Hybrid Approach for Addressing Uncertainty in Risk Assessments   总被引:3,自引:0,他引:3  
Parameter uncertainty is a major aspect of the model-based estimation of the risk of human exposure to pollutants. The Monte Carlo method, which applies probability theory to address model parameter uncertainty, relies on a statistical representation of available information. In recent years, other uncertainty theories have been proposed as alternative approaches to address model parameter uncertainty in situations where available information is insufficient to identify statistically representative probability distributions, due in particular to data scarcity. The simplest such theory is possibility theory, which uses so-called fuzzy numbers to represent model parameter uncertainty. In practice, it may occur that certain model parameters can be reasonably represented by probability distributions, because there are sufficient data available to substantiate such distributions by statistical analysis, while others are better represented by fuzzy numbers (due to data scarcity). The question then arises as to how these two modes of representation of model parameter uncertainty can be combined for the purpose of estimating the risk of exposure. This paper proposes an approach (termed a hybrid approach) which combines Monte Carlo random sampling of probability distribution functions with fuzzy calculus. The approach is applied to a real case of estimation of human exposure, via vegetable consumption, to cadmium present in the surficial soils of an industrial site located in the north of France. The application illustrates the potential of the proposed approach, which allows the uncertainty affecting model parameters to be represented in a way that is consistent with the information at hand. Also, because the hybrid approach takes advantage of the “rich” information provided by probability distributions, while retaining the conservative character of fuzzy calculus, it is believed to hold value in terms of a “reasonable” application of the precautionary principle.  相似文献

We have developed a tunable diode laser spectrometer, called SIMCO (spectrometer for isotopic measurements of CO(2)), for determining the concentrations of (12)CO(2) and (13)CO(2) in atmospheric air, from which the total concentration of CO(2) and the isotopic composition (expressed in delta units) delta(13)CO(2) are calculated. The two concentrations are measured using a pair of lines around 2290.1 cm(-1), by fitting a line profile model, taking into account the confinement narrowing effect to achieve a better accuracy. Using the Allan variance, we have demonstrated (for an integration time of 25 s) a precision of 0.1 ppmv for the total CO(2) concentration and of 0.3[per thousand] for delta(13)CO(2). The performances on atmospheric air have been tested during a 3 days campaign by comparing the SIMCO instrument with a gas chromatograph (GC) for the measurement of the total CO(2) concentration and with an isotopic ratio mass spectrometer (MS) for the isotopic composition. The CO(2) concentration measurements of SIMCO are in very good agreement with the GC data with a mean difference of Delta(CO(2))=0.16+/-1.20 ppmv for a comparison period of 45 h and the linearity of the concentration between the two instruments is also very good (slope of correlation: 0.9996+/-0.0003) over the range between 380 and 415 ppmv. For delta(13)CO(2), the comparison with the MS data shows a larger mean difference of Delta(delta(13)CO(2))=(-1.9+/-1.2)[per thousand], which could be partly related to small residual fluctuations of the overall SIMCO instrument response.  相似文献

The experiments reported in this paper demonstrate the causes of the failure of monolithic ceramics. The specimens are made of silicon nitride and tested at room temperature. The stress field within the specimen is different for each of four series of tests that have been conducted. Fractographic observations have also been made to identify the causes of the failures. This paper describes evidence of (n−3) and particularly of 22∶6 (n−3) fatty acid enrichment in trout lipoproteins as well as in vitellogenin, egg lipovitellin and oil globule. Among the lipoproteins, HDL and LDL were the main forms of blood lipid transport, whereas phospholipids and cholesteryl esters are the preferential chemical carriers for (n−3) fatty acid transport. However, cholesteryl esters were less important as esterified fatty acid carriers than in man. Taken together with the data obtained in mammals, our results suggest that there may be a relationship between EFA activity and the distribution of the EFA among the lipoprotein lipid fractions in vertebrates, irrespective of the EFA series. Administration of an (n−3) fatty acid deficient diet for three months prior to trout spawning produced a significant increase in egg lipid content, primarily as a result of the increase of the oil globule composed almost exclusively of triacylglycerols. This diet decreased the 22∶6 (n−3), as well as the (n−3) fatty acid contents of lipoproteins, lipovitellin, vitellogenin and the oil globule. In contrast, the (n−3) fatty acid level was always higher in lipoproteins and lipovitellin than in the vitellogenin and the oil globule. Moreover, the relative levels of 22∶6 (n−3) and total (n−3) fatty acids were quite similar in lipoproteins and lipovitellin on the one hand, and in vitellogenin and the oil globule on the other. These findings suggest a direct relationship between the two forms of plasma lipid transport and the two egg compartments. During ovogenesis, dietary lipids seemed to be diverted from the adipose tissue and essentially deposited in the egg.  相似文献

In the context of the high-level radioactive waste disposal CIGEO, the corrosion rate due to microbially influenced corrosion (MIC) has to be evaluated. In France, it is envisaged to dispose of high- and intermediate-level long-lived radioactive waste at a depth of 500 m in a deep geological disposal, drilled in the Callovo-Oxfordian claystone (Cox) formation. To do so, a carbon steel casing will be inserted inside disposal cells, which are horizontal tunnels drilled in the Cox. A specific cement grout will be injected between the carbon steel casing and the claystone. A study was conducted to evaluate the possibility of MIC on carbon steel in the foreseeable high radioactive waste disposal. The corrosiveness of various environments was investigated at 50°C and 80°C with or without microorganisms enriched from samples of Andra's underground research laboratory. The monitoring of corrosion during the experiments was ensured using gravimetric method and real-time corrosion monitoring using sensors based on the measurements of the electrical resistance. The corrosion data were completed with microbiological analyses including cultural and molecular characterizations.  相似文献

