The interaction and crosstalk of Toll-like receptors (TLRs) is an established pathway in which the innate immune system recognises and fights pathogens. In a single nucleotide polymorphisms (SNP) analysis of an Indian cohort, we found evidence for both TLR4-399T and TRL8-1A conveying increased susceptibility towards tuberculosis (TB) in an interdependent manner, even though there is no established TLR4 ligand present in Mycobacterium tuberculosis (Mtb), which is the causative pathogen of TB. Docking studies revealed that TLR4 and TLR8 can build a heterodimer, allowing interaction with TLR8 ligands. The conformational change of TLR4-399T might impair this interaction. With immunoprecipitation and mass spectrometry, we precipitated TLR4 with TLR8-targeted antibodies, indicating heterodimerisation. Confocal microscopy confirmed a high co-localisation frequency of TLR4 and TLR8 that further increased upon TLR8 stimulation. The heterodimerisation of TLR4 and TLR8 led to an induction of IL12p40, NF-κB, and IRF3. TLR4-399T in interaction with TLR8 induced an increased NF-κB response as compared to TLR4-399C, which was potentially caused by an alteration of subsequent immunological pathways involving type I IFNs. In summary, we present evidence that the heterodimerisation of TLR4 and TLR8 at the endosome is involved in Mtb recognition via TLR8 ligands, such as microbial RNA, which induces a Th1 response. These findings may lead to novel targets for therapeutic interventions and vaccine development regarding TB.  相似文献

Small extracellular vesicles isolated from urine (uEVs) are increasingly recognized as potential biomarkers. Meanwhile, different uEV preparation strategies exist. Conventionally, the performance of EV preparation methods is evaluated by single particle quantification, Western blot, and electron microscopy. Recently, we introduced imaging flow cytometry (IFCM) as a next-generation single EV analysis technology. Here, we analyzed uEV samples obtained with different preparation procedures using nanoparticle tracking analysis (NTA), semiquantitative Western blot, and IFCM. IFCM analyses demonstrated that urine contains a predominant CD9+ sEV population, which exceeds CD63+ and CD81+ sEV populations. Furthermore, we demonstrated that the storage temperature of urine samples negatively affects the recovery of CD9+ sEVs. Although overall reduced, the highest CD9+ sEV recovery was obtained from urine samples stored at −80 °C and the lowest from those stored at −20 °C. Upon comparing the yield of the different uEV preparations, incongruencies between NTA and IFCM data became apparent. Results obtained by both NTA and IFCM were consistent with Western blot analyses for EV marker proteins; however, NTA results correlated with the amount of the impurity marker uromodulin. Despite demonstrating that the combination of ultrafiltration and size exclusion chromatography appears as a reliable uEV preparation technique, our data challenge the soundness of traditional NTA for the evaluation of different EV preparation methods.  相似文献

The characteristic influences of increasing concentrations of graphene, expanded graphite (EG), carbon black (CB), and multiwall carbon nanotubes (MWNT) are investigated on pyrolysis, reaction to small flame, burning behavior, and on electrical, thermal, and rheological properties of flame retarded polypropylene (PP‐FR). The property‐concentration dependency is different for the various material properties, as threshold, linear, and leveling off functions were observed. Increasing concentrations of carbon nanoparticles resulted in a decrease in the electrical resistivity of the polymer by crossing the percolation threshold. The developing nanoparticle network changes melt flow behavior for small shear rates, increases thermal conductivity and therefore, affects the UL 94 classification and oxygen index. The onset temperature of PP decomposition is shifted to temperatures up to 37°C higher; the peak heat release rate is reduced by up to 74% compared to PP‐FR. Both effects leveled off with increasing particle concentration. Among the four carbon nanomaterials tested, graphene presents superior influence on composite properties over the tested concentration range and outperforms commercial CB, MWNT, and EG. POLYM. Robust and reproducible processes are necessary to achieve a constant and high quality of biotechnological products. Thus, suitable online measurement systems and an increased understanding of the parameters influencing the process are required. In this study, two online measurement devices were used to detect the influence of the initial pH value on the lag phase of three E. coli and one B. licheniformis strains cultivated in buffered mineral media. It was shown that the initial pH value, the type of buffer and its concentration have to be carefully chosen to avoid differences in the lag phase, unsuitable pH values during the cultivation, and osmotically inhibiting high buffer concentrations.  相似文献

