H2O) led to the formation of the thermodynamically stable monoclinic α-Bi2O3 and Bi5O7NO3 phases. The Bi-based precursors were prepared by the chemical precipitation method at room temperature in ethylenediamine-solvent varying the HNO3/Bi3+ molar ratio (10, 26 and 56). The physicochemical properties of the three as-prepared amorphous precursors and the formed-after-calcination β-Bi2O3, α-Bi2O3 and Bi5O7NO3 phases were analyzed by X-ray diffraction, scanning electron microscopy, thermogravimetry, X-ray photoelectron spectroscopy (XPS), FTIR analysis, diffuse reflectance spectroscopy and surface area by BET method. The photocatalytic activity of all annealed solids containing the β-Bi2O3 phase was tested in the photodegradation of the indigo carmine (IC) dye under specific blue light. A schematic diagram of the Bi2O3 phases obtained as a function of the annealing conditions and initial amorphous precursor is proposed and explained in terms of the amount of CO32-, NO3- and amine (ENH22+ ? ENH+) ions present in each bismuth precursor.  相似文献