This work describes a collaborative effort to define and apply a protocol for the rational selection of a general‐purpose screening library, to be used by the screening platforms affiliated with the EU‐OPENSCREEN initiative. It is designed as a standard source of compounds for primary screening against novel biological targets, at the request of research partners. Given the general nature of the potential applications of this compound collection, the focus of the selection strategy lies on ensuring chemical stability, absence of reactive compounds, screening‐compliant physicochemical properties, loose compliance to drug‐likeness criteria (as drug design is a major, but not exclusive application), and maximal diversity/coverage of chemical space, aimed at providing hits for a wide spectrum of drugable targets. Finally, practical availability/cost issues cannot be avoided. The main goal of this publication is to inform potential future users of this library about its conception, sources, and characteristics. The outline of the selection procedure, notably of the filtering rules designed by a large committee of European medicinal chemists and chemoinformaticians, may be of general methodological interest for the screening/medicinal chemistry community. The selection task of 200K molecules out of a pre‐filtered set of 1.4M candidates was shared by five independent European research groups, each picking a subset of 40K compounds according to their own in‐house methodology and expertise. An in‐depth analysis of chemical space coverage of the library serves not only to characterize the collection, but also to compare the various chemoinformatics‐driven selection procedures of maximal diversity sets. Compound selections contributed by various participating groups were mapped onto general‐purpose self‐organizing maps (SOMs) built on the basis of marketed drugs and bioactive reference molecules. In this way, the occupancy of chemical space by the EU‐OPENSCREEN library could be directly compared with distributions of known bioactives of various classes. This mapping highlights the relevance of the selection and shows how the consensus reached by merging the five different 40K selections contributes to achieve this relevance. The approach also allows one to readily identify subsets of target‐ or target‐class‐oriented compounds from the EU‐OPENSCREEN library to suit the needs of the diverse range of potential users. The final EU‐OPENSCREEN library, assembled by merging five independent selections of 40K compounds from various expert groups, represents an excellent example of a Europe‐wide collaborative effort toward the common objective of building best‐in‐class European open screening platforms.  相似文献

A 3‐MPa, 350 °C fixed‐bed reactor was designed to follow‐up gas‐liquid‐solid reactions on a millimetric size heterogeneous catalyst with Raman spectroscopy. The transparent reactor is a quartz cylinder enclosed in a Joule effect heated stainless‐steel tube. A methodology to determine how to focus the microscope for liquid and solid phase characterization is presented. The setup was validated by performing diesel hydrodesulfurization on a CoMo/alumina extrudate catalyst with a conversion very close to expected values along with the acquisition of Raman spectra of the solid catalyst showing an evolution of the catalyst phase during sulfidation.  相似文献

The work reported involved the fabrication of an electrospun tubular conduit of a gelatin and polycaprolactone (PCL) blend as an adventitia‐equivalent construct. Gelatin was included as the matrix for increased biocompatibility with the addition of PCL for durability. This is contrary to most of the literature available for biomaterials based on blends of gelatin and PCL where PCL is the major matrix. The work includes the assiduous selection of key electrospinning parameters to obtain smooth bead‐free fibres with a narrow distribution of pore size and fibre diameter. Few reports elucidate the optimization of all electrospinning parameters to fabricate tubular conduits with a focus on obtaining homogeneous pores and fibres. This stepwise investigation would be unique for the fabrication of gelatin–PCL electrospun tubular constructs. The fabricated microfibrous gelatin–PCL constructs had pores of size ca 50–100 μm reportedly conducive for cell infiltration. The measured value of surface roughness of 57.99 ± 17.4 nm is reported to be favourable for protein adhesion and cell adhesion. The elastic modulus was observed to be similar to that of the tunica adventitia of the native artery. Preliminary in vitro and in vivo biocompatibility tests suggest safe applicability as a biomaterial. Minimal cytotoxicity was observed using MTT assay. Subcutaneous implantation of the scaffold demonstrated acute inflammation which decreased by day 15. The findings of this study could enable the fabrication of smooth bead‐free microfibrous gelatin–PCL tubular construct as viable biomaterial which can be included in a bilayer or a trilayer scaffold for vascular tissue engineering. © 2019 Society of Chemical Industry  相似文献

Piezoelectric films were prepared by incorporation of lithium niobate (LiNbO3) nanoparticles into copolymer of vinylidene difluoride and trifluoroethylene. Nanoparticles of LiNbO3 with ferroelectric phase were successfully synthesized and dispersed homogenously by ultrasonication in the copolymer matrix without any surfactant or surface functionalization. The nanocomposites were fully characterized by electronic microscopy, X‐ray diffraction, differential scanning calorimetry, dynamical mechanical analysis, and piezometer. Surprisingly, the copolymer matrix crystallinity and morphology were not affected by the incorporation of nanoparticles. Therefore the nanocomposites remained good mechanicals properties and high ferroelectricity coupled to nonlinear optical activity thanks to the noncentro symmetric space group of lithium niobate.
