Inhibition of tumorigenicity of the teratoma PC cell line by transfection with antisense cDNA for PC cell-derived growth factor (PCDGF, epithelin/granulin precursor)

The PC cell line is a highly tumorigenic, insulin-independent, teratoma-derived cell line isolated from the nontumorigenic, insulin-dependent 1246 cell line. Studies of the PC cell growth properties have led to the purification of an 88-kDa secreted glycoprotein called PC cell-derived growth factor (PCDGF), which has been shown to stimulate the growth of PC cells as well as 3T3 fibroblasts. Sequencing of PCDGF cDNA demonstrated its identity to the precursor of a family of 6-kDa double-cysteine-rich polypeptides called epithelins or granulins (epithelin/granulin precursor). Since PCDGF was isolated from highly tumorigenic cells, its level of expression was examined in PC cells as well as in nontumorigenic and moderately tumorigenic cells from which PC cells were derived. Northern blot and Western blot analyses indicate that the levels of PCDGF mRNA and protein were very low in the nontumorigenic cells and increased in tumorigenic cell lines in a positive correlation with their tumorigenic properties. Experiments were performed to determine whether the autocrine production of PCDGF was involved in the tumorigenicity of PC cells. For this purpose, we examined the in vivo growth properties in syngeneic C3H mice of PC cells where PCDGF expression had been inhibited by transfection of antisense PCDGF cDNA. The results show that inhibition of PCDGF expression resulted in a dramatic inhibition of tumorigenicity of the transfected cells when compared with empty-vector control cells. These data demonstrate the importance in tumor formation of overexpression of the novel growth factor PCDGF.

Carbon fibre composites based on polyimide/silica ceramers: aspects of structure-properties relationship

Polyimide/silica ceramers, based on the products of the hydrolysis of tetraethoxysilane (TEOS) and a commercial poly(amic acid) solution, were used to fabricate unidirectional carbon fibre composites, which were subsequently evaluated with respect to thermal and mechanical properties. There is evidence to suggest that the silica component of these ceramers is present as dispersed discrete particles at low silica concentration (i.e. 7 wt%) and as fine interconnected domains trapped within the polyimide matrix at higher silica content (i.e. 14 wt%). The dimensions of the silica domains were in the region of 7–20 nm. Carbon fibre composites produced from ceramer solutions (CF/ceramers) were found to exhibit lower thermal expansion and a greater retention of flexural and interlaminar shear properties at elevated temperature than the corresponding polyimide-matrix composites (CF/polyimide). The properties of CF/ceramers were generally better for systems containing the higher amount of silica and were improved further by lowering the pH value of the precursor ceramer solution. This is believed to have resulted from the enhanced fluidity of the ceramer gel within the pre-impregnated fibres, giving rise to a higher packing density of the fibres and a more homogeneous distribution of fibres. CF/ceramers were also found to exhibit a better thermal oxidative stability at 350°C than the corresponding CF/polyimide, although a substantial amount of porosity developed in the case of ceramers with the higher silica content.     

A methodology for modeling the distributions of medical images and their stochastic properties

The probabilistic distribution properties of a set of medical images are studied. It is shown that the generalized Gaussian function provides a good approximation to the distribution of AP chest radiographs. Based on this result and a goodness-of-fit test, a generalized Gaussian autoregressive model (GGAR) is proposed. Its properties and limitations are also discussed. It is expected that the GGAR model will be useful in describing the stochastic characteristics of some classes of medical images and in image data compression and other applications.     

Influence of content and creator characteristics on sharing disaster-related information on social media

Identifying the key factors of the disaster-related information propagation process can provide decision support for disaster management. This study characterizes the effects of content types, location, and social capital of social media users on the virality of disaster-related information. We found through the Weibo dataset of the Yiliang earthquake that the virality of different types of information can vary on the basis of the social capital of users who post the information. This study fills the current research gaps by examining the individual and joint effects of the content and creator characteristics on the virality of disaster-related information.