A multi-sensor approach was used to determine high-resolution porewater gradients of Ca(2+), CO(3)(2-), H(+) and O(2) in sediment cores along a depth transect from eutrophic Lake Sempach (Switzerland). We quantified the reproducibility of measurements and analyzed concentration profiles with a one-dimensional diffusion-reaction model to quantify benthic fluxes. Calculation of oxic respiration in the sediment showed that almost all settled organic carbon was degraded with O(2) at shallow depths while only 28% was decomposed aerobically at the deepest location. Fluxes were highest at 8 m depth (24.4 mM m(-2) d(-1)) and lowest at the deepest site of 85 m (7.1 mM m(-2) d(-1)). Dissolution of calcite from the sediment was also depth dependent. A total of 1.1 mM m(-2) d(-1) was found for the shallowest site, and values decreased with depth to 0.6 mM m(-2) d(-1) at the deepest site.  相似文献

Abstract

This article analyses how firms use events and trade fairs for external knowledge sourcing, which barriers emerge and how event organisers strategically mediate and influence those processes. The research setting focuses on two major automotive events in Shanghai, highlighting that knowledge sourcing in these events do complement other types of knowledge accessed in permanent ‘sites’ and organisational configurations, such as in clusters and through joint-ventures. Firms use automotive events to access buzz, to monitor other firms and to explore options for new collaborations. Yet, it is also argued that a focus on existing relations, the defensive strategies deployed by lead firms and the intrinsic complexity of exhibited technologies hinder the process of knowledge sourcing that is influenced by event organisers’ content, matchmaking and access policies.  相似文献

The effects of two different low calcium fly ashes on the hydration of ordinary Portland cement (OPC) pastes containing 50 wt.% of fly ash were investigated over a hydration time of 550 days. The results were compared with a reference blend of OPC containing 50 wt.% of inert quartz powder allowing the distinction between "filler effect" and pozzolanic reaction.Until 2 days, no evidence of fly ash reaction was measured and its influence on the hydration is mainly related to the “filler effect”. From 7 days on, the effects of the pozzolanic reaction were observed by the consumption of portlandite, the change of the pore solution chemistry, the formation of a presumably water-rich inner hydration product and the change of the C–S–H composition towards higher Al/Si ratio compared to the C–S–H of neat OPC. Additional strength due to the pozzolanic reaction developed after 28 days of hydration.  相似文献

Commercially available antibacterial semisolid preparations intended for topical application provide only short-term drug release. A sustained kinetics is possible by exploitation of a biodegradable polymer carrier. The purpose of this work is to formulate a mucoadhesive system with aciclovir (ACV) based on a solid molecular dispersion of this drug in poly(lactic-co-glycolic acid) branched on tripenterythritol (PLGA/T). The ACV incorporation into PLGA/T was carried out either by solvent method, or melting method, or plasticization method using various plasticizers. The drug–polymer miscibility, plasticizer efficiency and content of residual solvent were found out employing DSC. Viscosity was measured at the shear rate range from 0.10 to 10.00?s?1 at three temperatures and data were analyzed by Newtonian model. The mucoadhesive properties were ascertained in the tensile test on a mucin substrate. The amount of ACV released was carried out in a wash-off dissolution test. The DSC results indicate a transformation of crystalline form of ACV into an amorphous dissolved in branched polyester carrier, and absence of methyl formate residuals in formulation. All the tested plasticizers are efficient at Tg depression and viscosity decrease. The non-conventional ethyl pyruvate possessing supportive anti-inflammatory activity was evaluated as the most suitable plasticizer. The ACV release was strongly dependent on the ethyl pyruvate concentration and lasted from 1 to 10 days. The formulated PLGA/T system with ACV exhibits increased adhesion to mucosal hydrophilic surfaces and prolonged ACV release controllable by degradation process and viscosity parameters.  相似文献

The challenge for unmanned aerial vehicles to sense and avoid obstacles becomes even harder if narrow passages have to be crossed. An approach to solve a mission scenario that tackles the problem of such narrow passages is presented here. The task is to fly an unmanned helicopter autonomously through a course with gates that are only slightly larger than the vehicle itself. A camera is installed on the vehicle to detect the gates. Using vehicle localization data from a navigation solution, camera alignment and global gate positions are estimated simultaneously. The presented algorithm calculates the desired target waypoints to fly through the gates. Furthermore, the paper presents a mission execution plan that instructs the vehicle to search for a gate, to fly through it after successful detection, and to search for a proceeding one. All algorithms are designed to run onboard the vehicle so that no interaction with the ground control station is necessary, making the vehicle completely autonomous. To develop and optimize algorithms, and to prove the correctness and accuracy of vision-based gate detection under real operational conditions, gate positions are searched in images taken from manual helicopter flights. Afterwards, the integration of visual sensing and mission control is proven. The paper presents results from full autonomous flight where the helicopter searches and flies through a gate without operator actions.  相似文献
